筋骨格系の状態、たとえば開放骨折および非癒合骨折、ならびに著しい骨損失
を伴う粗結合プロテーゼの処置には、予防用および治療用として抗生物質および
鎮痛薬を用いる。この薬物を局所的に送達すると、経口および非経口投与に伴う
全身毒性および副作用の危険性が低下し、一方では目的組織においてより高い薬
物濃度が得られることにより処置効率も改善されるという二重の利点がある。No

増殖に影響を与え、感染と戦い、および/または痛みを抑制するために、骨部
位において生物学的活性分子を放出しうる材料が求められている。理想的な送達
ビヒクルは、制御された様式で、かつその薬物の最適有効性をもたらすのに十分
なほど長期間、生物学的分子を放出すべきである。さらに、著しい骨再生が必要
な場合、骨組織増殖の足場としても役立ちうる生物学的分子キャリヤーも有益で

うな用途に重要である。
ゾル−ゲル誘導ガラスは機能性の生物学的分子を制御された様式で放出しうる
ので、骨の療法に用いるための制御放出材料としてきわめて有望である。Radin

れた。これらの新規材料は室温で調製され、シリカ系、無定形であり、かつ高い
超顕微鏡的多孔度をもつ。
ゾル−ゲル誘導処理は、低温で−−すなわち約40℃以下で−−かつ低pHで
実施できる。これらの条件は共に、生物学的活性分子を均一に取り込むために、
かつゾル−ゲルマトリックスに取り込まれた生物学的活性分子の機能性を維持す
るために、重要となる可能性がある。ゾル−ゲル誘導処理の利点には以下のもの
が含まれる:1)コロイドサイズ粒子の懸濁液であるゾルは、ゲル化する前は液
状である:2)反応全体を室温で実施できる:3)ゾル−ゲルガラスの微孔度を
、たとえば水分、プロトン添加時期、プロトン濃度、および乾燥時間により制御
できる。ゾル−ゲル処理により得られる孔径は一般にナノメートルの範囲である
。反応物が液相である間に、それがゲル化する前に、タンパク質その他の生物学
的活性分子を液状ゾルに添加できる。その際、これらの分子は固体マトリックス
中に封入された状態になる。その後、浸漬または移植されると、これらの分子が
制御放出される。
しかし、移植したゾル−ゲル誘導シリカ系ガラスについてのインビボ研究で、
これらのガラスは一部はその溶解性のためきわめて反応性である可能性があり、
炎症反応を起こす可能性のあることが明らかになった。室温で処理した4種類の
ゾル−ゲル誘導ガラスの顆粒を、ウサギの腸骨稜に形成した直径5mm、深さ2
.5mmの欠損部に2または4週間移植した。3種類の材料は100%SiO2
からなり(S100)、1種類は(重量%)70%SiO2−25%CaO−5
%P2O5を含有していた(S70)。S100材料のうち1種類は、さらに材料
1gにつき1.2mgのバンコマイシンを含有していた(S100V)。すべて
の材料を500〜710μmの顆粒として試験した。
移植期間終了時に回収した試料の組織学的分析により、移植したゾル−ゲル顆
粒の有意の分解、すなわちサイズの縮小が明らかになった。大部分の材料吸収に
典型的なように、分解により炎症反応が起きた。これは顆粒の周りや間に食細胞
が存在したことにより示された。分解および炎症反応の程度は、S70材料の場
合の方が大きかった。移植に際しての炎症反応を最小限に抑えるためには、分解
速度のより良い制御が確実である。
インビボ試験の所見は、インビトロでみられたものと一致した。インビボ状態
での非平衡を模倣するために溶液を頻繁に交換してインビトロで溶解実験を実施
すると、速やかなケイ素溶解がみられ、溶解速度はS100ガラスの場合よりS
70の方が速やかであった。
組成物中にバンコマイシンが存在すると炎症反応は減少したが、インビボ活性
をより良く制御するためにガラス組成物を安定化する手段が必要である。
発明の概要
1態様において本発明は、生理学的溶液に接触した際のケイ素の溶解を低下さ
せるために処理したゾル−ゲル誘導シリカ系ガラス組成物に関する。これらの組
