Abstract
【解決手段】 本発明は、トレハロースによる澱粉の老化防止方法、微生物汚染防止方法、安定化方法、品質改良方法、揮散防止方法、日持ちを向上又は商品寿命を延長させる方法と、それらの方法により得られるトレハロース含有組成物を確立することにより前記課題を解決する。

Description
実験に用いた非還元性糖質は、砂糖、結晶性無水トレハロース、非晶質無水トレハロース、トレハロース含水結晶、非晶質無水ネオトレハロース、ネオトレハロース含水結晶、非晶質無水エルロース、エルロース含水結晶、非晶質無水ラフィノース、ラフィノース含水結晶、非晶質無水メレチトースおよびメレチトース含水結晶である。砂糖、トレハロース含水結晶、エルロース含水結晶、ラフィノース含水結晶およびメレチトース含水結晶は、市販の試薬を用いた。結晶性無水トレハロース,非晶質無水トレハロースは、それぞれ参考例1、参考例3の方法で調製した。ネオトレハロース含水結晶は、特開平4−179490号公報実験2に開示される方法で調製した。非晶質無水ネオトレハロース、非晶質無水エルロース、非晶質無水ラフィノースおよび非晶質無水メレチトースは、それぞれ含水結晶を水に溶解した後、室温下で真空乾燥して調製した。
無水トレハロースとして結晶性無水トレハロースおよび非晶質無水トレハロースを用いて、その脱水作用、すなわち、脱水力の強さと脱水量について詳細に実験した。対照としてトレハロース含水結晶を用いた。実験は、実験1で述べた方法で調製したそれぞれのトレハロースを粒径約100乃至150μmの粉末品とし、直径5cmのプラスチックシャーレにそれぞれ1gずつ採り、相対湿度70%に調整された25℃の雰囲気に放置し、経時的にこれらの水分(%)を測定した。結果は表2に示す。
もち粉4重量部を水6重量部に溶いて、木枠に 濡れ布きんを敷いたものに流し込み、これを105℃で10分間蒸して糊化澱粉とする。これにトレハロース含水結晶または参考例1の方法で調製した結晶性無水トレハロース7重量部をミキサーで混和し、均一になったら、更に水飴2重量部を加え十分に捏ねて成形し、更に40℃の温風で2時間軽く乾燥して求肥を得た。
トレハロース含水結晶を少量の水で加熱溶解し、これを蒸発釜にとり、減圧下で煮詰め、水分5.0%のシラップとした。次いで助晶機に移し、これに種晶として結晶性無水トレハロースをシラップ固形物当たり1%加え、120℃で5分間攪拌助晶し、次いで、アルミ製バットに取り出し、100℃で6時間晶出熟成させてブロックを調製した。
高純度トレハロース水溶液を蒸発釜にとり、減圧下で煮詰め、水分4.0%のシラップとし、これに、種晶として結晶性無水トレハロースをシラップ固形物当たり5%加え95℃で攪拌助晶し、次いで押出造粒機で造粒、熟成乾燥し、粉砕機にて粉砕して、水分0.53%の結晶性無水トレハロース粉末を、原料に対して約95%の収率で得た。
トレハロース含水結晶を室温にて、水に完全に溶解した後、60℃で24時間真空乾燥を行う。得られた乾燥物を粉砕機にて粉砕し、水分0.83%の非晶質無水トレハロース粉末を、原料のトレハロース含水結晶に対して約89%の収率で得た。得られた非晶質トレハロースの粉末X線回折図形を図3に示す。
参考例1の方法で得た結晶性無水トレハロース粉末を用いて、紙製の透湿性小袋に20gずつ充填し、脱水剤を製造した。本品は、味付け海苔、クッキーなどの乾燥食品を封入した防湿容器内雰囲気の脱水剤として有利に利用できる。また、各種乾燥食品、油性食品などに対し、本脱水剤とともに脱酸素剤などを併用して、これら食品などを安定保存することも有利に実施できる。
餅粉4重量部を水6重量部で溶いて、木枠に濡れ布きんを敷いたものに流し込み、これを100℃で20分間蒸した後、参考例1の方法で得た結晶性無水トレハロース粉末7重量部および砂糖1重量部を捏り込み、次いで水飴2重量部を加えて、充分に捏ねた後に成形し、更に、室内に16時間放置して、本品の表層において無水トレハロースをトレハロース含水結晶に変換させ、これを軽くロール掛けして表面をひび割れさせ、そぼろ風の求肥を得た。本品は、風味良好で、微生物汚染を受けにくく、高品質を長期間に亙って維持した。
さつまいもを厚さ約1cmにスライスし、これを蒸した後放冷し、これに参考例1の方法で得た結晶性無水トレハロース粉末をまぶしトレハロース含水結晶に変換せしめて脱水し、表面にトレハロース含水結晶の付着したいも菓子を製造した。本品は風味良好で安定ないも菓子である。
