Claims (1)
- 【特許請求の範囲】 1.合成中間体から保護基を除去する方法であって、該方法は、以下の工程を 包含する: a)以下を含有する表面を形成する工程: i)感光性化合物または基であって、該感光性化合物または基は、照射すると 、触媒を生成する、および ii)自己触媒化合物または基であって、該自己触媒化合物または基は、該触 媒により活性化すると、保護基除去生成物を生じる;および b)該表面の少なくとも一部を照射して、該保護基を除去する工程。 2.前記感光性化合物が、PACである、請求項1に記載の方法。 3.前記自己触媒化合物が、遮蔽された酸である、請求項1に記載の方法。 4.前記自己触媒化合物が、ペンタフルオロ安息香酸である、請求項1に記載 の方法。 5.前記合成中間体が、リンカー分子である、請求項1に記載の方法。 6.前記合成中間体が、DMT保護ヌクレオチドである、請求項1に記載の方法 。 7.前記合成中間体が、ヌクレオチドである、請求項1に記載の方法。 8.前記合成中間体が、ポリヌクレオチドである、請求項1に記載の方法。 9.前記合成中間体が、アミノ酸である、請求項1に記載の方法。 10.前記合成中間体が、ポリペプチドである、請求項1に記載の方法。 11.前記除去可能保護基が、酸除去可能基である、請求項1に記載の方法。 12.前記感光性化合物が、トルエンスルホン酸である、請求項1に記載の方法 。 13.前記自己触媒化合物が、ペンタフルオロベノン酸(pentafluorobenoic aci d)である、請求項1に記載の方法。 14.前記除去可能保護基が、5'-ジメトキシトリチルである、請求項1に記載 の方法。 15.前記感光性基および前記自己触媒基が、同一化合物の一部である、請求項 1に記載の方法。 16.前記感光性基が、PACである、請求項1に記載の方法。 17.多様な配列の重合体を合成する方法であって、該方法は、以下の工程を包 含する: a)以下を含有する表面を形成する工程: i)感光性化合物または基であって、該感光性化合物または基は、照射すると 、触媒を生成する、および ii)自己触媒化合物または基であって、該自己触媒化合物または基は、該触 媒により活性化すると、保護基除去生成物を生じる; b)該表面の少なくとも一部を照射して、該保護基を除去する工程; c)該表面を第一モノマーと接触させる工程; d)該表面の少なくとも他の部分を照射して、該保護基を除去する工程;および e)該表面を第二モノマーと接触させる工程。 18.前記感光性化合物が、PACである、請求項17に記載の方法。 19.前記自己触媒化合物が、遮蔽された酸である、請求項17に記載の方法。 20.前記合成中間体が、リンカー分子である、請求項17に記載の方法。 21.前記合成中間体が、ヌクレオチドである、請求項17に記載の方法。 22.前記合成中間体が、ポリヌクレオチドである、請求項17に記載の方法。 23.前記合成中間体が、アミノ酸である、請求項17に記載の方法。 24.前記合成中間体が、ポリペプチドである、請求項17に記載の方法。 25.前記除去可能保護基が、酸除去可能基である、請求項17に記載の方法。 26.前記感光性化合物が、トルエンスルホン酸である、請求項17に記載の方法 。 27.前記自己触媒化合物が、ペンタフルオロベノン酸(pentafluorobenoic aci d)である、請求項17に記載の方法。 28.前記酸除去可能保護基が、5'-ジメトキシトリチル(5'-demethoxytrityl )である、請求項17に記載の方法。 29.固相化学合成のための装置であって、該装置は、以下を含有する基板を包 含する: i)除去可能保護基を含有する合成中間体; ii)感光性化合物または基であって、該感光性化合物または基は、照射すると 、触媒を生成する、および iii)自己触媒化合物または基であって、該自己触媒化合物または基は、該触媒 により活性化すると、保護基除去生成物を生じる。 