成物はカルシウム−ホスフェート(calcium-phosphate)表面層で囲まれたケイ素
含有ガラス材料を含む。他の態様において本発明は、ケイ素で飽和した溶液に浸
漬することによりゾル−ゲル誘導シリカ系ガラス組成物を安定化する方法に関す
る。これによりその後生理的溶液に接触した際のケイ素の溶解速度が低下する。
他の態様において本発明は、最初はケイ素を含有しない溶液に浸漬してケイ素
を速やかに溶液中へ溶解させ(ゾル−ゲルの損失は1重量%以下)、これによっ
てケイ素を飽和させることにより、ゾル−ゲル誘導シリカ系ガラスを処理してカ
ルシウム−ホスフェート表面層を形成する方法に関する。

発明の詳細な説明
本発明は、ゾル−ゲル誘導シリカ系ガラスをガラス表面にカルシウムホスフェ
ート(calcium phosphate)皮膜が形成されるように処理する方法、およびそれに
より得られた組成物を伴う。このカルシウム−ホスフェート(calcium-phosphate
)皮膜は、ガラスの著しい分解なしに形成される。カルシウム−ホスフェート皮
膜は、生物学的活性分子をも含むこともできる。
従来の研究で、SiO2、CaOおよびP2O5系の熱処理したゾル−ゲル誘導
ガラス(Pereira et al.,“インビトロでのゾル−ゲル誘導−生物学的活性ガラ
ス上のリン酸カルシウム形成”,J.Biomed.Res.,28:693-8,1994)が、純粋な
シリカゲル(Li et al.,“模擬体液中でシリカゲルにより誘発されるアパタイト
形成”,J.Amer.Ceram.Soc.,75:2094-7,1992)と同様に、模擬生理学的溶液
に浸漬した際にアパタイト形成を誘発しうることが示された。室温で調製したゾ
ル−ゲルが、Ca2+およびPO4 2-を含有する溶液中でもアパタイト形成を誘発
できる。たとえば米国特許出願第08/477,585号参照。
本発明によれば、ゾル−ゲル誘導シリカ系ガラスを、ケイ素で飽和した、カル
シウムおよびホスフェートを含有する溶液に該ガラスを浸漬することにより処理
して、ガラスの著しい分解なしにカルシウム−ホスフェート表面層を形成するこ
とができる。後記のように、これらのガラスは、生物学的活性分子をマトリック
スに取り込んだ状態で調製できる。分解は取り込まれた分子の損失をもたらすの
で、分解を阻止し、または最小限に抑えると、生物学的活性分子がより良く保持
される。これにより、生物学的分子を制御放出性の生体適合性キャリヤーに取り
込ませることを意図した製造プロセスの全収率が改良される。この処理の他の観
点は、酸触媒されたゾル−ゲル誘導ガラスが溶解−沈殿反応により分子レベルで
再構成され、これによってより大きな安定性がガラスに付与されることである。
本方法は、45S5ガラスのものと近似する組成以外の組成をもつ溶融誘導−
生物学的活性ガラスからの同様な組成物、すなわち約55〜約65%の濃度のケ
イ素を含有する組成物にも、カルシウム−ホスフェート表面層の形成のために適
用できるものとする。他の化合物および酸化物、たとえばP.Ducheyne,“移植
材料としてのバイオガラス皮膜およびバイオガラス複合材料”,J.Biomed.Mat
.Res.,19:273-291,1985(本明細書において援用される)に記載されるものが
存在してもよい。
ゾル−ゲル誘導ガラスは、たとえば米国特許出願第08/477,585号(本明細書に
おいて援用される)に開示される方法で製造できる。ゾル−ゲル誘導ガラスの組
成範囲は以下のとおりである(重量%):SiO2−60〜100;CaO−3
〜30;およびP2O5−0〜10。
本明細書において用いる“著しいガラスの分解なしに”とは、約10%以下の
ケイ素損失を意味する。
本明細書において用いる“約”とは、その数値のほぼ±10%の変更を意味す
る。
本明細書において用いる“シリカ系(silica-based)”とは、ガラス組成物に酸
