- 【特許請求の範囲】 1.a)液体および標的配列を含む試験試料を提供する段階と、 b)前記試験試料を酸性化して前記標的配列を第二の液体中に置く段階と、お よび c)前記第二の液体を、核酸増幅に適する緩衝液と置き換える段階と を含む、核酸増幅検定における阻害を軽減する方法。 2.段階b)の前に、前記標的配列を塩基性リン酸塩含有溶液と接触させる、請 求の範囲第1項に記載の方法。 3.前記試験試料が臨床試験試料である請求の範囲第2項に記載の方法。 4.前記試験試料かさらにカルシウムイオンを含む、請求の範囲第1項に記載の 方法。 5.前記酸性化が、 a)前記試験試料を遠心分離して、上清およびペレットを形成させる段階と、 b)前記上清を除去する段階と、および c)前記ペレットを酸性緩衝液中に懸濁させる段階と を含む、請求の範囲第1項に記載の方法。 6.前記酸性緩衝液が、100mMと2Mの間の酢酸ナトリウムを含み、3.0 と4.5の間のpHを有する請求の範囲第5項に記載の方法。 7.a)前記試料を塩基性溶液で処理し、それによって塩基性の試験試料を形成 させる段階と、および b)前記試験試料中に含有されるいずれもの標的配列を、核酸増幅に適した緩 衝液に置く段階と を含む核酸増幅用の試験試料を調製する方法において、 改良点が、段階a)の後でありかつ段階b)の前に、前記塩基性の試験試料を 酸性化する段階を含む方法。 8.前記試験試料が臨床試験試料である請求の範囲第7項に記載の方法。 9.前記方法が、段階b)の前に前記塩基性溶液をリン酸塩含有溶液で処理する 段階をさらに含む、請求の範囲第8項に記載の方法。 10.前記酸性化が、 a)前記塩基性試験試料を遠心分離して、上清およびペレットを形成させる段 階と、 b)前記上清を除去する段階と、および c)前記ペレットを酸性緩衝液に懸濁させる段階と を含む、請求の範囲第7項に記載の方法。
