Abstract

Es wird ein neues Verfahren zur Herstellung von N-(2-(((5-(Dimethylamino)-methyl-2-furanyl)methyl)thio)aethyl)-N'-methyl-2-nitro-1,1-aethendiamin (Ranitidin) der Formel I und seinen physiologisch vertraeglichen Saeureadditionssalzen beschrieben, bei welchem 2-(((5-(Dimethylamino)methyl-2-furanyl)methyl)thio)aethanaminder Formel II mit 1-Nitro-3-methyl-ketenimin der Formel CH tief3-N=C=CHNO tief2 zur gewuenschten Verbindung umgesetzt wird. 1-Nitro-3-methyl-ketenimin wird "in situ" durch Umsetzung von 1-Methylthio-1-methylamino-2-nitroaethen der Formel III in einem polaren organischen Loesungsmittel, wie Methanol oder Acetonitril, mit der Loesung eines Schwermetallsalzes, wie AgNO tief3, CuCl, in demselben Loesungsmittel in Anwesenheit von einem Protonenakzeptor, wie Triaethylamin, bei Raumtemperatur gebildet. Ranitidin ist ein wertvolles Arzneimittel in der Behandlung von Ulkus-Krankheiten. Wegen des sehr einfachen Reaktionsverlaufs und der ausserordentlich kurzen Reaktionszeit ist das Verfahren hervorragend fuer den industriellen Massstab geeignet.