化ケイ素が含有されることを意味する。他の酸化物が存在してもよい。
本発明は、取り込まれた生物学的活性分子を含むか、または含まないゾル−ゲ
ルガラスを、Ca、PおよびSi含有溶液に浸漬して、溶解により誘発された物
理化学的反応により表面Ca−P層を形成することを含む。浸漬中にシリカ網状
構造が溶解するのを防ぐために、その上にCa−P層を形成すべきシリカ系材料
を浸漬する処理溶液のSi含量は、その溶液のケイ素溶解度と等しいか、または
より大きくなければならない。処理中に、ゾル−ゲル合成の残留副生物、たとえ
ばアルコール類も脱離させることができる。
Ca−、P−およびSi−含有溶液に浸漬した際のCa−P層形成に伴う反応
は以下のものであると考えられる:
1)100%SiO2ゾル−ゲルガラスの場合、層形成は溶液中に存在する
CaおよびP−イオンが表面シラノール(SiOH)と錯体形成することに
より起きる;
2)CaP含有ゾル−ゲルガラスの場合、材料から溶液へのCaイオンの拡
散によりCaP−相に関して溶液の過飽和が高まり、次いでこの相がガラス
表面に析出する。
処理条件(W/V比、溶液の組成およびpH、温度、浸漬の程度など)は、後記
に詳述する具体例により限定されず、むしろおおはばに変更できる。当業者に自
明のとおり、これら種々の条件は関連しており、したがって本発明を達成するよ
うに調整できる。S100ガラスについてのパラメーターは、少なくとも以下の
ように変更できる:pH−約6〜約9;浸漬時間−最高約10日間;Ca含量−
約2〜約15mM;全PO4(HPO4およびH2PO4を含む)約1〜約20mM;
他のイオンも溶液中に存在してもよく、これにはNa、K、Cl、Mg、CO3
およびSO4、ならびにその組合わせが含まれるが、これらに限定されない。た
とえば10重量%の酸化カルシウムを含有するCaP含有ゾル−ゲルガラスにつ
いては、これらのパラメーターは少なくとも以下のように変更できる:pH−約
6〜約9;浸漬時間−最高約3日間;Ca含量(溶存)−約0.5〜約5mM;
全PO4含量(溶存)約1〜約20mM;他のイオンも溶液中に存在してもよく
、これにはNa、K、Cl、Mg、SO4およびCO3、ならびにその組合わせが
含まれるが、これらに限定されない。用いる溶液はいずれか適した緩衝剤で緩衝
化されていてもよく、これにはトリス緩衝液およびリン酸緩衝液が含まれるが、
これらに限定されない。
最初はケイ素を含有しない溶液に浸漬してケイ素を速やかに溶液中へ溶解させ
(ゾル−ゲルの損失は1重量%以下)、これによってケイ素を飽和させることに
より、ゾル−ゲル誘導シリカ系ガラスを含むカルシウム−ホスフェート表面層を
材料上に形成しうることも見出された。たとえば後記実施例3に詳述した条件下
では、カルシウム−ホスフェート皮膜が数時間以内に形成された。
処理されるガラス材料は、各種サイズの顆粒、ディスク、棒、またはブロック
であってよい。あるいは処理されるガラス材料は、他の材料上のコーティングで
あってもよい。
酸化カルシウムを含有するか、または含有しないゾル−ゲル誘導シリカ系ガラ
スは、たとえば米国特許出願第08/477,585号(本明細書において援用される)に
開示された方法で製造できる。カルシウムアルコキシドおよびカルシウム塩類を
共に酸化カルシウム源として使用できる。もちろん他のゾル−ゲル法もシリカ系
ガラスの製造に使用できる。
種々の生物学的活性分子を含有する溶液中で処理を行って、治癒や修復のプロ
セスを増強するために生物学的活性分子を取り込んだ保護コーティングを形成す
ることができる。生物学的活性分子は処理中に存在してもよく、またはその後、
たとえば血清その他の生理的媒質を溶存関連分子と共に含有する組織培養培地に
浸漬することにより吸収させてもよい。あるいは、前記の米国特許出願第08/477
,585号に開示された方法で生物学的活性分子をガラスマトリックスに取り込ませ
たゾル−ゲル誘導−生物学的活性ガラスを製造することができる。
