Claims (1)
- 【特許請求の範囲】 1.サンプル中に存在し得る広く分岐した微生物の少なくとも二つの群の中か らある微生物を同定することができる、サンプル中の微生物を同定するためのシ ステムであって、 あらかじめ定めた数の反応チャンバー中に配置された非冗長生化学テストのあ らかじめ定めた組合わせを含み、めいめいの生化学テストはある酵素または酵素 群のための基質を含み、該基質はもしその酵素または酵素群が作用すれば反応チ ャンバー中に検出可能な産物の生成をもたらし、そして 生化学テストの組合わせからの検出可能な産物がサンプル中の微生物を同定す るために使用される ことを特徴とする前記テストシステム。 2.微生物の同定は微生物の属または種またはその両方へ分類する請求項1の テストシステム。 3.非冗長テストのあらかじめ定めた組合わせは蛍光テスト、比色定量テスト またはその組合わせを含む請求項1のテストシステム。 4.蛍光テストはフルオロゲン法または蛍光定量法で実施される請求項3のテ ストシステム。 5.非冗長テストのあらかじめ定めた組合わせは約15分ないし約8時間内で 実施される請求項3のテストシステム。 6.非冗長テストのあらかじめ定めた組合わせは約25ないし約37℃の温度 において実施される請求項1のテストシステム。 7.比色定量テストは内眼によりまたは比色計によって読取られ る請求項6のテストシステム。 8.蛍光テストは蛍光計を使用して読取られる請求項4のテストシステム。 9.ペプチダーゼ、グリコシダーゼ、糖発酵酵素、ウレアーゼ、デカルボキシ ラーゼ、エステラーゼ、炭素利用酵素、フォスファターゼまたはトリプトファナ ーゼを検出するための少なくとも一つのテストを含んでいる請求項1のテストシ ステム。 10.少なくとも一つのペプチターゼおよび少なくとも一つグリコシダーゼの ためのテストを含んでいる請求項9のテストシステム。 11.少なくとも一つの炭素発酵酵素のためのテストをさらに含んでいる請求 項10のテストシステム。 12.少なくとも一つのウレアーゼのためのテストをさらに含んでいる請求項 11のテストシステム。 13.少なくとも一つのデカルボキシラーゼのためのテストをさらに含んでい る請求項12のテストシステム。 14.少なくとも一つのエステラーゼのためのテストをさらに含んでいる請求 項13のテストシステム。 15.少なくとも一つの炭素同化酵素のためのテストをさらに含んでいる請求 項14のテストシステム。 16.非冗長生化学テストのあらかじめ定めた組合わせは、腸内細菌および非 発酵細菌;嫌気性細菌,イースト;Staphylococcus sp.また はStreptococcus sp.またはEnterococcus sp ;および偏好性細菌の少なくとも二つの間で微生物を同定することが可能である 請求項1のテ ストシステム。 17.非冗長生化学テストのあらかじめ定めた組合わせは、Staphylo coccus sp.とStreptococcus sp.とそしてEnte rococcus sp;Corynebacterua sp.;Lacto bacillus sp.;Pediococcus sp.;Leucono stococcus sp.;Alloicoccus sp.;Vagoco ccus sp.;Kluyvera sp.;Leminorella sp .;Haemophilus sp.およびNeisseria sp.;Sa lmonella sp.;Clostridia sp.;およびListe ria sp.の少なくとも一つの間で微生物を同定することが可能である請求 項1のテストシステム。 18.非冗長生化学テストのあらかじめ定めた組合わせは、嫌気性菌、イース トまたは偏好性細菌の間で微生物を同定することが可能である請求項1のテスト システム。 19.非冗長生化学テストのあらかじめ定めた組合わせは、嫌気性菌およびイ ースト、または偏好性細菌の間で微生物を同定することが可能である請求項1の テストシステム。 20.非冗長生化学テストのあらかじめ定めた組合わせは、イーストおよび偏 好性細菌の間で微生物を同定することが可能である請求項1のテストシステム。 21.非冗長生化学テストのあらかじめ定めた組合わせは、Staphylo coccus,StreptococcusまたはEnterococcusの 少なくとも一つ、および嫌気性菌、イー ストおよび非発酵菌または偏好性細菌またはそれらの組合わせの少なくとも一つ の間で微生物を同定することが可能である請求項1のテストシステム。 22.テストシステムはイースト、嫌気性細菌および偏好性細菌の間で同定す ることが可能である請求項1のテストシステム。 23.テストシステムはEnterococcus,Staphylococ cus,またはStreptococcus;嫌気性細菌;イースト;および偏 好性細菌を同定することが可能である請求項1のテストシステム。 24.基質はリジン(AMC),ロイシン(AMC),メチオニン(AMC) ,グリシルプロリン(AMC),イソロイシン(AMC),トレハロース,マル トース,L−トリプトファン(7−AMC),4−MeU−フォスフェート,β −D−キシロシド−4MeU,ヒドロキシプロリン−AMC,β−D−グルクロ ニド−4MeU,チロシン(AMC),4−MeU−β−D−ガラクトシド,マ ンノース,スクロース,およびプロリン(AMC)を含んでいる請求項1のテス トシステム。 25.基質はフラクトース,グリセロース,L−ヒスチジン7−AMC,ピロ グルタミン酸(AMC)および4−MeU−β−D−グルコシドをさらに含んで いる請求項24のテストシステム。 26.基質はL−セリン7−AMC,セロビオース,アルギニン(AMC)お よび4−MeU−N−アセチル−β−D−ガラクトサミニドをさらに含んでいる 請求項25のテストシステム。 27.請求項1のテストシステムの使用により、サンプル中に存在し得る広く 分岐した微生物の少なくとも二つの群の中からある微 生物を同定する方法であって、 a)ある基質を含んでいる各反応チャンバーへサンプルを加え、 b)もし存在すれば酵素が該基質と反応することを許容し、 c)テスト中の検出可能な産物を検出することによってサンプル中の酵素の存 在を測定し、 d)あらかじめ定めたテストの組合わせの結果を少なくとも一つのあらかじめ 定めた標準と比較し、サンプル中の微生物を同定することを含む前記方法。 28.微生物による炭素源利用を検出するためのテストであって、少なくとも一 つの炭素源および少なくとも一つ蛍光定量指示薬を含み、微生物は指示薬の蛍光 の変化を生ずるpH変化を発生するように炭素源に作用し、蛍光変化は微生物の 炭素源利用の指標である前記テスト。
