Abstract
The use of a therapeutically effective amount of (i) an einkorn wheat extract rich in a peptide of sequence (H-Cys-Gly-OH)2 (ii) a hexapeptide of sequence Gly-Pro-Gln-Gly-Pro-Gln, and (iii) a tripeptide of sequence Arg-Gly-Ser, in the manufacture of a cosmetic composition for treating skin conditions is described. The composition may be used for treating skin conditions selected from the group consisting of wrinkling, sagging, photodamaged skin. Topical application of the composition to human skin has a photo-protecting and regenerating effect on the skin as well as combating cellular energy decline.
Description
The present invention relates to compositions for application to human skin and to 5 their use in improving the condition and appearance of the skin. In particular the invention relates to topical compositions for combating the effects of aging on the skin.
Background to the Invention As the skin is the outermost aspect of our bodies it is subject to deterioration through environmental abuse (wind, air conditioning, central heating) or through the normal aging process on non-sun exposed areas (chronological aging) which may be accelerated by exposure of skin to sun (photoaging). As a result of this the consumer demands highly effective products to prevent the aging process.
In this respect consumers are increasingly seeking "anti-aging" cosmetic products which treat or delay the visible signs of aging skin such as wrinkles, lines, sagging, hyperpigmentation and age spots in addition to cosmetic products improving the appearance and condition of dry and flaky skin and to soothe irritated and itchy skin.
Skin care cosmetic and dermatological compositions for improving the condition and appearance of skin comprising peptides are well known in the art.
EP0505108 describes a composition for topical application for regulating wrinkles in mammalian skin comprising a safe and effective amount of peptide having the amino acid sequence Qa-(Xaa)r-Arg-Ser-Arg-Lys-(Xaa)n-Qc; and a safe and effective amount of a pharmaceutically acceptable carrier.
FR2846885 describes a topical cosmetic composition for smoothing wrinkles and fine lines containing a synergistic combination of peptide with sequence based on SNAP 25 (synaptosoome-associated protein of 25 kDa) and calcium channel inhibitor.
FR2873918 describes a cosmetic and/or dermatological and/or pharmaceutical composition comprising, as active ingredient, an effective amount of an extract from diploidic vegetal species belonging to the Triticum family to prevent or treat the cellular damages provoked by free radicals induced by atmospheric and/or ultraviolet pollutants. In particular, it describes the use of einkorn wheat extract (Triticum monococcum) as an active ingredient in cosmetic and/or pharmaceutical compositions.
US 2004/0142873 Al describes cosmetic or dermatological use of peptides for 5 promoting adhesion between skin cells. It describes a method for preparing a cosmetic or dermatological composition, of a sufficient amount of peptides of sequence (GlyPro-Gln)n-NH2, wherein: n ranges between 1 and 3, and the amino acids can be in the form L, D or DL. The compositions described have been designed to provide curative and/or preventative treatment for aging skin symptoms and to enhance skin appearance.
US 2006/0013794 Al (corresponding to FR2846883) describes the use of at least one peptide, with the sequence (AA)n-Arg-Gly-Ser-(AA)n, where (AA) is any amino acid or a derivative thereof and n= 0 to 3, as an active ingredient in or for the preparation of a cosmetic and /or dermatological and/or pharmaceutical preparation for treatment of the effects of cutaneous aging.
However, no prior art exists for a synergistic combination of peptides of exclusively natural origin for use in cosmetic products.
There is therefore a need for alternative effective cosmetic compositions for topical application to skin for treating and/or delaying the visible signs of aging and 0 photodamaged skin such as wrinkles, lines, sagging, hyperpigmentation, skin dryness and age spots.
It is therefore an object of the invention to provide an improved cosmetic composition comprising a combination of peptides of exclusively natural origin for treating and or preventing normal skin conditions due to chronological aging or photoaging, such as wrinkles, lines, sagging, hyperpigmentation and age spots, and/or of dry, flaky, itchy, irritated skin. ο ό ύ ί $ % Summary of the Invention Accordingly, the invention provides the use of a therapeutically effective amount of (i) an einkorn wheat extract rich in a peptide of sequence (H-Cys-Gly-OH)2, (ii) a hexapeptide of sequence Gly-Pro-Gln-Gly-Pro-Gln, and (iii) a tripeptide of sequence Arg-Gly-Ser, in the manufacture of a cosmetic composition for treating skin conditions. The cosmetic composition is suitable for use in conditions selected from the group consisting of wrinkling, sagging, photodamaged skin, dry skin, flaky skin, irritated skin, itchy skin and age spots.
