Claims (1)
- 【特許請求の範囲】 1. ハウジングと、 第1及び第2の側がある組織とを、備え、 前記ハウジングは、前記ハウジングと前記組織の第1の側の間の第1流空間を 画定し、前記第1流空間は第1培地流を内部に配置し、第1濃度の溶質を有し、 前記組織の第1の側に接触し、 前記ハウジングは、前記ハウジングと前記組織の第2の側の間の第2流空間を 画定し、前記第2流空間は第2培地流を内部に配置し、第2濃度の溶質を有し、 前記組織の第2の側に接触し、溶質の第1濃度は溶質の第2濃度とは異なってお り、前記溶質は拡散により前記組織を通過することを特徴とするバイオリアクタ 。 2. 第1半透膜が前記組織と前記第1培地流の間に配置され、第2半透膜が前 記組織と前記第2培地流の間に配置され、前記第1および第2半透膜を選択され た溶質が通過できる請求項1に記載のバイオリアクタ。 3. 前記組織は基質細胞を有する請求項1に記載のバイオリアクタ。 4. 前記組織はさらに肝臓実質細胞を有する請求項2に記載のバイオリアクタ 。 5. 円筒形ハウジングと、 前記ハウジングの内周面上に配置された組織と、 前記組織と前記ハウジングの間の第1空間と、 前記組織が結合した前記ハウジング内の第2中央空間と、 第1濃度の溶質の第1培地流を前記第1空間に供給する少なくとも1つの培地 入口と、 第2濃度の溶質の第2培地流を前記第2空間に供給する少なくとも1つの培地 入口と、 少なくとも1つの培地出口と、を備えることを特徴とするバイオリアクタ。 6. 溶質の第1濃度は溶質の第2濃度とは異なっており、前記溶質は拡散勾配 により前記組織を通過する請求項5に記載のバイオリアクタ。 7. 内径と外径をもつ円筒形ハウジングと、 前記円筒形ハウジングの内径にほぼ等しい直径の第1円筒体部分と、前記第1 直径より小さい第2直径の第2円筒体部分と、前記第2直径より小さい第3直径 の第3円筒体部分をそれぞれ有し、各端キャップの前記第1円筒体部分は前記円 筒形ハウジングの端部に密封係合する2つの端キャップと、 内周と外周をそなえ、前記円筒形ハウジング内に同軸状に配置され、1端部が 1方の端キャップの前記第3体部分上に周面装着され、他端部が他の端キャップ の前記第3体部分上に周面装着される円筒形半透膜と、 前記円筒形ハウジングと前記半透膜の間に配置され、1端が1方の端キャップ の前記第2体部分上に周面装着され、他端が他方の端キャップの前記第2体部分 上に周面装着された円筒形支柱と、 前記膜および前記支柱の間の第1空間と、 前記膜の内周面上に配置された組織と、 前記組織が結合した第2中央空間と、 第1濃度の溶質を有する第1培地流を第1空間に供給する少なくとも1つの培 地入口と、 第2濃度の溶質を有する第2培地流を第2空間に供給する少なくとも1つの培 地入口と、 前記第1培地流の溶質が前記半透膜を介して通過可能な少なくとも1つの培地 出口と、を備えることを特徴とするバイオリアクタ。 8. 溶質の第1濃度は溶質の第2濃度とは異なっており、前記溶質は拡散によ り前記組織を通過する請求項7に記載のバイオリアクタ。 9. 前記組織は前記半透膜の外周面上に配置され、前記第2培地流の前記溶質 が前記膜を通過可能な請求項7に記載のバイオリアクタ。 10. 3次元組織を増殖させる方法において、 (a)内部にメッシュ部分を配置して前記メッシュ部分の対向両側の個々の流 空間が画定されるハウジングを備えるバイオリアクタを用意する工程と、 (b)前記メッシュ部分上に細胞を接種する工程と、 (c)第1濃度の溶質を有する第1培地を前記メッシュ部分の1方の側上の細 胞に接触して流す工程と、 (d)第2濃度の溶質を有する第2培地を前記メッシュ部分の他方の側の上の 細胞に接触して流し、ここで溶質の第1濃度は溶質の第2濃度とは異なっており 、前記溶質は拡散により前記組織を通過するものである工程と、 (e)多機能細胞を有する3次元組織を増殖させ、多機能細胞の発達は、拡散 溶質にさらすことで前記培地流に関して極性を有することになる工程と、を備え ることを特徴とする前記方法。 11. 細胞を接種する前記工程は基質細胞を接種するものである請求項10に 記載の方法。 12. 前記基質細胞を形成した後で肝実質細胞を追加する工程をさらに備える 請求項11に記載の方法。 13. 前記3次元組織は肝臓組織であり、 (a)前記培地流を停止させる工程と、 (b)前記3次元肝臓細胞の1方の側と接触して血液を流す工程と、 (c)前記3次元肝臓組織の反対側と接触して培地を流し、拡散により前記肝 臓組織を介して血液廃棄物を通過させる工程と、をさらに備える請求項10に記 載の方法。 14. (a)前記血液を血漿と血球に分離して、前記肝臓組織の1方の側に前 記血漿を流す工程と、 (b)前記血液を前記患者に戻す前に前記血漿と血球を再合体する工程と、を さらに含む請求項15に記載の方法。
