Claims (1)
- 【特許請求の範囲】 1.外科手術用及び/又は治療用の、特に、生体組織同士又は生体組織と移植 された生体材料とを接着するための、生体適合性、生体吸収性、及び無毒性の接 着剤組成物であって、酸化でんぷん又はそれに類似の安定特性を有するポリアル デヒドのごとき、天然物由来の少なくとも1つの生分解性マクロモレキュラーポ リアルデヒドの水溶液を含むことを特徴とする接着剤組成物。 2.更に、水性媒体に溶解した、下記のものから選ばれたコラーゲンベースの 組成分を含有することを特徴とする請求項1記載の接着剤組成物。 −少なくとも部分的にヘリカル構造を消失した、主としてα鎖からなる加水分 解されてないコラーゲン、 −濃度5%以下の天然コラーゲン。 3.自然に得られた酸性pHの、天然物由来の少なくとも1つの生分解性マクロ モレキュラーポリアルデヒド水性溶液を含有することを特徴とする請求項1又は 2記載の接着剤組成物。 4.ポリアルデヒドが酸化された多糖類又はムコ多糖類からなることを特徴と する請求項1〜3のいずれかに記載の接着剤組成物。 5.多糖類又はムコ多糖類が、でんぷん、デキストラン、かんてん、セルロー ズ、キチン、キトサン、アルギン酸、グリコサミノグリカン、ヒアルロン酸若し くはコンドロイチン硫酸、又はそれらの誘導体を含む群から選ばれることを特徴 とする請求項1〜4のいずれかに記載の接着剤組成物。 6.多糖類がでんぷん及びデキストランから選択されたもの、又好ましくはで んぷんであることを特徴とする請求項1〜5のいずれかに記載の接着剤組成物。 7.ポリアルデヒドが、過ヨウ素酸又はその塩によって酸化された多糖類又は ムコ多糖類からなることを特徴とする請求項1〜6のいずれかに記載の接着剤組 成物。 8.多糖類が約10,000〜200 万ダルトンの分子量を有することを特徴とする請 求項1〜7のいずれかに記載の接着剤組成物。 9.1又は複数のポリアルデヒドの濃度 0.5〜5重量%水溶液を含有すること を特徴とする請求項1〜8のいずれかに記載の接着剤組成物。 10.酸性の凍結乾燥した形態のポリアルデヒドを水又は生理的緩衝剤を用いた 水に溶解することにより水溶液を得ることを特徴とする請求項1〜9のいずれか に記載の接着剤組成物。 11.少なくとも部分的にヘリカル構造を消失した、加水分解されてない、濃度 10〜20%、好ましくは15〜18%のコラーゲン溶液を含有することを特徴とする請 求項2〜10のいずれかに記載の接着剤組成物。 12.濃度 0.2〜5%、好ましくは1〜3%の天然コラーゲン水溶液を含有する ことを特徴とする請求項2〜10のいずれかに記載の接着剤組成物。 13.混合に供する、コラーゲンに対するポリアルデヒドの割合が0.5〜10重量 %、好ましくは3〜10重量%であることを特徴とする請求項2〜12のいずれかに 記載の接着剤組成物。 14.1〜2日と30〜60日との間に身体に吸収可能であることを特徴とする請求 項1〜13のいずれかに記載の接着剤組成物。 15.組織及び/又は生体材料に用いるための、外科手術用及び/又は治療用の 生体適合性、生体吸収性、及び無毒性接着剤組成物の製造方法において、架橋に 先立って、水性媒体に溶解した下記のものから選ばれたコラーゲンベースの組成 分と、 −少なくとも部分的にヘリカル構造を消失した、本質的にα鎖からなる加水分 解されてないコラーゲン、 −濃度5%以下の天然コラーゲン、 酸化でんぷん又は類似の安定特性を有するポリアルデヒドのごとき、天物由来 の少なくとも一つの生分解性マクロモレキュラーポリアルデヒドの水溶液とを混 合する工程を含むことを特徴とする製造方法。 16.コラーゲンベースの組成分の溶液及びマクロモレキュラーポリアルデヒド 溶液が、請求項2〜13のいずれかに規定されたものであることを特徴とする請求 項15に記載の製造方法。 17.混合物が20〜45℃の温度で製造されることを特徴とする請求項15又は16の いずれかに記載の製造方法。 18.混合物が37〜42℃の温度で製造されることを特徴とする請求項17記載の製 造方法。 19.混合物が生理的、中性pHで製造されることを特徴とする請求項15〜18のい ずれかに記載の製造方法。 20.架橋が5分以内で行われることを特徴とする請求項15〜19のいずれかに記 載の製造方法。 21.外科手術用及び/又は治療用、特に、生体組織同士又は生体組織と前記ポ リアルデヒドに対するアミン含有反応性官能基を有する移植された生体材料とを 接着する、生体適合性、生体吸収性、及び無毒性の接着剤を得るための、酸化で んぷん又は類似の安定特性を有するポリアルデヒドのごとき天然物由来の生分解 性マクロモレキュラーポリアルデヒドの水溶液の使用。 22.水性媒体に溶解した下記のものから選ばれたコラーゲンベースの組成分を 組み合わせた請求項21記載の使用。 −少なくとも部分的にヘリカル構造を消失した、主としてα鎖か らなる加水分解されてないコラーゲン、 −濃度5%以下の天然コラーゲン。 23.請求項1〜14のいずれかに規定された接着剤組成物を得るための請求項21 及び22のいずれかに記載の使用。 