- 【特許請求の範囲】 1.実質的に精製されたJaggedタンパク質。 2.通常会合されているタンパク質を含まず、そして配列番号1に対応するアミ ノ酸配列を有する、請求項1に記載のタンパク質。 3.前記タンパク質が配列番号1に対応するアミノ酸配列、その機能的に等価な 誘導体、または対立遺伝子改変体もしくは種改変体を含む、請求項2に記載のタ ンパク質。 4.前記タンパク質が配列番号1に対応するアミノ酸配列、その機能的に等価な 誘導体、または対立遺伝子改変体もしくは種改変体を含み、そしてNotchに結合 する能力によって特徴づけられる、請求項3に記載のタンパク質。 5.Jaggedタンパク質、あるいはその機能的に等価な誘導体、または対立遺伝子 改変体もしくは種改変体をコードする実質的に精製された核酸分子またはそのDN Aセグメント。 6.前記核酸が、配列番号1に対応する配列を有する配列、またはそのセグメン トを含む、請求項5に記載の核酸分子。 7.前記核酸が、配列番号1に対応する核酸配列またはそのセグメント、あるい はその機能的に等価な誘導体、または対立遺伝子改変体もしくは種改変体を含む 、請求項6に記載の核酸分子。 8.前記核酸が、配列番号1に対応する核酸配列またはそのセグメント、あるい はその機能的に等価な誘導体、または対立遺伝子改変体もしくは種改変体を含み 、そしてNotchに結合する能力によって特徴づけられる、請求項6に記載の核酸 分 子。 9.ベクターおよび請求項5に記載の核酸配列を含む組換え分子。 10.請求項9に記載の組換え分子を含む宿主細胞。 11.請求項9に記載の組換え分子の発現産物。 12.Jagged cDNAのアンチセンス鎖またはそのセグメントを含む、実質的に精 製された一本鎖核酸分子。 13.前記核酸配列が、配列番号1に対応するアンチセンスヌクレオチド配列、 またはそのセグメントを含む、請求項12に記載の核酸分子。 14.前記アンチセンス核酸が、配列番号1に対応するアンチセンス核酸配列、 あるいはセンス方向に読まれれば、その機能的に等価な誘導体、または対立遺伝 子改変体もしくは種改変体をコードするDNAセグメントを含む、請求項13に記 載の核酸分子。 15.前記アンチセンス核酸が、配列番号1に対応するアンチンセンス核酸配列 、あるいはセンス方向に読まれれば、その機能的に等価な誘導体、または対立遺 伝子改変体もしくは種改変体をコードするDNAセグメントを含み、そしてJagged に結合する能力によって特徴づけられる、請求項14に記載の核酸分子。 16.請求項12〜15に記載の核酸分子によりコードされたポリペプチド。 17.前記ポリペプチドが、Jaggedへの結合親和性を有し、そしてJaggedの活性 を阻害する、請求項12〜15に記載の核酸分子によりコードされたポリペプチ ド。 18.Jaggedまたはその独特部分への結合親和性を有する抗体。 19.抗Jaggedまたはその独特部分への結合親和性を有する二次抗体。 20.微小直径血管上の部位への内皮細胞の移動を減少させる方法であって、請 求項1または19に記載のタンパク質を、内皮細胞が取り除かれているか、損傷 を受けているか、または実質的に減少している該部位に送達する工程を包含する 、方法。 21.巨大直径血管上の部位への内皮細胞の移動を増大させる方法であって、請 求項1または19に記載のタンパク質を、内皮細胞が取り除かれているか、損傷 を受けているか、または実質的に減少している該部位に送達する工程を包含する 、方法。 22.微小直径血管上の部位への内皮細胞の移動を増大させる方法であって、請 求項16、17または18に記載のタンパク質を、内皮細胞が取り除かれている か、損傷を受けているか、または実質的に減少している該部位に送達する工程を 包含する、方法。 23.巨大直径血管上の部位への内皮細胞の移動を減少させる方法であって、請 求項16、17または18に記載のタンパク質を、内皮細胞が取り除かれている か、損傷を受けているか、または実質的に減少している該部位に送達する工程を 包含する、方法。 24.巨大直径血管上の部位への平滑筋細胞の移動を減少させる方法であって、 請求項16、17または18に記載のタンパク質を、内皮細胞が取り除かれてい るか、損傷を受けているか、または実質的に減少している部位に送達する工程を 包含する、方法。 25.治療有効量のJaggedタンパク質、あるいはその機能的に等価な誘導体、ま たは対立遺伝子改変体もしくは種改変体;および薬学的に受容可能なキャリアを 含む薬学的組成物。 