Claims (1)
- 【特許請求の範囲】 1.ヒトDNアーゼI過反応性変異体。 2.直線状DNA切断アッセイにおいて測定すると天然のヒトDNアーゼIのものより 少なくとも50%大きいDNA加水分解活性を有する請求項1の変異体。 3.直線状DNA切断アッセイにおいて測定すると天然のヒトDNアーゼIのものより 少なくとも2倍大きいDNA加水分解活性を有する請求項1の変異体。 4.図1に示された天然のヒトDNアーゼIのアミノ酸配列と少なくとも90%の同 一性を有するアミノ酸配列を含む請求項1の変異体。 5.図1に示された天然のヒトDNアーゼIのアミノ酸配列と少なくとも95%の同 一性を有するアミノ酸配列を含む請求項1の変異体。 6.図1の配列内で単一の位置でのみの一アミノ酸からもう一つのアミノ酸への 置換によって、図1に示されたアミノ酸配列とは異なるアミノ酸配列を有するヒ トDNアーゼI過反応性変異体。 7.アミノ酸置換が、図1の配列内で以下の位置:Gln9,Glu13,Thr14,His44,Asn7 4,Ser75及びThr205の一つである請求項6の変異体。 8.図1の配列内で二つ以上の位置での一アミノ酸からもう一つのアミノ酸への 置換によって、図1に示されたアミノ酸配列とは異なるアミノ酸配列を有するヒ トDNアーゼI過反応性変異体。 9.少なくとも一つのアミノ酸置換が、図1の配列内で以下の位置:Gln9,Glu13, Thr14,His44,Asn74,Ser75及びThr205の一つでなされる請求項8の変異体。 10.ヒトDNアーゼI過反応性変異体をコードする単離された核酸。 11.図1に示された天然のヒトDNアーゼIのアミノ酸配列と少なくとも90%の同 一性を有するアミノ酸配列をコードする核酸配列を含む請求項10の核酸。 12.図1に示された天然のヒトDNアーゼIのアミノ酸配列と少なくとも95%の同 一性を有するアミノ酸配列をコードする核酸配列を含む請求項10の核酸。 13.図1の配列内で単一の位置でのみの一つのアミノ酸からもう一つのアミノ酸 への置換によって、図1に示されたアミノ酸配列とは異なるアミノ酸配列をコー ドする核酸配列を含む請求項10の核酸。 14.図1の配列内で二つ以上の位置での一つのアミノ酸からもう一つのアミノ酸 への置換によつて、図1に示されたアミノ酸配列とは異なるアミノ酸配列をコー ドする核酸配列を含む請求項10の核酸。 15.ヒトDNアーゼI過反応性変異体の治療上の有効量を患者に投与することを含 む、肺疾患または肺病を有する患者の治療方法。 16.該疾患または病気が嚢胞性線維症である請求項15の方法。 17.ヒトDNアーゼI過反応性変異体の治療上の有効量を患者に投与することを含 む、全身性エリテマトーデスを有する患者の治療方法。 18.ヒトDNアーゼI過反応性変異体及び場合により製薬学的に許容可能な賦形剤 を含む製薬学的組成物。 19.該組成物が液体形態で存在する請求項18の組成物。 20.該組成物がパウダー形態で存在する請求項18の組成物。
