Claims (10)

  1. RIVENDICAZIONI 1. Sistema (10) per rivelare una sequenza nucleotidica bersaglio (3) a singolo filamento comprendente: - almeno una prima sonda (1) di acido nucleico da 10 a 14 basi, all’estremità 5’ della quale è legato almeno un fluoroforo (11); - almeno una seconda sonda (2) di acido nucleico da 35 a 50 basi, comprendente, dall’estremità 5’ all’estremità 3’: - un primo segmento (21) di sequenza nucleotidica complementare alla prima sonda (1) di acido nucleico, - almeno un quencher (23), e - un secondo segmento (22) di sequenza nucleotidica complementare ad almeno parte della sequenza nucleotidica bersaglio (3). 2. Sistema (10) secondo la rivendicazione 1, in cui è rispettata la seguente relazione: <|>∆Gibr. bers.3 − sonda2<|>><|>∆Gibr. sonda1 − sonda2<|>, dove - ∆G ibr.bers.3-sonda2 è l’energia libera di formazione del duplex tra sequenza nucleotidica bersaglio (3) e seconda sonda (2) di acido nucleico, e - ∆G ibr.sonda1-sonda2 è l’energia libera di formazione del duplex tra prima sonda (1) di acido nucleico e seconda sonda (2) di acido nucleico. 3. Sistema (10) secondo la rivendicazione 2, in cui 10 Kcal/mol ><|>∆Gibr. bers.3 − sonda2<|>−<|>∆Gibr. sonda1 − sonda2<|>> 50 Kcal/mol 4. Sistema (10) secondo la rivendicazione 3, in cui la sequenza nucleotidica bersaglio (3) a singolo filamento è DNA e 35 Kcal/mol ><|>∆Gibr. bers.3 − sonda2<|>−<|>∆Gibr. sonda1 − sonda2> 45 Kcal/mol. 5. Sistema (10) secondo la rivendicazione 3, in cui la sequenza nucleotidica bersaglio (3) a singolo filamento è miRNA e 10 Kcal/mol ><|>∆Gibr. bers.3 − sonda2<|>−<|>∆Gibr. sonda1 − sonda2> 25 Kcal/mol. 6. Sistema (10) secondo una qualsiasi delle rivendicazioni precedenti, in cui l’almeno una prima sonda (1) di acido nucleico ha lunghezza da 11 a 13 basi. 7. Sistema (10) secondo una qualsiasi delle rivendicazioni precedenti, in cui la sequenza nucleotidica bersaglio (3) a singolo filamento ha una lunghezza da 15 a 100 basi. 8. Sistema (10) secondo la rivendicazione 7, in cui la sequenza nucleotidica bersaglio (3) a singolo filamento ha una lunghezza da 20 a 40 basi. 9. Sistema (10) secondo una qualsiasi delle rivendicazioni precedenti, in cui la sequenza nucleotidica bersaglio (3) a singolo filamento è in concentrazione da 1·10<-17>M a 1·10<-19>M. 10. Sistema (10) secondo una qualsiasi delle rivendicazioni precedenti, comprendente inoltre almeno una microparticella (4) legata covalentemente all’estremità 3’ dell’almeno una prima sonda (1) di acido nucleico. TITLE: PROBE SYSTEM FOR DETECTING A SINGLE STRAND TARGET NUCLEOTIDE SEQUENCE 1. A system (10) for detecting a single strand target nucleotide sequence (3) comprising: - at least one first nucleic acid probe (1) from 10 to 14 bases, to the 5’ end of which at least one fluorophore (11) is bound; - at least one second nucleic acid probe (2) from 35 to 50 bases, comprising, from the 5’ to the 3’ end: - a first segment (21) having a nucleotide sequence complementary to the first nucleic acid probe (1), - at least one quencher (23), and - a second segment (22) having a nucleotide sequence complementary to at least part of the target nucleotide sequence (3).
  2. 2. The system (10) according to claim 1, wherein the following relation is met: , where: - ∆G hybr.target3-probe2 is the free energy of duplex formation between the target nucleotide sequence (3) and the second nucleic acid probe (2), and - ∆G hybr.probe1-probe2 is the free energy of duplex formation between the first nucleic acid probe (1) and the second nucleic acid probe (2).
  3. 3. The system (10) according to claim 2, wherein: 10 Kcal/mol 50 Kcal/mol
  4. 4. The system (10) according to claim 3, wherein the single strand target nucleotide sequence (3) is DNA and 35 Kcal/mol 45 Kcal/mol.
  5. 5. The system (10) according to claim 3, wherein the single strand target nucleotide sequence (3) is miRNA and 10 Kcal/mol 25 Kcal/mol.
  6. 6. The system (10) according to any of the preceding claims, wherein the at least one first nucleic acid probe (1) has a length from 11 to 13 bases.
  7. 7. The system (10) according to any of the preceding claims, wherein the single strand target nucleotide sequence (3) has a length from 15 to 100 bases.
  8. 8. The system (10) according to claim 7, wherein the single strand target nucleotide sequence (3) has a length from 20 to 40 bases.
  9. 9. The system (10) according to any of the preceding claims, wherein the single strand target nucleotide sequence (3) is in a concentration from 1·10<-17>M to 1·10<-19>M.
  10. 10. The system (10) according to any of the preceding claims, also comprising a microparticle (4) bound covalently to the 3’ end of the at least one first nucleic acid probe (1).
