Claims (1)
- 【特許請求の範囲】 1. ヒトRHAMM1,ヒトRHAMM2,ヒトRHAMM3,ヒトRHAMM 4およびヒトRHAMM5からなる群から選ばれるタンパク質をコードするヌク レオチド配列を含んでなる単離された核酸。 2. 配列番号4のアミノ酸配列をコードする請求項1に記載の単離された核酸。 3. 該ヌクレオチド配列が (イ)ヒトRHAMMのゲノム配列と、 (ロ)配列番号3のヌクレオチド配列と からなる群から選ばれる請求項2に記載の単離された核酸。 4.(イ)連続した配列として配列番号9〜25のヌクレオチド配列を含むヌクレオ チド配列と、 (ロ)連続した配列として配列番号9、10、11および13のヌクレオチド配列 を含むヌクレオチド配列と、 (ハ)連続した配列として配列番号9、10および12〜25のヌクレオチド配列 を含むヌクレオチド配列と からなる群から選ばれる請求項1に記載の単離された核酸。 5.(イ)配列番号3からの少なくとも10個の連続したヌクレオチドのヌクレオチ ド配列と、 (ロ)配列番号3からの少なくとも15個の速続したヌクレオチドのヌクレオ チド配列と、 (ロ)配列番号3からの少なくとも20個の速続したヌクレオチドのヌクレオ チド配列と からなる群から選ばれるヌクレオチド配列を含んでなる単離された核酸。 6. HA結合親和性を保持している、ヒトRHAMMタンパク質の結合ドメイン 少なくとも1個をコードするヌクレオチドまたはそのフラグメントまたは類似体 を含んで なる単離された核酸。 7.配列番号1または配列番号7のアミノ酸配列をコードする請求項6に記載の 単離された核酸。 8.配列番号9、配列番号10、配列番号11、配列番号12、配列番号13、配列番号15 、配列番号16、配列番号17、配列番号18、配列番号19、配列番号20、配列番号21 、配列番号22、配列番号23、配列番号24、および配列番号25からなる群から選ば れるヌクレオチド配列からなる単離された核酸。 9. ヌクレオチド配列が配列番号16である請求項8に記載の単離核酸。 10.配列番号50のアミノ酸配列をコードするヌクレオチド配列を含んでなる単離 された核酸。 11.配列番号26〜49からなる群から選ばれるヌクレオチド配列を含んでなる単離 された核酸 12.請求項1〜11のいずれかに記載の単離された核酸からなる組み替え発現ベク ター。 13.請求項12に記載の組み替え発現ベクターによってトランスフォーメーション された宿主細胞。 14.動物またはその祖先のゲノムに、 (イ)ヒトRHAMM遺伝子のエキソン少なくとも1個のヌクレオチド配列の 挿入と、 (ロ)ヒトRHAMMタンパク質を少なくとも1個コードするヌクレオチド配 列の挿入と、 (ハ)内因性RHAMM遺伝子の失活と からなる群から選ばれる変更を引き起こす組み替え構成体少なくとも1個が挿入 されることによって変更されている遺伝子導入動物。 15.ヒトHRAMM1,ヒトHRAMM2,ヒトHRAMM3,ヒトHRAMM 4およびヒト HRAMM5からなる群から選ばれる実質的に純粋なタンパク質。 16.ヒアルロナンに結合することができる配列番号4のアミノ酸配列またはその フラグメントまたは類似体を含んでなる請求項15に記載のタンパク質。 17.(イ)配列番号4のアミノ酸配列からの少なくとも5個の連続したアミノ酸 残基と、 (ロ)配列番号4のアミノ酸配列からの少なくとも10個の連続したアミノ酸 残基と、 (ハ)配列番号4のアミノ酸配列からの少なくとも15個の連続したアミノ酸 残基と からなる群から選ばれるアミノ酸配列を含んでなる実質的に純枠なペプチド。 18.ヒトRHAMMの結合ドメイン少なくとも1個からなる実質的に純粋なペプ チド。 19.配列番号1および配列番号17からなる群から選ばれる請求項18に記載のペ プチド。 20.配列番号50のアミノ酸配列を持つ実質的に純粋なペプチド。 21.ヒトRHAMMタンパク質の抗原決定基に選択的に結合する抗体。 22.請求項20のペプチドの抗原決定基に選択的に結合する抗体。 23.請求項21または22に記載の抗体を産生する細胞系。 24.ヒトRHAMMタンパク質少なくとも1個からなる調製物を用意するステッ プと、 該調製物を候補化合物と接触させるステップと、 RHAMMタンパク質が候補化合物と結合しているのを検出するステップと を含んでなるヒトRHAMMタンパク質と選択的に結合する化合物の同定方法。 25.RHAMMタンパク質の結合をアフィニテイクロマトグラフィ、イースト2 ハイブリッド系およびファージ・ディスプレー・ライブラリからなる群から選ば れる方法によって検出する請求項24に記載の方法。 26.哺乳動物の腫瘍サンプルを採取し、腫瘍サンプル中のRHAMMタンパク質 発現 濃度を測定し、RHAMMタンパク質発現濃度が高い場合を予後不良と判定する 、腫瘍を持つ哺乳動物の予後評価方法。 27.組織化学方法、生検サンプル中のRHAMM mRNA濃度測定を含んでな る方法、生検サンプル中のヒトRHAMMエキソン8の発現測定を含んでなる方 法からなる群から選ばれる方法によってRHAMM発現を測定する請求項26に記 載の方法。 28.哺乳動物がヒトであり、、腫瘍が乳房腫瘍である請求項26または27に記 載の方法。 29.RHAMM遺伝子の発現過多を特徴とするヒトの疾患を予防または治療する 医薬組成物において、 (イ)RHAMM遺伝子に対する優勢なサプレッサー変異体と、 (ロ)ヒトRHAMM cDNAに対するアンチセンス配列と、 (ハ)薬学的に許容できる担体に担持されている、ヒトRHAMM遺伝子のエ キソン8に対するアンチセンス配列と からなる群から選ばれるヌクレオチド配列の有効量を含んでなる医薬組成物。 30.RHAMM遺伝子の発現過多を特徴とするヒトの傷害を予防または治療する 方法において、 (イ)RHAMM遺伝子に対する優勢なサプレッサー変異体と、 (ロ)ヒトRHAMM cDNAに対するアンチセンス配列と、 (ハ)ヒトRHAMM遺伝子のエキソン8に対するアンチセンス配列と からなる群から選ばれるヌクレオチド配列の有効量を哺乳動物に投与することか らなる方法。 31.該疾患が癌である請求項30に記載の方法。 32.ヒトにおける細胞移動を抑制する方法であって、 (イ)ヒトRHAMMタンパク質またはそのフラグメントに特異的に結合する 抗体と、 (ロ)ヒトRHAMMのHA結合ドメインを含んでなるペプチドと からなる群から選ばれる薬剤をヒトに投与することからなる方法。
