Claims (1)
- 【特許請求の範囲】 1. キシロースリダクターゼ、キシリトールデヒドロゲナーゼ及びキシルロキ ナーゼをコードする遺伝子が、酵母の複数反復リボゾームDNA部位の各々に組 込まれた酵母を含む、キシロースをエタノールに発酵する酵母。 2. さらにグルコースのエタノールへの発酵も行なう請求項1記載の酵母。 3. Saccharomycesである請求項2記載の酵母。 4. 前記部位が非転写DNA部位である請求項3記載の酵母。 5. 前記遺伝子が、非グルコース抑制プロモーターに融合されており、酵母が グルコースとキシロースのエタノールへの発酵を同時に行なう請求項1記載の酵 母。 6. 前記プロモーターが誘導にキシロースを要求しない請求項5記載の酵母。 7. 前記遺伝子が、非グルコース抑制プロモーターに融合されており、酵母が グルコースとキシロースのエタノールへの発酵を同時に行なう請求項3記載の酵 母。 8. 前記遺伝子が、非グルコース抑制プロモーターに融合されており、酵母が グルコースとキシロースのエタノールへの発酵を同時に行ない、該プロモーター が誘導にキシロースを要求しない請求項4記載の酵母。 9.前記キシロースリダクターゼ及びキシリトールデヒドロゲナーゼ遺伝子が、 キシロースをエタノールに発酵する天然酵母由来である請求項6記載の酵母。 10. 前記天然酵母が、Candida Shehatae,Pichia stipitisまたはPachysolen tannophilusである請求項9の酵母。 11. 前記キシルロキナーゼ遺伝子が酵母または細菌由来である請求項9記載 の酵母。 12. 前記キシルロキナーゼ遺伝子が、Candida Shehatae,Pichiastipitis、P achysolen tannophilus、Saccharomyces cerevisiae、Schizosaccaromyces pomb eまたはEscherichia coli由来である請求項11記載の酵母。 13. 酵母の少なくとも約10個のリボソームDNA部位に組込まれた前記遺 伝子を有する請求項1記載の酵母。 14. (a)第1の選択マーカーを含む外来DNAを有する複製及び組込みプ ラスミドで細胞を形質転換し、そして (b)工程(a)で得られた細胞を繰り返し複製し、選択マーカー含む細胞を選 択しながら複数の世代の子孫細胞を生じさせ、その結果、子孫細胞の次の世代に おける複製及び組込プラスミドの保持を促進し、外来DNAの複数の挿入コピー を有する子孫細胞を生じさせることを含む方法。 15. 前記プラスミドDNAが、プラスミドを含む細胞を選択するための第二 の選択マーカーをも含む請求項14記載の方法。 16. 細胞が酵母または真核細胞であり、さらに、工程(b)で得られた子孫 細胞を、選択マーカーを含む細胞の選択をせずに繰り返し複製して、複数の世代 の子孫細胞を生じさせ、その結果次の世代の子孫細胞におけるプラスミドの喪失 を促進し、外来DNAの複数のコピーが各々染色体DNAに組込まれた酵母細胞 を回収することを含む請求項14記載の方法。 17. 前記細胞が酵母細胞であり、前記外来のDNAが、第一の選択マーカー としても機能するキシロースリダクターゼ、キシリトールデヒドロゲナーゼ、及 びキシルロキナーゼをコードする遺伝子を含む請求項16記載の方法。 18. (i)酵母細胞を選択マーカーを含む外来DNAを有する複製プラスミ ドで形質転換し、該外来DNAが、宿主のDNAの反復配列に相同なDNA配列 により各末端にフランキングしており、 (ii)工程(i)で得られた形質転換された酵母細胞を繰り返し複製し、選択マ ーカーを含む細胞を選択しながら複数の世代の子孫細胞を生じさせ、その結果、 子孫細胞の次の世代における複製プラスミドの保持を促進し、外来DNAの複数 の挿入コピーを有する子孫細胞を生じさせ、そして (iii)工程(ii)からの子孫細胞を、選択マーカーを含む細胞の選択をせずに 複製して、複数の世代の子孫細胞を製造し、その結果、次の世代の子孫細胞にお けるプラスミドの喪失を促進し、外来DNAの複数のコピーが各々染色体DNA に組込まれた酵母細胞を回収することを含む請求項14記載の方法。 