Claims (1)
- 【特許請求の範囲】 1. (a)腫瘍細胞株を得て、 (b)該腫瘍細胞株を、未修飾の腫瘍細胞株と比べて増加したレベルのサイト カインを産生することができるように修飾し、 (c)該腫瘍細胞株を得た腫瘍と同じタイプの腫瘍である少なくとも1つの腫 瘍を有する哺乳類宿主に、該腫瘍細胞株(該腫瘍細胞株は該宿主と同種異系であ り、かつMHCがマッチしていない(not MHC-matched))を投与することを含 む癌の治療方法。 2. 該腫瘍細胞株を該腫瘍のごく近傍に投与する請求の範囲1記載の方法。 3. 該腫瘍細胞株を投与の前に放射線照射する請求の範囲1記載の方法。 4. 該腫瘍細胞株を、免疫原性を増強するために投与の前に処理する請求の範 囲1記載の方法。 5. 該処理が遺伝子操作または非特異的アジュバントとの混合を含む請求の範 囲4記載の方法。 6. (a)哺乳類宿主から腫瘍のサンプルを得て、 (b)該腫瘍サンプルから単細胞懸濁液を形成させ、 (c)該腫瘍細胞をペレットにし、 (d)該腫瘍細胞をプレーティングすることにより該腫瘍細胞株を得る請求の 範囲1記載の方法。 7. 該腫瘍細胞株が腫瘍遺伝子または腫瘍抑制遺伝子中に変異を含む請求の範 囲1記載の方法。 8. 該サイトカインがGM−CSFである請求の範囲1記載の方法。 9. (a)膵臓腫瘍細胞株を得て、 (b)該腫瘍細胞株を、未修飾の腫瘍細胞株と比べて増加したレベルのサイト カインを産生することができるように修飾し、 (c)少なくとも1つの膵臓腫瘍を有する哺乳類宿主に、該腫瘍細胞株(該腫 瘍細胞株は該宿主と同種異系であり、かつMHCがマッチしていない)を投与す ることを含む膵臓癌の治療方法。 10. 該腫瘍細胞株を該腫瘍のごく近傍に投与する請求の範囲9記載の方法。 11. 該腫瘍細胞株を投与の前に放射線照射する請求の範囲9記載の方法。 12. 該腫瘍細胞株を、免疫原性を増強するために投与の前に処理する請求の 範囲9記載の方法。 13. 該処理が遺伝子操作または非特異的アジュバントとの混合を含む請求の 範囲12記載の方法。 14. (a)哺乳類宿主から膵臓腫瘍のサンプルを得て、 (b)該腫瘍サンプルから単細胞懸濁液を形成させ、 (c)該腫瘍細胞をペレットにし、 (d)該腫瘍細胞をプレーティングすることにより該膵臓腫瘍細胞株を得る請 求の範囲9記載の方法。 15. 該腫瘍細胞株が腫瘍遺伝子または腫瘍抑制遺伝子中に変異を含む請求の 範囲9記載の方法。 16. 該変異がγas 遺伝子中にある請求の範囲15記載の方法。 17. 該サイトカインがGM−CSFである請求の範囲9記載の方法。 18. 実質的に精製された膵臓腫瘍細胞株。 19. 該腫瘍細胞株がPanc 10.5.92である請求の範囲18記載の腫瘍細胞株。 20. 該腫瘍細胞株が、未修飾の腫瘍細胞株と比べて増加したレベルのサイト カインを産生することができるように修飾されている請求の範囲18記載の腫瘍 細胞株。 21. 該サイトカインがGM−CSFである請求の範囲20記載の腫瘍細胞株 。 22. 医薬的に許容される担体および請求の範囲18記載の腫瘍細胞株の細胞 を含有する医薬組成物。 23. 医薬的に許容される担体および請求の範囲19記載の腫瘍細胞株の細胞 を含有する医薬組成物。 24. (a)哺乳類宿主から膵臓腫瘍のサンプルを得て、 (b)該腫瘍サンプルから単細胞懸濁液を形成させ、 (c)該腫瘍細胞をペレットにし、 (d)該腫瘍細胞をプレーティングすることを含む、請求の範囲18記載の腫 瘍細胞株を得る方法。 25. 該腫瘍細胞を、胎児血清、インスリン、およびインスリン様増殖因子1 および2を含有する増殖培地にプレーティングする請求の範囲24記載の方法。 26. 胎児血清、インスリン、およびインスリン様増殖因子1および2を含有 する膵臓腫瘍細胞の培養用増殖培地。 27. 該培地がインスリンを約0.1〜約1.0U/mlの濃度で、インスリ ン様増殖因子1を約0.005〜約0.05μg/mlの濃度で、およびインス リン様増殖因子2を約0.005〜約0.05μg/mlの濃度で含有する請求 の範囲26記載の培地。
