Claims (1)
- 【特許請求の範囲】 1. 組換え真菌宿主細胞を獲得する方法であって、核酸配列中の少なくとも1 つの陰性スプライス部位が変更されている異種ポリペプチドをコードする核酸配 列を、真菌宿主細胞に導入することを含んで成る方法。 2. 前記少なくとも1つの陰性スプライス部位が、少なくとも1つの陰性共通 配列を非共通配列で置き換えることにより変更される、請求項1に記載の方法。 3. 前記陰性共通配列が5’陰性共通配列である、請求項2に記載の方法。 4. 前記5’陰性共通配列がGT,GCまたはCTである、請求項3に記載の 方法。 5. 前記5’陰性共通配列がGTANGT,GCANGTまたはCTANGT であり、ここでNはA,C,GまたはTである、請求項4に記載の方法。 6. 前記陰性共通配列が3’陰性共通配列である、請求項2に記載の方法。 7. 前記3’陰性共通配列がAGである、請求項6に記載の方法。 8. 前記3’陰性共通配列がCAG,TAGまたはAAGである、請求項6に 記載の方法。 9. 前記真菌宿主細胞により生産される異種ポリペプチドのアミノ酸配列が野 性型ポリペプチドである、請求項1に記載の方法。 10.前記少なくともつの陰性スプライス部位が、少なくとも1つの陰性イント ロンまたはその部分を含んで成る第一領域を、約40%〜約70%の範囲のG+C含 量を有する第二領域で置き換えることにより変更される、請求項1に記載の方法 。 11.前記G+C含量が約40%〜約60%の範囲である、請求項10に記載の方法。 12.前記G+C含量が約40%〜約50%の範囲である、請求項11に記載の方法。 13.少なくとも2つの陰性イントロンまたはその部分が置き換えられる、請求 項10に記載の方法。 14.前記核酸配列中の陰性共通配列を非共通配列で置き換えることと、1つの 陰性イントロンを含む第一領域を約40%〜約70%の範囲のG+C含量を有する第 二領域で置き換えることとの両方により、少なくとも1つの陰性スプライス部位 が変更される、請求項1に記載の方法。 15.少なくとも2つの陰性スプライス部位が変更される、請求項1に記載の方 法。 16.前記真菌宿主細胞により生産される異種ポリペプチドが、対応する野性型 ポリペプチドと同じ数のアミノ酸残基を含む、請求項1に記載の方法。 17.前記非共通配列が前記共通配列と同じ数のヌクレオチドを有する、請求項 1に記載の方法。 18.前記核酸配列がホルモン、酵素、レセプターまたはレポーターをコードす る、請求項1に記載の方法。 19.前記核酸配列が酵素をコードする、請求項1に記載の方法。 20.前記酵素がオキシドレダクターゼ、トランスフェラーゼ、ヒドロラーゼ、 リアーゼ、イソメラーゼまたはリガーゼである、請求項18に記載の方法。 21.前記酵素がアミノペプチダーゼ、アミラーゼ、カルボヒドラーゼ、カルボ キシペプチダーゼ、カタラーゼ、セルラーゼ、キチナーゼ、クチナーゼ、デオキ シリボヌクレアーゼ、エステラーゼ、α −ガラクトシダーゼ、β−ガラクトシダーゼ、グルコアミラーゼ、α−グルコシ ダーゼ、β−グルコシダーゼ、ハロペルオキシダーゼ、インベルターゼ、ラッカ ーゼ、リパーゼ、マンノシダーゼ、ムタナーゼ、オキシダーゼ、ペクチン分解酵 素、ペルオキシダーゼ、フィターゼ、ポリフェノールオキシダーゼ、タンパク質 分解酵素、リボヌクレアーゼおよびキシラナーゼから成る群より選ばれる、請求 項19に記載の方法。 22.前記核酸配列がレポーターをコードする、請求項18に記載の方法。 23.前記レポーターがクラゲ(Aequorea victoria)緑色蛍光タンパク質であ る、請求項22に記載の方法。 24.前記真菌細胞が糸状菌細胞である、請求項1に記載の方法。 25.前記糸状菌細胞がアクレモニウム、アスペルギルス、フザリウム、フミコ ラ、ミセリオフトラ、ムーコル、ニューロスポラ、ペニシリウム、チエラビア、 トリポクラディウムまたはトリコデルマの種の細胞である、請求項24に記載の方 法。 26.前記糸状菌細胞がアスペルギルス細胞である、請求項24に記載の方法。 27.