The inventor has surprisingly and unexpectedly found that the use of a therapeutically effective amount of a combination of the above mentioned peptides, each of exclusively natural origin, and which each individually target different biological mechanisms demonstrates a synergistic effect in the treatment of skin conditions due to chronological aging or photoaging, such as wrinkles, lines, sagging, hyperpigmentation and age spots, and/or of dry, flaky, itchy, irritated skin.
In a preferred embodiment, each of the peptides used in the present invention is of exclusively natural origin. Preferably the triple peptide combination comprises a therapeutically effective amount of (i) einkorn wheat extract rich in a peptide of sequence (H-Cys-Gly-OH)2, (ii) hexapeptide of sequence Gly-Pro-Gln-Gly-Pro-Gln extracted from rice and (iii) at least one tripeptide of sequence Arg-Gly-Ser extracted from Saccharomyces Cerevisiae. It has been surprisingly found that effective treatment and prevention of skin conditions due to chronological aging or photoaging, such as wrinkles, lines, sagging, hyperpigmentation and age spots, and/or of dry, flaky, itchy, irritated skin may be obtained through the application of cosmetic compositions to the skin which comprise a specific synergistic combination of these three peptides.
Preferably, the therapeutically effective triple peptide combination comprises the peptides in an amount (i) 15-20 ppm of dipeptide dimmer from einkorn wheat extract, (ii) 45-55 ppm of the hexapeptide, and (iii) 15-20 ppm of the tripeptide.
Further preferably, the total amount of the peptide combination comprises between 0.0001% to 10% by weight of the composition, preferably between 0.001% to 5% by Ά -ι weight of the composition and more preferably between 0.01% to 3% by weight of the composition.
The combination of peptides used in accordance with the present invention suitably comprises an anti-oxidant peptide, a collagen promoting peptide and an energy boosting peptide.
The einkorn peptidic extract is known to promote a cellular antioxidant mechanism. It reinforces the activity of anti-oxidant enzymes such as Superoxide Dismutase (SOD) and catalase, thus favouring the elimination of Reactive Oxygen Species (ROS) within cells. It diminishes protein carbonylation and lipid peroxidation resulting from oxidative damages. It decreases DNA damage and increase cell viability.
The peptide of sequence (Gly-Pro-Gln)2, which is homologue to sequences found in human collagen IV and XVII, is known to promote dermal collagen synthesis, dermoepidermal junction regeneration and epidermal differentiation.
The peptide of sequence Arg-Gly-Ser binds signalling molecules implicated in the maintenance and regulation of cellular function and is known to trigger both calcium signalling and ATP synthesis, which act in synergy to increase energy rebound. Energy rebound is essential for cell physiology and represents the capacity to recover rapidly from physiological or environmental stress situations. In healthy and young skin, a balance exists between energy metabolites and the capacity for a cell to fulfil 0 energy requirements. Energy rebound allows a rapid and efficient recovery from energy-consuming processes. In aged skin, this balance is disrupted and energy rebound is significantly delayed or weakened. Therefore improving energy rebound helps the skin to preserve its structural and functional integrity and act against aging mechanisms.
The triple peptide combination provided a remarkable synergistic effect on collagen IV and pan-keratin expression compared to the use of the individual peptides alone.
In one embodiment, the invention provides a topical composition comprising: (a) a therapeutically effective amount of (i) an einkorn wheat extract rich in a peptide of IE Ο 6 Ο 1 5 4 sequence (H-Cys-Gly-OH)2, (ii) a hexapeptide of sequence Gly-Pro-GIn-GIy-Pro-Gln, and (iii) a tripeptide of sequence Arg-Gly-Ser, together with (b) a dermatologically acceptable carrier.
Such compositions are particularly useful for topical application to human skin for 5 cosmetically treating and/or preventing skin conditions selected from the group consisting of wrinkling, sagging, photodamaged skin, dry skin, flaky skin, irritated skin, itchy skin and age spots.