本明細書中で用いる“生物学的活性分子”は、生物学的系中で作用を示す有機
分子であると定義され、その系はインビトロ、インビボ、または原位置(in sit
u)のいずれであってもよい。生物学的活性分子には以下のカテゴリーが含まれ
るが、これらに限定されない:増殖因子、サイトカイン、抗生物質、抗炎症薬、
鎮痛薬および他の薬物、ならびに細胞接着分子。
“抗生物質”という用語には、実質的に水溶性である殺菌性、殺真菌性、およ
び感染防止性の薬物、たとえばゲンタマイシン、バンコマイシン、ペニシリンお
よびセファロスポリン類が含まれる。
“増殖因子”という用語は、造骨細胞、線維芽細胞、神経細胞、内皮細胞、上
皮細胞、ケラチノサイト、軟骨細胞、筋細胞、関節靭帯由来の細胞、髄核由来の
細胞などの細胞の機能に影響を与える因子を意味するが、これらに限定されない
。血小板由来増殖因子(PDGF)、トランスフォーミング増殖因子(TGF−β)
、インスリン様増殖因子(IGF)、線維芽細胞増殖因子(FGF)、および骨
形成タンパク質(BMP)が、本発明による粒子に含有される増殖因子の例である
。
本明細書中で用いる“細胞接着分子”という用語には、フィブロネクチン、ビ
トロネクチン、コラーゲン−タイプI、オステオポンチン、骨シアロプロテイン
であるトロンボスポンジン、およびフィブリノーゲンが含まれるが、これらに限
定されない。それらの分子は足場依存性細胞の接着に重要である。
本明細書中で用いる“接触”という用語には、浸漬、移植および埋込みによる
接触が含まれるが、これらに限定されない。
実施例1
ゾル−ゲルの調製
3種類のゾル−ゲル誘導粒子を合成した:100%シリカ(S100)、バン
コマイシン−シリカ複合体(S100V)、ならびにカルシウムおよびリン含有
シリカ(S70)。S100Vはバンコマイシン1.2mg/gを含有していた。
S70は70%SiO2、25%CaOおよび5%P2O5(重量%)からなって
いた。
アルコキシド類であるテトラメチルオルトシリケート(TMOS−アルドリッ
チ・ケミカル社、ワイオミング州ミルウォーキー)、カルシウムメトキシエトキ
シド(CME−ゲレスト社、ペンシルベニア州タリータウン)およびリン酸トリ
エチル(TEP−ストレム・ケミカル社、マサチュセッツ州ニューベリーポート
)を、それぞれケイ素、カルシウムおよびリン源として用いた。
S100およびS100Vは、1:10のモル比のTMOSと脱イオン(DI
)水の混合物から調製された。以下の方法を用いた。調製した容量につきモル比
を維持するように予め定めた量のTMOSおよび脱イオン水をビーカーに注入し
、これを直ちに氷冷超音波浴に入れた。バンコマイシン含有ゾル−ゲルについて
は、この時点で添加する水の量は、後に抗生物質と共に添加する水の量を除いた
ものである。相分離を防ぐために溶剤の不在下で超音波処理を行った。超音波は
エネルギーを生じるので、過熱を防ぐために氷冷を採用した。次いで少量の酸触
媒(1N HCl)を添加した。数分以内に混合物は均質になった。超音波撹拌
を20〜25分間続け、溶液のpHを測定した。pHは3を越えなかった。この
ゾルをなお撹拌および冷却しながら、予め調製したDI中バンコマイシン(バン
コールド(Vancold)−リーデルレ・パレンタールズ社、カロライナ、プエルト
リコ)溶液を、必要な場合には添加した。この溶液をさらに5分間撹拌した。こ
れらのゾルを、予め秤量したポリスチレンバイアルにメスピペットで注型した。
各バイアルに3mlを分注した。次いでバイアルを秤量し、シールし、放置して
ゲル化を起こさせた。ゲル化時間は約15時間であった。
バイアルをシールしたまま、ゲルを3日間放置して熟成させた。次いでゲルを
周囲空気に暴露し、乾燥させると、注型直後の重量の70%が失われた。次いで
得られたディスクをセラミック製の乳鉢と乳棒で破砕し、超音波シフターでふる
い分けして、2直径範囲の粒子を得た:210〜500μmおよび500〜71
