Description
Claims (1)
- 【特許請求の範囲】 1.プレ−治療用分子であって、 a)標的プレ−mRNAに対する結合領域、 b)3’スプライス部位、5’スプライス部位、または3’および5’スプライ ス部位、 c)RNAとして機能する配列、または治療用タンパク質、有用なタンパク質、 もしくは遺伝子産物をコードする配列、 d)スプライス調節配列、および e)治療用RNAのスプライスと産生に必要な配列成分、を含む上記分子。 2.結合は相補性または他の手段により仲介される、請求の範囲第1項記載の分 子。 3.治療用または有用なタンパク質をコードする配列を含む、請求の範囲第1項 記載の分子。 4.特定の標的細胞中の機能性の治療用RNAまたは核酸類似体の産生方法であ って、請求の範囲第1項記載のプレ−治療用分子を投与するか、またはプレ−治 療用分子のin situ発現を起こさせて、ユニークなプレ−mRNAまたは プレ−mRNAの集団を発現する細胞を標的とし、ここでユニークなプレ−mR NAは、プレ−治療用RNAによるトランス−スプライスの基質であり、トラン ス−スプライスは機能性の治療用RNAを産生する、上記方法。 5.治療用RNAは、治療用または有用なタンパク質をコードする配列を含む、 請求の範囲第3項記載の方法。 6.動物、植物、および下等真核生物よりなる群から選択される被験体に、特定 の標的細胞中の治療用RNAを提供する方法であって、被験体(被験体は、ユニ ークなプレ−mRNAまたはプレ−mRNAの集団を発現する細胞を有し、この ユニークなプレ−mRNAは、プレ−治療用分子によるトランス−スプライスの 基質であり、トランス−スプライスは機能性の治療用RNAを産生する)に請求 の範囲第1項のプレ−治療用分子を投与することからなる、上記方法。 7.動物はヒトである、請求の範囲第6項記載の方法。 8.治療用RNAは、治療用または有用なタンパク質をコードする配列を含む、 請求の範囲第6項記載の方法。 9.分子は、RNA、DNA、ペプチド核酸、および他の核酸類似体よりなる群 から選択される、請求の範囲第1項記載の分子。 10.請求の範囲第1項記載の分子と分子を投与するための手段を含むキット。
