Abstract
1450976 Insolubilized enzymes RHONE PROGIL SA 19 Aug 1974 [22 Aug 1973] 36399/74 Heading C3H In fixing an enzyme on a support by covalent bonding, the enzyme is reacted with a support possessing functional groups capable of reacting with the groups by which the enzyme can be fixed, in the presence of an anhydrous organic medium that does not dissolve the enzyme of the support, at a temperature of from 30 to 120 C. for, say, from 30 minutes to 2 hours. The reaction may be carried out under slightly increased pressure. The organic medium is exemplified by hexane, benzene, toluene, xylene, chloroform, carbon tetrachloride, dichloroethane, trichloroethane, trichloroethylene, perchloroethylene, dioxane, tetrahydrofuran and dimethylsulphoxide. The support may be of brick, silica, alumina, clay, sand, agarose, starch, polydextran, cellulose, polybutadiene, polystyrene, methacrylic acid copolymer, or a copolymer of maleic anhydride and ethylene. Examples 1 to 3 are directed to fixing pectinase, papain and trypsin, respectively, on activated brick particles of diameter between 0À5 and 0À8 mm. dispersed in boiling benzene or hexane. The brick particles were activated in a preliminary treatment with sulphuryl chloride. Example 4 relates to the fixing in benzene of ribonuclease on polybutadiene activated by reaction with acetyl chloride. According to Example 5 glycoamylase is fixed on particles of silica, which had been activated by the grafting of a silane with expoxy function, in boiling chloroform. In comparative experiments the bonding was effected in an aqueous medium.