0μm。得られた粒子を4℃で密閉ポリスチレン容器内に保存した。
カルシウムおよびリンを含有するゾルは、グローブボックス内でアルゴン雰囲
気下に調製された。以下の方法を用いた。最終組成70%SiO2、25%Ca
Oおよび5%P2O5(重量%)のゾル−ゲル組成物になるように予め配分した量
のTMOS、CMEおよびTEPを、連続してビーカーに注入した。この混合物
を約5分間、磁気撹拌した。このゾルを予め秤量したポリスチレンバイアルに、
バイアル当たり2.23ml注型した。注型後、試料をグローブボックスから取
り出した。ゲル化を遅延させるために、メタノールをTMOSに対し1:1のモ
ル比で添加した。その後、直ちに試料を渦撹拌した。次いで酸性溶液に溶解した
生物学的活性分子の添加を模倣して、各試料に0.1N酢酸を添加した。1分以
内にゲル化が起きた。残りの操作はシリカゾル−ゲルの場合と同じであった。
実施例2
ケイ素を補充した溶液中での表面層形成
直径500〜710μmのS100およびS70粒子を、それらの表面にCa
−P皮膜を形成するために処理した。2.5mMケイ素(37℃でpH=7.6
)を補充したTE(ヒト血漿のものに類似する電解質含量をもつトリス緩衝液、
37℃でpH=7.4)に、試料を5日間(S100)および2日間(S70)
浸漬した。このケイ素濃度は、ゾル−ゲルを浸漬したpH7.4のTE中におけ
るケイ素の溶解度に相当する。材料重量−対−溶液容量比0.5mg/mlでコ
ンディショニングを行い、溶液を連続的に振とうした。溶液のカルシウムおよび
リンの濃度を測定することにより、表面析出反応を連続的に監視した。浸漬時間
終了時に粒子を採集し、室温で空気流下に乾燥させた。
実施例3
合成;最初はケイ素を含有しない溶液中での表面層形成
3種類のアルコキシド類−−テトラメチルオルトシラン(TMOS)、カルシ
ウムメトキシエトキシド(CME)およびリン酸トリエチル(TEP)を用いて
、70%SiO2−25%CaO−5%P2O5(重量%)の組成をもつシリカ系
ゾ
ル−ゲルディスク(S70)を合成した。詳細には、3.46mlのTMOS、
0.24mlのTEP、および8.4mlのCMEを、30mlのビーカー内で
アルゴン雰囲気下に磁気撹拌により混合した。5分後に、この混合物をポリスチ
レン容器に試料0.74mlで配分し、これにタンパク質溶液0.26mlずつ
を添加した。タンパク質溶液は0.26mlの0.1N酢酸および1%ウシ血清
アルブミン(BSA)からなっていた。得られたゲルを室温で7日間熟成し(密
閉)、その元の重量の50%にまで乾燥させて、ディスクを得た。乾燥を室温で
行い、得られたディスクが亀裂を生じるのを避けるために、慎重に監視した。デ
ィスクはほぼ直径10mm、高さ4mmであり、表面積1.257cm2となっ
た。粒子を調製するために、ディスクを乳鉢と乳棒で破砕し、ふるい分けして、
直径範囲210〜710μmの粒子を得た。
ディスクおよび粒子を、殺菌したポリスチレン密閉容器内で無菌のダルベッコ
−リン酸緩衝塩類溶液(DPBS)(ギブコBRL/ライフ・テクノロジーズ、
ニューヨーク州グランド・アイランド)に37℃で3時間浸漬することにより、
予備処理した。ディスクについては、表面積−対−溶液容量比0.1cm-1を用
い、したがって表面積1.257cm2につき12.57mlのDPBSを用い
た。粒子は、重量−対−溶液容量比1mg/mlで浸漬された。DPBSは溶液
1リットルにつき下記配合の成分を含有する(37℃での最終pH7.4):0
.10gのCaCl2(無水)、0.20gのKCl、0.20gのKH2PO4
、0.10gのMgCl2(6H2O)、0.80gのNaCl、1.15gのN
a2HPO4、2.16gのNa2HPO4(7H2O)。浸漬試料につき、フーリ
エ変換赤外分光法(FTIR)(ニコレット5DXC)によりP−O変角ピーク
がみられた。これはカルシウムホスフェート層の形成を示す。