The composition according to the invention may comprise a dermatologically/cosmetically acceptable vehicle to act as a dilutant, dispersant or carrier for the peptides composition. The carrier may be selected from the group consisting of water, liquid or solid emollients, silicone oils, emulsifiers, solvents, humectants, thickeners, powders, propellants and the like.
The carrier will usually form from 5% to 99.9%, preferably from 25% to 80% by weight of the composition, and can, in the absence of other cosmetic adjuncts, form the balance of the composition.
The composition may also comprise other specific skin-benefit actives such as sunscreens, moisturising agents, skin lightening agents. The vehicle may also further include adjuncts such as fragrances, opacifiers, preservatives, colourants, buffers and other anti-aging ingredients. 0 The topical composition according to the present invention may be prepared in a manner well known in the art for preparing skin care products. The active components are generally incorporated in a dermatologically acceptable carrier in conventional manner. The active components can suitably first be dissolved or dispersed in a portion of the water or another solvent or liquid to be incorporated in the composition.
The preferred compositions are oil-in-water or water-in-oil emulsions.
The composition may be in the form of conventional skin-care products such as a cream, gel or lotion or the like. The composition can also be in the form of a cleanser e.g. a bath or shower gel, possibly containing a delivery system for the actives to It it Η (J 1 5 4 promote adherence to the skin during rinsing. The composition may also be a foundation or other colouring product e.g. mascara or lipstick. Most preferably the product is a "leave-on" topical product; a product to be applied to the skin without a deliberate rinsing step soon after its application to the skin.
The composition may be packaged in any suitable manner such as in ajar, a bottle, tube, roll-ball, or sachets.
The invention further provides a cosmetic method of treating/preventing skin conditions selected from the group consisting of wrinkling, sagging, photodamaged skin, dry skin, flaky skin, irritated skin, itchy skin and age spots, the method comprising applying to the skin a topical composition comprising (i) an einkorn wheat extract rich in a peptide of sequence (H-Cys-Gly-OH)2, (ii) a hexapeptide of sequence Gly-Pro-Gln-Gly-Pro-Gln, and (iii) a tripeptide of sequence Arg-Gly-Ser.
The method of the present invention may be carried out one or more times daily to the skin which requires treatment. The improvement in skin appearance will usually become visible after 3 to 6 months, depending on skin condition, the concentration of the active components used in the inventive method, the amount of composition used and the frequency with which it is applied. In general, a small quantity of the composition, for example from 0.1 to 5 ml is applied to the skin from a suitable container or applicator and spread over and/or rubbed into the skin using the hands or fingers or a suitable device. A rinsing step may optionally follow depending on whether the composition is formulated as a "leave-on" or a cleansing product.
The inventive compositions and methods thus provide anti-aging benefits which result in the promotion of smooth and supple skin with improved skin elasticity and a reduced or delayed appearance of wrinkles and aged skin with improved skin colour.
A general improvement in the appearance, texture and condition, in particular with respect to the radiance, clarity, and general youthful appearance of skin is achieved.
Thus the inventive methods and compositions provide a wide range of skin care benefits.
The term "treating" as used herein includes within its scope reducing, delaying and/or preventing the appearance of wrinkled, aged, photodamaged, dry and/or dry and ΪΪ06 0 1 5 4 irritated skin and generally enhancing the quality of skin by improving its appearance and texture, by preventing or reducing wrinkling and increasing flexibility, firmness, smoothness, suppleness and elasticity of the skin. The cosmetic compositions, methods and the use of the combination of peptides according to the invention may be useful for treating skin which is already in a wrinkled, aged, photodamaged, dry and irritated condition or for treating youthful skin to prevent or reduce deteriorative changes due to the normal aging but particularly the photoaging process.
Detailed Description of the Invention Skin aging is a complex process controlled by a number of factors broken down into intrinsic and extrinsic factors that include genetics, exposures to environmental stress (UV, mechanical stress), hormonal changes or metabolism. The combined results of these different factors are the deterioration of the structure of the skin, its function and its appearance (Rittie et al., 2002). The influence of the environment and UV irradiation in particular is very important in the process of skin aging (Rittie et al., 2002). It is now known that these effects are directly related to the flattening of the dermal-epidermal junction, to the degradation of the fiber system of the skin, the extracellular matrix (ECM) and to the alteration of epidermis.