24.外科手術用及び/又は治療用の生体適合性、生体吸収性、及び無毒性の接 着剤組成物を含有するキットにおいて、 −酸化でんぷん又は類似の安定特性を有するポリアルデヒドのごとき天然物由 来の少なくとも1つの生分解性クロモレキュラーポリアルデヒド含有の水溶液を 含有することを特徴とするキット。 25.シリンジの形態であることを特徴とする請求項24記載のキット。 26.濃度0.5〜5重量%である1又は複数のポリアルデヒド水溶液を含有する ことを特徴とする請求項24又は25記載のキット。 27.更に下記のものを含有することを特徴とする請求項24記載のキット: −下記のものから選ばれたコラーゲンベースの組成分の水溶液、 ・少なくとも部分的にヘリカル構造を消失した、主としてα鎖からなる加水 分解されてないコラーゲン、 ・濃度5%以下の天然コラーゲン、 −酸化でんぷん又は類似の安定特性を有するポリアルデヒドのごとき、天然物 由来の少なくとも1つの生体分解性マクロモレキュラーポリアルデヒドの水溶液 、 −前記各溶液を即座に混合するための混合手段。 28.混合手段を接合した、一方のシリンジがコラーゲンベースの組成分溶液を 内蔵し、又他方のシリンジがマクロモレキュラーポリアルデヒド溶液を内蔵する 2本のシリンジで構成されることを特徴とする請求項27記載のキット。 29.自然に得られた酸性pHの天然物由来の少なくとも1つの生分解性マクロモ レキュラーポリアルデヒド含有の水溶液を含むことを特徴とする請求項24〜28の いずれかに記載のキット。 30. 0.5〜5%ポリアルデヒド溶液を含むことを特徴とする請求項27及び29の いずれかに記載のキット。 31.ポリアルデヒド溶液を、酸性の凍結乾燥形態のポリアルデヒドを水又は生 理的緩衝剤を用いた水に溶解して得ることを特徴とする請求項24〜30のいずれか に記載のキット。 32.少なくとも部分的にヘリカル構造を消失した、主としてα鎖からなる加水 分解されてないコラーゲンの10〜20%、好ましくは15〜18%溶液を含むことを特 徴とする請求項24〜31のいずれかに記載のキット。 33.一方で、少なくとも部分的にヘリカル構造を消失した、主としてα鎖から なる加水分解されてないコラーゲンの18%水溶液と、他方で酸化でんぷんの3% 溶液とを、1/24の割合で含むことを特徴とする請求項27〜32のいずれかに記載 のキット。 34.濃度 0.2〜5、好ましくは1〜3%の天然コラーゲン水溶液を含むことを 特徴とする請求項27〜31のいずれか記載のキット。 35.1の溶液又は複数の溶液が+1〜25℃、好ましくは+2℃〜+8℃の温度 に維持されることを特徴とする請求項27〜34のいずれかに記載のキット。 36.請求項1〜14のいずれかに記載の組成物を含むことを特徴とする請求項24 〜35のいずれかに記載のキット。 37.生体材料と生体組織とを接着する方法において、酸化でんぷん又は類似の 安定特性を有するポリアルデヒドのごとき、天然物由来の少なくとも1つの生分 解性マクロモレキュラーポリアルデヒドを含有する水溶液を、20〜45℃の温度で 、前記生体組織及び/又は 生体材料に適用し、その生体材料は前記ポリアルデヒドに反応性のある官能基、 特にアミン含有官能基を有することを特徴とする接着方法。 38.コラーゲンで作られたレンズを接着するための請求項37記載の方法。 39.生体組織同士又は生体組織と移植された生体材料とを接着する方法におい て: −下記のものから選ばれたコラーゲンベースの組成分の水溶液、 ・少なくとも部分的にヘリカル構造を消失した、主としてα鎖からなる加水 分解されてないコラーゲン、 ・濃度5%以下の天然コラーゲン、及び −酸化でんぷん又は類似の安定特性を有するポリアルデヒドのごとき、天然物 由来の少なくとも1つの生体分解性マクロモレキュラーポリアルデヒドの水溶液 を生理的、中性pHで混合し、 −得られたゲルを速やかに前記生体組織及び/又は前記ポリアルデヒドに反応 性のある官能基、特にアミン含有官能基を有する前記生体材料に、20〜45℃の温 度で適用し、 −その混合物を重合させることを特徴とする接着方法。 40.自然に得られた酸性pHのポリアルデヒド水溶液を使用する請求項37〜39の いずれかに記載の方法。 41.あらかじめpHを中性にしたポリアルデヒド水溶液を使用する請求項37〜39 のいずれかに記載の方法。 42.少なくとも部分的にヘリカル構造を消失した、主としてα鎖からなる加水 分解されてないコラーゲンの場合、温度37〜42℃で適用することを特徴とする請 求項39〜41のいずれかに記載の方法。 43.天然コラーゲンの場合、室温で適用することを特徴とする請求項39〜41の いずれかに記載の方法。 44.請求項1〜14のいずれかに記載の組成物を使用することを特徴とする請求 項37〜43のいずれかに記載の方法。 45.請求項24〜36のいずれかに記載のキットを使用することを特徴とする請求 項37〜44のいずれかに記載の方法。 46.手術後の癒着を回避するための請求項37〜45のいずれかに記載の方法。