実施例4
材料の分析
実施例2に示した粒子を、合成およびコンディショニングの後、ガス収着(ク
アンタクロム・オートソルブ1(Quantachrome Autosorb 1))およびフーリエ
変換赤外分光法(FTIR、ニコレット5DXC)により分析した。
S100、S100VおよびS70につき得られた等温線(分圧に対する吸収
物の容量のプロット)を図1a〜cに示す。等温線の形は、S100が微孔質(
細孔半径<20Å)であり、一方S70が中孔質(20Å<細孔半径<500Å
)であることを示す。ヒステリシスの型は、両組成物につき円筒形の細孔形状で
あることを示唆する。S100にバンコマイシンを添加しても、等温線の形状が
類似することにより示されるように複合材料の超微細構造に有意の影響はなかっ
た。
すべての材料につき、等温線から多点BET表面積、全細孔容量、および平均
細孔半径を判定した。それらのデータを表Iに挙げる。S70は等温線の形状か
ら推定されたように、S100およびS100Vより小さな表面積およびより大
きな孔径を示す。S100およびS70の両方とも、コンディショニング工程に
よりかなり比表面積が縮小し、孔径が増大する。コンディショニングに際し、前
者は中孔質材料の特性を獲得する。さらに、処理済みS100およびS70の孔
径は二ピーク分布をもち、第2相の出現を示唆する。この追加ピークは強度が小
さく、20Å付近にある。したがってそれは、平均孔径が約100ÅであるS7
0の場合の方がより容易に識別できる。追加実験により、このピークは新たに形
成されたCa−P相に帰属させることができる。 処理前の各材料につき得たFTIRスペクトルを図2aに示す。S100の網
状構造振動モードに伴う主な特徴は、465、787、1088および1246
cm-1のバンドである。1088および1246cm-1のバンドが形成する二重
線はSi−O−Si非対称伸縮と帰属される。787cm-1の振動は、Si−O
−Si非対称伸縮に付随する。最後に465cm-1の振動は、Si−O−Si変
角モードと帰属される。さらに561および937cm-1に2つのバンドを観察
できる。それらは、それぞれ四員環構造のSi−O−Si変角およびSi−OH
の伸縮の結果である可能性が大きい。バンコマイシンを添加してもこれらのバン
ドの外観および位置は変化しない。主振動モードに付随するピークの急角度は、
十分に重合したシリカに典型的である。さらに、2800〜3000cm-1の領
域に残存メトキシ基中のC−H伸縮によるバンドがないことは、これらの基が完
全に加水分解されたことを示す。S70については、非対称伸縮および変角に付
随するバンドがS100の場合より幅広く、かつより低い波数へ、すなわちそれ
ぞれ1088から1038cm-1へ、および465から452cm-1へシフトし
ている。シラノール基中のヒドロキシル結合に付随するバンドを、S100につ
いては3740cm-1、S70については3695cm-1にみることができる。
処理後のS100およびS70材料のFTIRスペクトルを図2bに示す。5
60cm-1および604cm-1のP−O変角振動に特徴的な2つのピークの出現
は、血漿質Ca−P相が両キセロゲルの表面に形成されたことを示す。炭素化ヒ
ドロキシアパタイト(c−HA)に特徴的な870cm-1のP−O変角振動も、
コンディショニングした両キセロゲルに検出される。したがって新たに形成され
た相は炭素化ヒドロキシアパタイトである。
実施例5
インビトロ溶解模擬実験
前記実施例2に従って処理したS100およびS70、ならびに未処理S10
0およびS70を、インビトロで模擬生理的溶液、すなわちTEに浸漬した。T
Eは、ヒト血漿の電解質成分を同様な濃度で含有する非タンパク性溶液である(
表II参照)。TEは、試薬用のNaCl、KCl、NaHCO3、MgCl2・
6H2O、MgSO4・7H2O、無水KHPO4およびCaCl2・2H2Oを、0