19. 請求項18記載の方法により生産される酵母細胞。 20. 前記外来DNAが、キシロースリダクターゼ、キシリトールデヒドロゲ ナーゼ、及びキシルロキナーゼをコードする遺伝子を含み、前記酵母細胞が、キ シロースをエタノールに発酵する請求項19記載の酵母細胞。 21. 前記遺伝子が、誘導にキシロースを要求しない非グルコース抑制プロモ ーターに融合しており、前記酵母細胞が、グルコースとキシロースを同時にエタ ノールに発酵する請求項20記載の酵母細胞。 22. 非選択性条件で培養した場合に、キシロースをエタノールに発酵する能 力を、少なくとも20世代の間実質的に維持する請求項21記載の酵母細胞。 23. キシロースをエタノールに発酵する酵母であって、 該酵母の染色体DNAに組み込まれた外来DNAの複数のコピーを有し、前記 外来DNAが、非グルコース抑制プロモーターに融合したキシロースリダクター ゼ、キシリトールデヒドロゲナーゼ、及びキシルロキナーゼをコードする遺伝子 を含み、前記酵母が、グルコースとキシロースを同時にエタノールに発酵し、そ して非選択性条件で培養した場合に、キシロースをエタノールに発酵する能力を 、少なくとも20世代の間実質的に維持する酵母を含む酵母。 24. 前記プロモーターが誘導にキシロースを要求しない請求項23記載の酵 母。 25. キシロースをエタノールに発酵する酵母であって、 キシロースリダクターゼ、キシリトールデヒドロゲナーゼ、及びキシルロキナ ーゼをコードする導入されたDNAの複数のコピーを含み、前記酵母がキシロー スをエタノールに発酵し、そして非選択性条件で培養した場合に、キシロースを エタノールに発酵する能力を、少なくとも20世代の間実質的に維持する酵母を 含む酵母。 26. 前記プロモーターが誘導にキシロースを要求しない請求項25記載の酵 母。 27. 請求項1、22、23、24、25または26記載の酵母を用いて、キ シロース含有培地を発酵してエタノールを生産することを含む、キシロースのエ タノールへの発酵方法。 28. 第一の選択マーカーを含む外来DNA配列を標的酵母細胞の染色体DN Aに組込むためのプラスミドベクターであって、機能的な酵母DNA複製起点と 、標的酵母細胞のリボソームDNAの反復配列に相同なDNAフランキング配列 に より各末端にフランキングする外来DNAを含み、該プラスミドは、さらに第二 の選択マーカーをDNAフランキング配列の問以外の位置に有するプラスミドベ クター。 29. 外来DNA配列を酵母に組み込んで、キシロースをエタノールに発酵す る安定な組込み体を形成するためのプラスミドベクターであって、機能的な酵母 DNA複製起点と、標的酵母細胞の反復DNA配列に相同なDNAフランキング 配列により各末端にフランキングするキシロースリダクターゼ、キシリトールデ ヒドロゲナーゼ、及びキシルロキナーゼをコードする遺伝子含む外来DNAを含 むプラスミドベクター。 30. 外来DNA断片のコピーが複数組み込まれた細胞を形成するための方法 であって、反復ゲノムDNAを有し、外来DNAを含む複製及び組込プラスミド を含む細胞を複製し、選択マーカーを含む細胞を選択しながら複数の世代の子孫 細胞を生じさせ、その結果、子孫細胞の次の世代における複製及び組込みプラス ミドの保持を促進し、外来DNAの複数の組込コピーを有する子孫細胞を生じさ せることを含む方法。
