1394845 Process for penicillamine DEUTSCHE GOLD-UND SILBER-SCHEIDEANSTALT 1 Nov 1972 [15 Nov 1971] 50375/72 Addition to 1346198 Heading C2C The invention comprises a process for the preparation of penicillamine or a homologue thereof which comprises reacting an aliphatic aldehyde branched on the α-carbon atom with sulphur and ammonia to form a #<SP>3</SP>-thiazoline, converting the #<SP>3</SP>-thiazoline with anhydrous hydrogen cyanide into a thiazolidine-4-carbonitrile, hydrolysing the thiazolidine-4-carbonitrile into a salt of a thiazolidine-4-carbonamide by introducing it into aqueous hydrochloric acid containing at least 30% by weight of hydrogen chloride and heating to a temperature of from 40 to 70‹ C., converting the salt of a thiazolidine-4- carbonamide at a temperature above 80‹ C. into a salt of a thiazolidine-4-carboxylic acid and an ammonium salt, working up the mixture and separating off the ammonium salt followed by hydrolytic degradation of the thiazolidine-4- carboxylic acid to produce the penicillamine or its homologue.