.05M[ヒドロキシメチル]アミノメタン塩酸塩緩衝溶液に溶解することによ
り調製された。得られたpHは37℃で7.4であった。
浸漬期間中の種々の時点で新鮮な溶液と交換することにより、差別浸漬を行っ
た。この実施例では、浸漬の3、6、9、24、48、72、96、124およ
び168時間後に、試料を新鮮な溶液に浸漬した。これらの間隔は、溶液中に飽
和濃度の2/3未満のSi最大濃度を維持するように選択された。この浸漬プロ
トコールは、移植部位での連続的な体液補給を反映することを意図した。試料を
最高7日間浸漬した。
3試料を試験した。重量−対−溶液容量比は0.5mg/mlであった。試料
を5%CO2雰囲気で37℃のインキュベーターに入れ、連続的に振とうした(
200回転/分)。ガス交換を妨げずに蒸発を最小限に抑えるために、バイアル
にゆるく蓋をした。浸漬が終了した時点で溶液を採集し、回収した粒子をエタノ
ールですすぎ、周囲空気中で乾燥させた。
炎光原子吸光分光法(FAAS、パーキン−エルマー5100PC)によりSi
濃度を測定した。
予め形成したc−HA層がS100およびS70粒子の溶解に与える影響を図
3aおよびbに示す。両試料につき、コンディショニング工程はSi溶解の速度
および量を有意に低下させた。この低下効果はS100の場合の方がはるかに大
きい。処理済みS100からのSiの放出は、相関係数0.998で時間との一
次関係に従う。
以上の実施例は本発明を説明するためのものであり、決して本発明を限定する
ためのものではない。当業者は、請求の範囲に示した本発明の精神および範囲内
での変更をなしうることを理解するであろう。

Claims (1)
- 【特許請求の範囲】 1.ケイ素濃度約55〜約100%のシリカ系ガラス組成物を、ケイ素で飽和 した溶液に、ガラスの著しい分解なしに材料上にカルシウム−ホスフェート外層 を形成するのに十分な時間浸漬することにより製造された材料。 2.シリカ系ガラスがゾル−ゲル誘導によるものである、請求項1記載の組成 物。 3.シリカ系ガラスがさらにカルシウムおよびホスフェートを含む、請求項1 記載の組成物。 4.ガラスがさらに生物学的活性分子を含む、請求項2記載の組成物。 5.ケイ素濃度約55〜約100%のシリカ系ガラス組成物を、ケイ素で飽和 した溶液に、ガラスの著しい分解なしに該ガラス上にカルシウム−ホスフェート 外層を形成するのに十分な時間浸漬することを含む、該ガラス組成物の生体適合 性を高める方法。 6.シリカ系ガラスがゾル−ゲル誘導によるものである、請求項5記載の方法 。 7.シリカ系ガラスがさらにカルシウムおよびホスフェートを含む、請求項5 記載の方法。 8.溶液がさらに生物学的活性分子を含む、請求項5記載の方法。 9.生物学的活性分子が抗生物質を含む、請求項8記載の方法。 10.生物学的活性分子が細胞接着分子を含む、請求項8記載の方法。 11.生物学的活性分子が増殖因子を含む、請求項8記載の方法。 12.最初はケイ素を含有しないがガラスからのケイ素溶解を誘発することに よりガラスからのケイ素損失1重量%以下で該溶液のケイ素飽和が達成される溶 液に、カルシウムおよびホスフェートの外層を形成するのに十分な時間、シリカ 系ガラスを浸漬することを含む、シリカ系ガラスを処理してカルシウムおよびホ スフェートを含む外層を形成する方法。 13.溶液がリン酸緩衝化塩類溶液である、請求項12記載の方法。 14.シリカ系ガラスがゾル−ゲル誘導によるものである、請求項12記載の 方法。 15.シリカ系ガラスがさらにカルシウムおよびホスフェートを含む、請求項 12記載の方法。 16.溶液がさらに生物学的活性分子を含む、請求項12記載の方法。 17.生物学的活性分子が抗生物質を含む、請求項16記載の方法。 18.生物学的活性分子が細胞接着分子を含む、請求項16記載の方法。 19.生物学的活性分子が増殖因子を含む、請求項16記載の方法。