The ECM is produced by fibroblasts and keratinocytes. It is composed of macromolecules including collagens, elastin, proteoglycans and fibronectin. Collagen and elastin are the major components of the ECM and are contained in tissues in the form of insoluble, rigid and stable fibers. They are tightly interwoven, organized in a network of microfibrils that imparts solidity and mechanical resistance to connective tissues. ECM thus acts like a physical support, but also carries out biological functions of intercellular exchange and communication that are essential for tight connection between the dermis and epidermis and the integrity of skin tissue.
Chronological aging or environmental stress (especially sunlight) destroys this balance. Cellular metabolic activity decreases and the renewal of collagen fibers are reduced. In parallel, enzymatic activities increase dramatically and result in an excessive and anarchic degradation of collagen fibers and a loss of structure in the ECM. The skin loses its suppleness and elasticity and becomes flaccid.
IE 1 54 Since the work of McCord and Fridovich in 1969 on superoxide dismutase, considerable interest has been focused on the relation between free radicals [superoxide (O2- and hydroxyl (OH-) anions and other reactive oxygenated species (ROS)] and aging of the skin. The origin of free radicals may be endogenous (activation of arachidonic acid metabolism, activation of phagocytosis or accumulation of reduced metabolites) or exogenous (radiation, UV from sunlight, air pollution, tobacco smoke, pesticides and more).
All cells have varying degrees of a chemical and enzymatic anti-free radical arsenal.
This enables them to exert a natural protection against these highly reactive molecules and to optimally control their deleterious effects. Cellular responses may differ, however, depending on the extent of the oxidative stress. Low quantities of ROS stimulate cell proliferation and the over expression of antioxidant enzymes that can adapt and react to the stress. When produced in excessive quantities, on the other hand, ROS attack a series of biological substrates. These defense systems are often overwhelmed and cannot handle an overpowering radical flux. This saturation phenomenon is accentuated with age. In this case, ROS cause cell damage that is often irreversible and can even lead to cell death (McCord et al., 1996). The major effects of this uncontrolled oxidative stress are intense, targeted and irreversible: oxidation of proteins and enzyme inactivation, DNA chain breaks and lipid peroxidation.
The three peptides used in accordance with the present invention are preferably of natural origin. The combination of peptides used comprises an anti-oxidant peptide, a collagen promoting peptide and an energy boosting peptide. The triple peptide combination used comprises a therapeutically effective amount of (i) einkorn wheat extract rich in a peptide of sequence (H-Cys-Gly-OH)2, (ii) hexapeptide of sequence Gly-Pro-Gln-Gly-Pro-Gln extracted from rice and (iii) at least one tripeptide of sequence Arg-Gly-Ser extracted from Saccharomyces Cerevisiae. Most preferably the combination of the three individual peptides comprises (i) 15-20 ppm of the dipeptide dimer from the einkorn wheat extract specified above, (ii) 45-55 ppm of the hexapeptide specified above, and (iii) 15-20 ppm of the tripeptide specified above.
Therapeutically effective amounts of the triple peptide combination described are used in the manufacture of the topical compositions described herein. Preferably the •j I 5 * total amount of the triple peptide combination is present in an amount between 0.0001% to 10% by weight of the composition. More preferably the amount is from 0.001% to 5% by weight of the composition and most preferably from 0.01% to 3% by weight of the composition, in order to maximise benefits at a minimum cost.
The inventor has surprisingly found that the combination of each of the above peptides produces a remarkable synergistic effect on collagen IV and pan-keratin expression compared to the use of the individual peptides alone.
In order that the present invention may be more readily understood, the following examples are given, by way of illustration only.
Example 1 Effect of the triple peptide combination at protecting human fibroblasts from UVA induced oxidative stress.
Active oxygen species and other free radicals are presumed to participate in actinic aging. In order to determine the protective activities of the combination of the three peptides, different conditions of UVA-induced oxidative stress were conducted.
Cultures of human fibroblasts were artificially subjected to UVA stresses. The free radicals formed during those stresses were measured. The anti-oxidant activity of the triple peptide combination tested was determined by different methods.
