Claims (1)
- 【特許請求の範囲】 1.標的ポリペプチド配列上のオーファン結合部位を同定する方法であって、 (a)潜在的リガンドのライブラリーを提供する工程、 (b)該標的ポリペプチドの公知のリガンドと接触している該標的ポリペプチド を提供する工程、 (c)該標的ポリペプチドおよび公知のリガンドを該潜在的リガンドと接触させ る工程、および (d)該公知のリガンドの存在下で、該標的ポリペプチドに結合し、該標的ポリ ペプチドと結合対を形成する該潜在的リガンドを同定する工程、 を包含する、方法。 2.前記該潜在的リガンドのライブラリーが、バクテリオファージライブラリー 含む、請求項1に記載の方法。 3.前記標的ペプチド配列が、レセプター、リガンド、成長因子、ポリペプチド ホルモン、サイトカイン、分化因子、シグナル伝達可能な分子、酵素、細胞外マ トリックス相互作用に関与するポリペプチドからなる群より選択される配列を含 む、請求項1に記載の方法。 4.前記レセプターが、ウロキナーゼプラスミノーゲンアクチベーターレセプタ ー、ならびにビトロネクチンおよびuPAからなる群より選択される1つを含む公 知のリガンドである、請求項3に記載の方法。 5.前記潜在的リガンドが、ランダム合成ペプチド、低分子、ペプトイド、ポリ ペプチド、およびポリヌクレオチドからなる群より選択される1つを含む、請求 項1に記載の方法。 6.前記潜在的リガンドを、前記標的ポリペプチドおよび未知のリガンドと接触 させ、該標的ポリペプチドと未知のリガンドとの間の結合対相互作用を拮抗する 潜在的リガンドを同定する、請求項1に記載の方法。 7.前記未知のリガンドがインテグリンである、請求項6に記載の方法。 8.ウロキナーゼプラスミノーゲンアクチベーターレセプター(uPAR)と結合し、 そしてuPARのインテグリンとの結合を阻害する、単離されたペプチド。 9.AESTYHHLSLGYMYTLN(配列番号4)、YHXLXXGYMYT(配列番号5)(Xは任意の アミノ酸)からなる群より選択されるアミノ酸配列を含む、請求項8に記載の単 離されたペプチド。 10.ウロキナーゼプラスミノーゲンアクチベーターレセプター(uPAR)と結合し 、そしてuPARのビトロネクチンとの結合を阻害する、単離されたペプチド。 11.AEPVYQYELDSYLRSYY(配列番号1)、およびAELDLSTFYDIQYLLRT(配列番号 3)からなる群より選択されるアミノ酸配列を含む、請求項10に記載の単離さ れたペプチド。 12.AEFFKLGPNGYVYLHSA(配列番号2)、およびFKLXXXGYVYL(配列番号6)( Xは任意のアミノ酸)からなる群より選択されるアミノ酸配列を含む、ウロキナ ーゼプラスミノーゲンアクチベーターレセプター(uPAR)と結合する単離されたペ プチド。 13.ウロキナーゼプラスミノーゲンアクチベーターレセプター(uPAR)と結合し 、そしてuPARのインテグリンとの結合を阻害するペプチドをコードする配列を含 む、単離されたポリヌクレオチド。 14.前記配列が、AESTYHHLSLGYMYTLN(配列番号4)、およびYHXLXXGYMYT(配 列番号5)(Xは任意のアミノ酸)からなる群より選択されるアミノ酸配列を含む ペプチドをコードする、請求項13に記載の単離されたポリヌクレオチド。 15.ウロキナーゼプラスミノーゲンアクチベーターレセプター(uPAR)と結合し 、そしてuPARのビトロネクチンとの結合を阻害するペプチドをコードする配列を 含む、単離されたポリヌクレオチド。 16.前記配列が、AEPVYQYELDSYLRSYY(配列番号1)、およびAELDLSTFYDIQYLL RT(配列番号3)からなる群より選択されるアミノ酸配列を含むペプチドをコー ドする、請求項15に記載の単離されたポリヌクレオチド。 17.ウロキナーゼプラスミノーゲンアクチベーターレセプター(uPAR)と結合す るペプチドをコードする配列を含む、単離されたポリヌクレオチドであって、該 ペプチドが、AEFFKLGPNGYVYLHSA(配列番号2)、およびFKLXXXGYVYL(配列番号 6)(Xは任意のアミノ酸)からなる群より選択されるアミノ酸配列を含む、単 離されたポリヌクレオチド。 18.uPAおよびuPARのアップレギュレーションによって特徴づけられる障害を 有する患者を処置する方法であって、 (a)uPAR:インテグリン結合対の有効量のアンタゴニストを提供する工程、 (b)該アンタゴニストを該患者に投与する工程 を包含する、方法。 