30.前記感光性化合物が、PACである、請求項29に記載の装置。 31.前記自己触媒化合物が、遮蔽された酸である、請求項29に記載の装置。 32.前記合成中間体が、リンカー分子である、請求項29に記載の装置。 33.前記合成中間体が、ヌクレオチドである、請求項29に記載の装置。 34.前記合成中間体が、ポリヌクレオチドである、請求項29に記載の装置。 35.前記合成中間体が、アミノ酸である、請求項29に記載の装置。 36.前記合成中間体が、ポリペプチドである、請求項29に記載の装置。 37.前記除去可能保護基が、酸除去可能基である、請求項29に記載の装置。 38.前記PACが、トルエンスルホン酸である、請求項29に記載の装置。 39.前記自己触媒化合物が、ペンタフルオロベノン酸(pentafluorobenoic aci d)である、請求項29に記載の装置。 40.前記酸除去可能保護基が、5'-ジメトキシトリチルである、請求項29に記 載の装置。 41.核酸をハイブリダイズする方法であって、該方法は、以下の工程を包含す る: a)以下を含有する表面を形成する工程: i)感光性化合物または基であって、該感光性化合物または基は、照射すると 、触媒を生成する、および ii)自己触媒化合物または基であって、該自己触媒化合物は、該触媒により 活性化すると、保護基除去生成物を生じる; b)該表面の少なくとも一部を照射して、該保護基を除去する工程; c)該表面を第一モノマーと接触させる工程; d)該表面の少なくとも他の部分を照射して、該保護基を除去する工程; e)該表面を第二モノマーと接触させる工程; f)工程a)〜e)を繰り返して、所望長のポリヌクレオチドアレイを合成する工程 ;および g)標的核酸を該ポリヌクレオチドアレイとハイブリダイズする工程。 42.前記合成中間体が、リンカー分子である、請求項41に記載の方法。 43.前記合成中間体が、ヌクレオチドである、請求項41に記載の方法。 44.前記合成中間体が、ポリヌクレオチドである、請求項41に記載の方法。 45.前記除去可能保護基が、酸除去可能基である、請求項41に記載の方法。 46.前記感光性化合物が、トルエンスルホン酸である、請求項41に記載の方法 。 47.前記自己触媒化合物が、ペンタフルオロ安息香酸である、請求項41に記載 の方法。 48.前記酸除去可能保護基が、5'-ジメトキシトリチルである、請求項41に記 載の方法。 49.基板上で重合体アレイを合成する方法であって、該方法は、以下の工程を 包含する: a)その上に1個またはそれ以上の合成中間体を結合した該基板の表面、および 触媒系を提供する工程であって、該合成中間体は、保護基により反応から保護さ れた反応性基を有する; b)該基板の該表面を、一定波長の放射線で照射する工程; c)該合成中間体から該保護基を除去する触媒反応を開始して、未保護反応性基 を生成する工程; d)引き続いて追加した合成中間体を該未保護反応性基と反応させる工程であっ て、該引き続いて追加した合成中間体は、保護基により反応から保護した反応性 基を有する; e)所望の重合体配列が得られるまで、必要なら、工程b)〜d)を繰り返す工程。 50.重合体アレイを製造する際に使用する基板であって、該基板は、以下を含 有する: a)その上に1個またはそれ以上の合成中間体を結合した基板表面であって、該 合成中間体は、保護基により反応から保護した反応性基を有する; b)該合成中間体と接触する触媒系であって、該触媒系は、該保護基の除去を触 媒できる。 51.重合体アレイを製造する際に使用する基板を製造する方法であって、該方 法は、以下を包含する: a)その上に1個またはそれ以上の合成中間体を結合した基板表面を提供する工 程であって、該合成中間体は、保護基により反応から保護した反応性基を有する ; b)該基板の表面に、該保護基の除去を触媒できる触媒系を有する重合体層を付 与する工程。