Fibroblasts were grown to 70% of confluence, were treated with 1% of peptide combination for 24h prior to and during UVA exposure. Before irradiation, the medium was removed and cells were rinsed with 5 ml of PBS. Fibroblasts were then irradiated in 5 ml PBS in uncovered dishes with 5 J/cm2 using lamp type Vilber Lourmat (VL-215M fisher Bioblock). After irradiation, cells were treated as described previously and replaced in incubator for 4h (SOD, catalase & carbonylation) or for 24h (lipid peroxidation).

The objective of the study was to investigate the effect of the triple peptide 5 combination on the synthesis of collagen IV and pan-keratin. 20μ1 of cream A (peptide alone) or B (triple peptide complex) were applied twice a day on the epidermal side of 6 mm fresh punch biopsies. Treated & untreated skin samples were cultured for 24h. Double immunofluorescence staining was performed using a specific antibody directed against collagen IV and pankeratin, followed by the specific secondary antibody covalently conjugated to Alexa Fluor® 488 (green). Nuclear staining was performed using DAPI (blue).

This increase has been observed within 24 hours of the application of the creams.
This study demonstrates that the three peptides act in synergy on the synthesis of collagen IV and pankeratin in a stronger manner compared to each peptide alone.
Example 5 Anti-wrinkle effect of the triple peptide combination.
The aim of this study was to quantify in vivo the anti-wrinkle efficacy of the products N°472 (placebo + 3% of triple peptide combination) and 638 (placebo) on the crow's feet. The study was conducted double blind on 22 volunteers presenting wrinkles on the crow's feet. The area of application for the products is randomized (left crow’s feet/right crow’s feet). Replicas of the two crow's feet were made before and after 28 days of twice daily application.
The anti-wrinkle effect of the products N°472 and 638 was analyzed by observing replicas with the help of a profilometer equipped with an image analyzer. Silicone polymer replicas were made before and after application of the product and examined by S.I.A. (silicone image analysis). Grazing light at an incidence of 35° casts shadows on the surface of the impression that are then observed with a CCD camera connected to a computer. The field examined was 1 cm2. The gray level analysis of the resulting computer image provides the following parameters using QUANTIRIDES and QUANTILINES (MONADERM) software: - The total wrinkle surface - The average length of wrinkles - The average depth of wrinkles The results were calculated using the means of variations of wrinkles parameters observed under the effect of the product compared to DO.
The results are shown in table 1: if - 4 ς A Table 1: Anti-wrinkle effect of the products 472 and 638 • Total wrinkled surface Product 472 Product 638 D28-D0 D28-D0 Mean SEM Mean SEM s -2,4 7.1 1,8 7.9 P 0,0368 • Average length Product 472 Product 638 D28-D0 D28-D0 Mean SEM Mean SEM L -0,1 0.2 -0,1 0.3 P 0,02845 » Average depth Product 472 Product 638 D28-D0 D28-D0 Mean SEM Mean SEM D -2,5 18.1 7,7 18.2 P 0,0327 After 28 days of twice daily application, and in the conditions of this study: - The product NO638 (placebo) tends to increase the different wrinkle parameters.
- The product N°472 (placebo + 3% of triple peptide combination) decreases significantly (P<0.05) the different wrinkle parameters. : , 5 5 Example 6 Effect of the triple peptide combination on skin elasticity.
The aim of this study was to quantify in vivo the effect of the products N°472 (placebo + 3% of triple peptide combination) and 638 (placebo) on skin elasticity. The study was conducted double blind on the face of 22 volunteers. The area of application for the products is randomized (left face/right face). Measurements were made on the cheek before and after 28 days of twice-daily treatment using the same instrument and principles as previously described but this time measuring the parameter of average roughness (Ra).
The results were calculated using the means of variations of average roughness observed under the effect of the product compared to DO.
The results are shown in table 2: Table 2: Effect of the products 472 and 638 on skin elasticity • Average roughness Product 472 Product 638 D28-D0 D28-D0 Mean SEM Mean SEM Ra -0,483 0,409 P 0,01365 After 28 days of application, and in the conditions of this study: - The product N°638 (placebo) tends to increase the Ra parameter, which represent the elasticity of the skin.
- The product N°472 (placebo + 3% of triple peptide combination) decreases 2 0 significantly (P<0.05) the Ra parameter which represent the elasticity of the skin.