19.前記アンタゴニストが、AESTYHHLSLGYMYTLN(配列番号4)およびYHXLXXGY MYT(配列番号5)(Xは任意のアミノ酸)からなる群より選択されるアミノ酸配 列を含むペプチドである、請求項18に記載の方法。 20.前記アンタゴニストが、AESTYHHLSLGYMYTLN(配列番号4)およびYHXLXXGY MYT(配列番号5)(Xは任意のアミノ酸)からなる群より選択されるアミノ酸配 列を含むペプチドをコードするポリヌクレオチドを含む、請求項18に記載の方 法。 21.前記障害が、細胞移動をさらに含むuPAおよびuPARのアップレギュレーシ ョンによって特徴づけられる、請求項18に記載の方法。 22.前記障害が、ガンおよび慢性の炎症からなる群より選択される1つをさら に含む、請求項18に記載の患者を処置する方法。 23.uPAR:インテグリン相互作用のアンタゴニストをスクリーニングする方法 であって、 (a)uPAR:インテグリン相互作用のペプチドアンタゴニストを提供する工程; (b)uPARへの結合に対する候補アンタゴニストと該ペプチドアンタゴニストを 競合させる工程; (c)uPAR結合に対する該ペプチドアンタゴニストとの競合能により候補アンタ ゴニストを同定する工程; を包含する、方法。 24.前記ペプチドアンタゴニストが、AESTYHHLSLGYMYTLN(配列番号4)および YHXLXXGYMYT(配列番号5)(Xは任意のアミノ酸)からなる群より選択されるア ミノ酸配列を含む、請求項23に記載の方法。 25.前記候補アンタゴニストが、低分子、ペプチド、およびペプトイドからな る群より選択される、請求項24に記載の方法。 26.uPAR:ビトロネクチン相互作用のアンタゴニストをスクリーニングする方 法であって、 (a)uPAR:ビトロネクチン相互作用のペプチドアンタゴニストを提供する工程; (b)uPARへの結合に対する候補アンタゴニストと該ペプチドアンタゴニストを 競合させる工程; (c)uPAR結合に対する該ペプチドアンタゴニストとの競合能により候補アンタ ゴニストを同定する工程; を包含する、方法。 27.前記ペプチドアンタゴニストが、AEPVYQYELDSYLRSYY(配列番号1)、お よびAELDLSTFYDIQYLLRT(配列番号3)からなる群より選択されるアミノ酸配列 を含む、請求項26に記載の方法。 28.ウロキナーゼプラスミノーゲンアクチベーターレセプター(uPAR)に結合 する候補をスクリーニングする方法であって、 (a)AEFFKLGPNGYVYLHSA(配列番号2)、およびFKLXXXGYVYL(配列番号6)(X は任意のアミノ酸)からなる群より選択されるアミノ酸配列を含むペプチドを提 供する工程; (b)uPARへの結合に対する候補と該ペプチドを競合させる工程; (c)uPAR結合に対する該ペプチドとの競合能により候補を同定する工程; を包含する、方法。 29.uPAR:インテグリン結合対の有効量のアンタゴニストおよび薬学的に受容 可能なキャリアを含む、uPAおよびuPARのアップレギュレーションによって特徴 づけられる障害を処置するための薬学的組成物。 30.前記アンタゴニストが、AESTYHHLSLGYMYTLN(配列番号4)およびYHXLXXGY MYT(配列番号5)(Xは任意のアミノ酸)からなる群より選択されるアミノ酸配 列を含むペプチドを含む、請求項29に記載の薬学的組成物。 31.前記障害が、細胞移動をさらに含むuPAおよびuPARのアップレギュレーシ ョンによって特徴づけられる、請求項29に記載の薬学的組成物。 32.AEPVYQYELDSYLRSYY(配列番号1)、AEFFKLGPNGYVYLHSA(配列番号2)、 AELDLSTFYDIQYLLRT(配列番号3)、AESTYHHLSLGYMYTLN(配列番号4)、YHXLXXG YMYT(配列番号5)およびFKLXXXGYVYL(配列番号6)(Xは任意のアミノ酸)から なる群より選択されるアミノ酸配列を含むペプチドをコードする有効量の核酸を 含む、uPAおよびuPARのアップレギュレーションによって特徴づけられる障害を 有する患者を処置するための薬学的組成物。 33.前記薬学的受容可能なキャリアが、リポソーム、ゲル、ポリマーマトリッ クス、泡、および緩衝液からなる群より選択される1つを含む、請求項32に記 載の薬学的組成物。
