Claims (1)
- 【特許請求の範囲】 1.ナタネシステインプロテアーゼ遺伝子プロモーターのDNA配列を含むプロ モーターであって、該プロモーターが、図19、20、もしくは21に示すDNA配列の 少なくとも一部、またはそれとの実質的な相同性を有する配列、あるいはその改 変体を含む、プロモーター。 2.図19に示すDNA配列の少なくとも一部、またはそれとの実質的な相同性を 有する配列、あるいはその改変体を含む、プロモーター。 3.図20に示すDNA配列の少なくとも一部、またはそれとの実質的な相同性を 有する配列の少なくとも一部、あるいはその改変体を含む、プロモーター。 4.図21に示すDNA配列の少なくとも一部、またはそれとの実質的な相同性を 有する配列の少なくとも一部、あるいはその改変体を含む、プロモーター。 5.目的の遺伝子に作動可能に連結された請求項1〜4のいずれかに記載のプ ロモーターを含む、組換えDNA構築物。 6.細胞機能を破壊し得る産物をコードするディスラプター遺伝子、および該 ディスラプター遺伝子についての請求項1〜4のいずれかに記載のプロモーター を含む、植物において機能的な組換えDNA構築物であって、該ディスラプター遺 伝子が、化学インデューサーの外部適用によって誘導可能であるプロモーターを 含む外部調節可能な遺伝子制御領域に機能的に連結されそしてこれによって制御 される、組換えDNA構築物。 7.請求項6に記載の組換えDNA構築物であって、前記誘導性プロモーターが 、リプレッサータンパク質遺伝子に機能的に連結されそしてこれを制御し、そし て前記ディスラプター遺伝子プロモーターが、該リプレッサータンパク質によっ て 認識されるオペレーター配列を含み、その結果前記インデューサーの存在下、該 オペレーター配列と相互作用する該リプレッサータンパク質が産生され、それに よって第2のプロモーターを無能にしそして該ディスラプター遺伝子の発現を阻 害する、組換えDNA構築物。 8.請求項6または7に記載の組換えDNA構築物であって、前記ディスラプタ ー遺伝子がヌクレオチド配列であり、これが植物発生に必須である内因性植物遺 伝子、または植物上に所望の特徴を与える遺伝子にセンス配向であるか、あるい は内因性植物遺伝子の部分センス配列を含む、組換えDNA構築物。 9.前記ディスラプター遺伝子が、植物発生に必須である内因性植物遺伝子ま たは植物に所望の特徴を与える遺伝子に対してアンチセンス配向であるヌタレオ チド配列である、請求項6または7に記載の組換えDNA構築物。 10.前記内因性植物遺伝子が、種子発芽または初期実生発生に必須である、 請求項8または9に記載の組換えDNA構築物。 11.前記外部調節可能な遺伝子制御領域が、グルタチオンS-トランスフェラ ーゼ系、Alc系、またはエクジソン系からの化学的誘導性遺伝子プロモーター配 列である、請求項6〜10のいずれかに記載の組換えDNA構築物。 12.前記リプレッサータンパク質遺伝子が、細菌リプレッサーをコードする 、請求項8〜11のいずれかに記載の組換えDNA構築物。 13.前記リプレッサータンパク質遺伝子が、lacリプレッサー、あるいは434 、P22、またはラムダバクテリオファージによって使用されるリプレッサーをコ ードする、請求項12に記載の組換えDNA構築物。 14.前記ディスラプター遺伝子またはディスラプタープロモーターが、「偽 オペレーター」を含む、請求項6〜13のいずれかに記載の組換えDNA構築物。 15.前記ディスラプター遺伝子が、細胞傷害性遺伝子である、請求項6〜1 4のいずれかに記載の組換えDNA構築物。 16.前記ディスラプター遺伝子が、リコンビナーゼ、またはリコンビナーゼ 認識部位に隣接するヌクレオチド配列を切除するために適合されるトランスポゼ ースをコードする、請求項6〜14のいずれかに記載の組換えDNA構築物。 17.図12に示される配列の少なくとも一部、またはそれとの実質的な相同性 を有する配列、あるいはその改変体を含み、そしてシステインプロテアーゼをコ ードする、DNA。 18.図13に示される配列の少なくとも一部、またはそれとの実質的な相同性 を有する配列、あるいはその改変体を含み、そしてシステインプロテアーゼをコ ードする、DNA。 19.図14に示される配列の少なくとも一部、またはそれとの実質的な相同性 を有する配列、あるいはその改変体を含み、そしてシステインプロテアーゼをコ ードする、DNA。 20.図15に示される配列の少なくとも一部、またはそれとの実質的な相同性 を有する配列、あるいはその改変体を含み、そしてシステインプロテアーゼをコ ードする、DNA。 21.図16に示される配列の少なくとも一部、またはそれとの実質的な相同性 を有する配列、あるいはその改変体を含み、そしてシステインプロテアーゼをコ ードする、DNA。 22.図17に示される配列の少なくとも一部、またはそれとの実質的な相同性 を有する配列、あるいはその改変体を含み、そしてシステインプロテアーゼをコ ードする、DNA。 23.プロモーターに作動可能に連結された請求項17〜22のいずれかに記 載のDNAを含む植物において機能的な、組換えDNA構築物。 24.請求項5〜16および23のいずれかに記載の組換えDNA構築物であっ て、該構築物が、穀果または実生または植物のゲノムDNA内に統合され、好まし くは安定に統合される場合、発芽中の実生または発生中の穀果または植物の1つ または複数の組織で発現され得る、組換えDNA構築物。 25.植物材料の1つまたは複数の組織についての発現系であって、該発現系 が、請求項1〜4のいずれかに記載の遺伝子プロモーターに融合した目的の遺伝 子を含み、ここで、該発現系が、植物材料の1つまたは複数の組織で発現され得 る、発現系。 26.前記発現系が、発芽中の実生または発生中の穀果または植物(例えば、 根、子葉、葉、および幹)の少なくとも組織についての発現系である、請求項2 5に記載の発現系。 27.前記発現系が、発芽中の実生のゲノムDNAまたは発生中の穀果のゲノムD NAまたは植物のゲノムDNA内に統合され、好ましくは安定に統合される、請求項 25または26に記載の発現系。 28.請求項5〜16および23のいずれかに記載の構築物を含む、発現系。 29.請求項1〜4のいずれかに記載のプロモーター、請求項17〜22のい ずれかに記載のDNA、請求項5〜16または23のいずれかに記載の組換えDNA構 築物、または請求項24〜28のいずれかに記載の発現系を含む、組換え植物ゲ ノム。 30.請求項29に記載の組換え植物ゲノムを有する植物、植物の種子、また は植物細胞。 31.請求項5〜16または23のいずれかに記載の組換えDNA構築物を含む 植物を含む、防御された植物胚形質。 32.請求項5〜16または23のいずれかに記載の組換えDNA構築物を含む 成熟状態に生長し得ない、植物または種子。 33.植物材料の1つまたは複数の組織で遺伝子プロモーターに融合される場 合に目的の遺伝子の発現を誘導するための、請求項1〜4のいずれかに記載の遺 伝子プロモーターの使用。 34.前記遺伝子プロモーターが、発芽中の実生または発生中の穀果または植 物(例えば、根、子葉、葉、および幹)の少なくとも1つまたは複数の組織で、 該遺伝子プロモーターに融合される場合に目的の遺伝子の発現を誘導するために 使用される、請求項33に記載の使用。 35.図12〜17および図19〜21のいずれか1つに関して上記のように実質的に 記載される、プロモーター、構築物、または発現系。