前記アスペルギルス細胞がアスペルギルス・オリゼ細胞、アスペルギルス ・ニガー細胞、アスペルギルス・フェティダス細胞、またはアスペルギルス・ジ ャポニカス細胞である、請求項26に記載の方法。 28.前記糸状菌細胞がフザリウム細胞である、請求項24に記載の方法。 29.前記フザリウム細胞がフザリウム・オキシスポラム細胞またはフザリウム ・グラミネアラム細胞である、請求項28に記載の方法。 30.前記糸状菌細胞がフミコラ細胞である、請求項24に記載の方 法。 31.前記フミコラ細胞がフミコラ・インソレンス細胞またはフミコラ・ラヌギ ノザス細胞である、請求項30に記載の方法。 32.前記糸状菌細胞がミセリオフトラ細胞である、請求項24に記載の方法。 33.前記ミセリオフトラ細胞がミセリオフトラ・テルモフィラ細胞である、請 求項32に記載の方法。 34.前記糸状菌細胞がムーコル細胞である、請求項24に記載の方法。 35.前記ムーコル細胞がムーコル・ミーヘイ細胞である、請求項34に記載の方 法。 36.前記糸状菌細胞がニューロスポラ細胞である、請求項24に記載の方法。 37.前記ニューロスポラ細胞がニューロスポラ・クラッサ細胞である、請求項 36に記載の方法。 38.前記糸状菌細胞がペニシリウム細胞である、請求項24に記載の方法。 39.前記ペニシリウム細胞がペニシリウム・プルプロゲナム細胞である、請求 項38に記載の方法。 40.前記糸状菌細胞がチエラビア細胞である、請求項24に記載の方法。 41.前記チエラビア細胞がチエラビア・テレストリス細胞である、請求項40に 記載の方法。 42.前記糸状菌細胞がトリコデルマ細胞である、請求項24に記載の方法。 43.前記トリコデルマ細胞がトリコデルマ・リーセイ細胞、トリコデルマ・ビ リデ細胞、トリコデルマ・ロンギブラキアタム細胞、 トリコデルマ・ハージアナム細胞またはトリコデルマ・コニンギイ細胞である、 請求項42に記載の方法。 44.前記真菌細胞が酵母細胞である、請求項1に記載の方法。 45.前記酵母細胞がカンジダ、クルイベロミセス、サッカロミセス、シゾサッ カロミセス、ピヒアまたはヤロウィアの種の細胞である、請求項44に記載の方法 。 46.前記酵母細胞がサッカロミセス細胞である、請求項45に記載の方法。 47.前記サッカロミセス細胞がサッカロミセス・セレビシエ細胞、サッカロミ セス・カールスバーゲンシス細胞、サッカロミセス・ジアスタティクス細胞、サ ッカロミセス・ドウグラシイ細胞、サッカロミセス・クルイベリ細胞、サッカロ ミセス・ノルベンシス細胞またはサッカロミセス・オビフォルミス細胞である、 請求項46に記載の方法。 48.前記酵母細胞がクルイベロミセス細胞である、請求項45に記載の方法。 49.前記クルイベロミセス細胞がクルイベロミセス・ラクティス細胞である、 請求項48に記載の方法。 50.前記酵母細胞がヤロウィア細胞である、請求項45に記載の方法。 51.前記ヤロウィア細胞がヤロウィア・リポリティカ細胞である、請求項50に 記載の方法。 52.真菌宿主細胞中での異種発現のために単離された核酸配列から少なくとも 1つの陰性スプライス部位を変更する方法であって、前記核酸配列中の陰性共通 配列を非共通配列で置き換え、そして/または、1つの陰性イントロンを含む第 一領域を、前記第一領域のG+C含量に比較して約40%〜約70%の範囲に調整さ れたG+C含 量を有する第二領域で置き換えることを含んで成る方法。 53.請求項52の方法に従って得られる単離された核酸配列。 54.請求項53の核酸配列を含んで成る核酸構成物。 55.請求項54の核酸構成物を含んで成る組換え発現ベクター。 56.前記核酸配列がプロモーター配列に作用可能に連結されている、請求項55 に記載のベクター。 57.前記核酸配列が転写終結シグナルに作用可能に連結されている、請求項55 に記載のベクター。 58.選択マーカーを更に含んで成る、請求項55に記載のベクター。 59.請求項54の核酸構成物を含んで成る組換え真菌宿主細胞。 60.請求項1の方法に従って得られる組換え真菌宿主細胞。 61.真菌宿主細胞中でのポリペプチドの異種発現方法であって、請求項59の真 菌細胞を栄養培地中で培養しそして前記培地から前記ポリペプチドを回収するこ とを含んで成る方法。
