Claims (1)
- 【特許請求の範囲】 1.一次非げっ歯類細胞を形質転換するための方法であって、下記の工程:p 53をコードする核酸を、p53の発現を指令することのできる遺伝子ベクターの中 にメタロチオネインプロモーターのコントロール下となるように配置する; 当該遺伝子ベクターを一次非げっ歯類細胞に導入する;そして1日当り約0.6 〜約1.5集団倍化の集団倍化時間を有する細胞フォーカスを選択する、ここで当 該細胞は逆転写酵素陰性であり、且つ非腫瘍原性である; を含んで成る方法。 2.前記一次非げっ歯類細胞が鳥類に由来する、請求項1記載の方法。 3.前記一次非げっ歯類細胞がヒトに由来する、請求項1記載の方法。 4.前記細胞が皮膚細胞である、請求項1記載の方法。 5.前記細胞が胸筋細胞である、請求項1記載の方法。 6.前記細胞が心筋細胞である、請求項1記載の方法。 7.メタロチオネインプロモータ−のコントロール下でp53の発現を指令する ことのできる遺伝子ベクターを含む不死化細胞。 8.前記細胞が繊維芽細胞である、請求項7記載の細胞。 9.前記細胞が鳥類に由来する、請求項7記載の細胞。 10.ウィルスを繁殖するための方法であって、下記の工程: 少なくとも一種の感染性ウィルス粒子を少なくとも一種の不死化細胞培養物と 接触させる、ここで当該培養物の細胞はメタロチオネインプロモーターのコント ロール下でp53を発現する遺伝子ベクターを含む;そして 当該細胞により産生されるウィルスを回収する; ことを含んで成る方法。 11.前記ウィルスがレオウィルスである、請求項10記載の方法。 12.前記ウィルスがHVTである、請求項10記載の方法。 13.前記ウィルスがフォウル・ポックス・ウィルスである、請求項10記載の方 法。 14.ウィルスを繁殖するための方法であって、下記の工程: 少なくとも一種の感染性ウィルス粒子を一次細胞と接触させる; 当該一次細胞を細胞培養物においてメタロチオネインプロモーターのコントロ ール下でp53を発現する遺伝ベクターを含む不死化細胞と一緒に増殖させる;そ して 当該細胞培養物から産生されたウィルスを回収する、 ことを含んで成る方法。 15.不死化非形質転換細胞であって、メタロチオネインプロモーターのコント ロール下にあるp53を含み、且つ当該細胞中での組換タンパク質の発現を指令す ることのできる少なくとも一種のベクターを含む細胞。 16.組換タンパク質を発現する請求項15記載の細胞。 17.前記ベクターが組換ウィルスの少なくとも一部をコードする、請求項16記 載の細胞。 18.前記ベクターがレトロウィルスベクターである、請求項15記載の細胞。
