Claims (1)
- 【特許請求の範囲】 1.約445アミノ酸を有するNPY−Y5受容体または機能的に等価なその断 片をコードする、単離されたDNA分子。 2.前記DNA分子がヒト、マウスまたはラットNPY−Y5受容体をコードす る、請求項1に記載の単離されたDNA分子。 3.前記DNA分子がヒトNPY−Y5受容体をコードする、請求項2に記載の 単離されたDNA分子。 4.NPY−Y5受容体をコードする単離されたDNA分子であって、下記に示 すヌクレオチド配列と少なくとも80%相同である該DNA分子: (i) 図1のヌクレオチド6291から7625、 (ii) 図2のヌクレオチド63から1397、 (iii)図3のヌクレオチド115から1449、または (iv) 図4のヌクレオチド73から1470。 5.NPY−Y5受容体をコードする単離されたDNA分子であって、下記に示 すヌクレオチド配列と少なくとも95%相同である該DNA分子: (i) 図1のヌクレオチド6291から7625、 (ii) 図2のヌクレオチド63から1397、 (iii)図3のヌクレオチド115から1449、または (iv) 図4のヌクレオチド73から1470。 6.NPY−Y5受容体をコードする単離されたDNA分子であって、図1のヌ クレオチド6291から7625に示すヌクレオチド配列に実質的に一致するヌクレオチ ド配列を有する該DNA分子。 7.NPY−Y5受容体をコードする単離されたDNA分子であって、図2のヌ クレオチド63から1397に示すヌクレオチド配列に実質的に一致するヌクレオチド 配列を有する該DNA分子。 8.NPY−Y5受容体をコードする単離されたDNA分子であって、図3のヌ クレオチド115から1449に示すヌクレオチド配列に実質的に一致するヌクレオチ ド配列を有する該DNA分子。 9.NPY−Y5受容体をコードする単離されたDNA分子であって、図4のヌ クレオチド73から1470に示すヌクレオチド配列に実質的に一致するヌクレオチド 配列を有する該DNA分子。 10.上記の請求項のいずれか一項に記載のDNA分子を含むプラスミドまたは発 現ベクター。 11.請求項1から9のいずれか一項に記載のDNA分子によって形質転換された 宿主細胞。 12.前記細胞が哺乳動物細胞または細菌細胞である、請求項11に記載の宿主細胞 。 13.前記細胞がチャイニーズハムスター卵巣(CHO)細胞、ヒト胎児腎臓(H EK)293細胞または昆虫Sf9細胞である、請求項12に記載の宿主細胞。 14.前記細胞が細胞表面にNPY−Y5受容体を発現する、請求項11から13のい ずれか一項に記載の宿主細胞。 15.実質的に純粋な形態のNPY−Y5受容体。 16.前記受容体が約445アミノ酸からなる、請求項15に記載のNPY−Y5受容 体。 17.前記NPY−Y5が図5に示すアミノ酸配列のいずれか一つに実質的に一致 するアミノ酸配列を有する、請求項15または16に記載のNPY−Y5受容体。 18.請求項15から17のいずれか一項に記載のNPY−Y5受容体に特異的に結合 することができる抗体。 19.請求項1から9のいずれか一項に記載のDNA分子によって形質転換された 非ヒト動物。 20.NPY−Y5受容体のアゴニストまたはアンタゴニスト作用物質を検出する 方法であって、請求項15から17のいずれか一項に記載のNPY−Y5受容体また は請求項1から9のいずれか一項に記載のDNA分子によって形質転換され、そ して該分子を発現する細胞を、NPY−Y5受容体の活性化を可能とする条件下 で被験作用物質に接触させる工程、及びNPY−Y5受容体活性の増大または減 少を検出する工程を包含する、方法。 21.請求項1から9のいずれか一項に記載のDNA分子内の独特な配列に特異的 にハイブリダイズすることができる、10個以上のヌクレオチドからなるヌクレオ チド配列を含む核酸プローブ。 22.NPY−Y5受容体をコードするmRNA分子に特異的にハイブリダイズし て該mRNA分子の翻訳を阻止することができるヌクレオチド配列を含むアンチ センス核酸分子。