26.治療有効量のJagged核酸、あるいはその機能的に等価な誘導体、または対 立遺伝子改変体もしくは種改変体;および薬学的に受容可能なキャリアを含む薬 学的組成物。 27.治療有効量のJagged抗体、あるいはその機能的に等価な誘導体、または対 立遺伝子改変体もしくは種改変体;および薬学的に受容可能なキャリアを含む薬 学的組成物。 28.治療有効量のJaggedアンチセンス分子、あるいはその機能的に等価な誘導 体、または対立遺伝子改変体もしくは種改変体;および薬学的に受容可能なキャ リアを含む薬学的組成物。 29.治療有効量の抗Jagged抗体、あるいはその機能的に等価な誘導体、または 対立遺伝子改変体もしくは種改変体;および薬学的に受容可能なキャリアを含む 薬学的組成物。 30.被験体において疾患または状態を予防または処置する方法であって、この ような予防または処置が必要な被験体に、治療有効量の、Notchタンパク質の機 能を拮抗するか、阻害するか、または予防する分子を投与する工程を包含する、 方法。 31.被験体において疾患または状態を予防または処置する方法であって、この ような予防または処置が必要な被験体に、治療有効量の、Notchタンパク質の機 能を作動するか、増強するか、または刺激する分子を投与する工程を包含する、 方法。 32.被験体において疾患または状態を予防または処置する方法であって、この ような予防または処置が必要な被験体に、治療有効量の、Jaggedタンパク質の機 能を拮抗するか、阻害するか、または予防する分子を投与する工程を包含する、 方法。 33.被験体において疾患または状態を予防または処置する方法であって、この ような予防または処置が必要な被験体に、治療有効量の、Notchタンパク質の機 能を作動するか、増強するか、または刺激する分子を投与する工程を包含する、 方法。 34.被験体において新脈管形成を阻害または予防する方法であって、このよう な阻害または予防が必要な被験体に、治療有効量のJaggedまたはJaggedアゴニス トを投与する工程を包含する、方法。 35.前記阻害または予防される新脈管形成が、固形腫瘍新脈管形成を含む、請 求項34に記載の方法。 36.前記阻害または予防される新脈管形成が、慢性関節リウマチ新脈管形成を 含む、請求項34に記載の方法。 37.前記阻害または予防される新脈管形成が、炎症性新脈管形成を含む、請求 項34に記載の方法。 38.前記阻害または予防される新脈管形成が、血管の管腔の再狭窄を含む、請 求項34に記載の方法。 39.前記再狭窄が、脈管の脈管からの新脈管形成を抑制すること、および大血 管の管腔を修復するように大血管内皮細胞細胞移動を促進することによって阻害 または予防される、請求項38に記載の方法。 40.前記Jaggedアゴニストが、フィブリンおよびその機能的誘導体、および薬 理学的に受容可能な化学物質を包含する、Jaggedの発現を促進する薬剤、および γ-イディオタイプJagged抗体を含む、請求項34に記載の方法。 41.前記阻害または予防が、インビボおよび/またはインビトロで生じる、請 求項34に記載の方法。 42.被験体において新脈管形成を促進または増強する方法であって、このよう な促進または増強が必要な被験体に、治療有効量の抗JaggedまたはJaggedアンタ ゴニストを投与する工程を包含する、方法。 43.前記促進または増強される新脈管形成が、創傷または傷害修復の新脈管形 成を含む、請求項42に記載の方法。 44.前記修復される創傷または傷害が、手術、外傷および/または疾患もしく は状態において引き起こされた、請求項43に記載の方法。 45.前記疾患または状態が、糖尿病関連である、請求項44に記載の方法。 46.前記Jaggedアンタゴニストが、Jagged抗体、アンチセンスJagged、Jagged 変異体および薬理学的に受容可能な化学物質を含む、請求項42に記載の方法。 47.前記促進または増強が、インビボおよび/またはインビトロで生じる、請 求項42に記載の方法。 48.中胚葉、内胚葉、外胚葉および/または神経胚葉を含む細胞の細胞分化に 影響を及ぼす方法。 49.前記影響される細胞型が、造血幹細胞、上皮細胞、血管平滑筋細胞および 樹状細胞を含む、請求項48に記載の方法。 50.請求項30〜48のいずれかに記載の方法において用いられる薬学的組成 物。
