Abstract
1393654 Making viral vaccines MERCK & CO INC 11 June 1973 [14 June 1972] 27636/73 Heading A5B [Also in Divisions Bl and C6] In a process for the production of cells and/ or vaccines cells are grown and/or infected with virus in a rotary multiplate propagator which comprises a cylindrical tank 2 and a plurality of plates 19 mounted normally to the axis of the cylindrical tank and rotatable thereabout, the plate and tank diameters being in the ratio 0.8:1 to 0.9:1. Preferably the surface area to volume ratio of the apparatus lies between 5.5:1 and 6.0:1. The tank may be of stainless steel, and the plates of titanium, and typical diameters are 7“ inches and 6 inches or 6 inches and 5.4 inches respectively. Drive to the shaft 20 on which the plates are mounted includes a magnetic coupling 25, 26 and the tank is provided with inlet and outlet connections for supply and egress of air or air/CO 2 mixture, culture medium and product or spent liquor. Coating of the plates with cells is effected with the apparatus placed with its axis vertical and the apparatus may be inverted to ensure coating of both sides of the plates. Cell growth or infection of cells grown on the plates is carried out with the apparatus horizontal, about half full of liquid medium and with the plates rotating e.g. at about 1/5 r.p.m. Reference is made and examples given relating to production of viral vaccines such as mumps, measles, rubella, Marek's, influenza, parainfluenza, varicella, trachoma, and respiratory syncytia and cells such as WI-38, chick embryo and duck embryo cells.
