Abstract
E' descritto un procedimento per la purificazione di big endotelina (Big ET) da estratti grezzi che comprende le fasi di:- esprimere mediante metodologie di ingegneria genetica in cellule ospiti la proteina Big ET;- lisare dette cellule ospiti e trattarle con un enzima proteolitico, in modo da avere un estratto proteolizzato;- passare detto estratto proteolizzato su una colonna cromatografica di affinità, preparata immobilizzando un ligando specifico per la Big ET comprendente peptidi idropaticamente complementari alla Big ET, costituiti da aminoacidi in conformazione D e equilibrata in un opportuno tampone salino;- lavare da detta colonna il materiale non specificamente legato con un opportuno tampone di lavaggio;- eluire il materiale specificamente trattenuto dalla colonna con un opportuno tampone di eluizione.