ブランデイー2重量部にプルラン10重量部を溶解し、これに参考例2の方法で得た結晶性無水トレハロース粉末8重量部を混合した後バットに移し、2日間放置してトレハロース含水結晶に変換させブロックを調製した。本ブロックを切削機にかけて粉末化し、分級して風味良好な粉末ブランディーを得た。本品を口に含めば、適度の甘味を有し、ブランディー香の充分な粉末香料である。本品は紅茶用香り付けとして、また、プレミックス、キャンデイー類などの製菓材料などとして有利に利用できる。また、本粉末を顆粒成形機、打錠機にかけて成型し、顆粒、錠剤として利用するこも有利に実施できる。
赤味噌2重量部に参考例2の方法で得た結晶性無水トレハロース粉末5重量部を混合し、多数の半球状凹部を設けた金属板に流し込み、これを室温下で一夜静置して固化し、離型して1個当り約4gの固形味噌を得、これを粉砕機にかけて粉末味噌を得た。本品は、即席ラーメン、即席吸物などの調味料として有利に利用できる。また、固形味噌は、固形調味料としてだけでなく味噌菓子などとしても利用できる。
参考例3の方法で得た非晶質無水トレハロース粉末4重量部および市販のトレハロース含水結晶0.02重量部をコンベア上で流動させつつ、これに対して薄口醤油を1重量部の割合になるように噴霧し、次いで熟成塔に移し、30℃で一夜放置して無水トレハロースをトレハロース含水結晶に変換せしめて粉末醤油を得た。本品は、即席ラーメン、即席吸物などの調味料として有利に利用できる。
生卵から調製した卵黄を、プレート式加熱殺菌機で60乃至64℃で殺菌し、得られる液状卵黄1重量部に対して、参考例1の方法で得た結晶性無水トレハロース粉末4重量部の割合で混合した後、実施例4と同様にブロック化し粉末化して粉末卵黄を得た。本品は、プレミックス、冷菓、乳化剤などの製菓用材料としてのみならず、経口流動食、経管流動食などの離乳食、治療用栄養剤などとしても有利に利用できる。また美肌剤、育毛剤などとしても有効に利用できる。
プレーンヨーグルト2重量部に参考例1の方法で得た結晶性無水トレハロース粉末8重量部を混合した後、実施例4と同様にブロック化し、粉末化して粉末ヨーグルトを得た。本品は、風味良好であるだけでなく、乳酸菌を生きたまま長期に安定化し得る。また、プレミックス、冷菓、乳化剤などの製菓用材料としてのみならず、経口流動食、経管流動食などの離乳食、治療用栄養剤などとして、更には、例えば、マーガマーガリン、ホィップクリーム、スプレッド、チーズケーキ、ゼリーなどに含有せしめヨーグルト風味の製品にするなどに有利に利用できる。さらに本粉末を顆粒成型機、打錠機などで成型して乳酸菌製剤とし、整腸剤などとして利用することも有利に実施できる。
小麦粉200重量部に実施例7の方法で得られた粉末卵黄60重量部、バター25重量部、砂糖10重量部、ベーキングパウダー12重量部および食塩0.5重量部を配合してホットケーキミックスを得た。本品は、水、牛乳などで溶いて焼くことにより、簡単に風味良好なホットケーキを調製することができる。
薬用人参エキス0.5重量部に参考例2の方法で得た結晶性無水トレハロース粉末1.5重量部を混捏した後、実施例4と同様にブロック化し、粉末化して粉末薬用人参エキスを得た。本品を適量のビタミンB1およびビタミンB2粉末とともに顆粒成型機にかけ、ビタミン含有顆粒状薬用人参エキスとした。本品は、疲労回復剤、強壮、強精剤などとして有利に利用できる。また、育毛剤などとしても利用できる。
参考例2の方法で得た結晶性無水トレハロース粉末500重量部、実施例7の方法で得られた粉末卵黄270重量部、脱脂粉乳209重量部、塩化ナトリウム4.4重量部、塩化カリウム1.85重量部、チアミン0.01重量部、L−アスコルビン酸ナトリウム0.1重量部、ビタミンEアセテート0.6重量部およびニコチン酸アミド0.04重量部からなる配合物を調製し、この配合物25gずつを防湿性ラミネート小袋に充填し、ヒートシールして流動食用固形製剤を製造した。
新生児のハムスターに、ウサギから公知の方法で調製した抗血清を注射して、ハムスターの免疫反応を弱めた後、その皮下にBALL−1細胞を移植し、その後通常の方法で3週間飼育した。皮下に生じた腫瘤を摘出して細断し、生理食塩水中で分散させてほぐした。得られた細胞を血清無添加のPRMI1640培地(pH7.2)で洗浄し、同培地に約2×106個/mlになるように懸濁し、35℃に保った。
ヒト由来のリンパ芽級細胞BALL−1細胞を牛胎児血清を20%補足したEagleの最少基本培地(pH7.4)に接種し、37℃で常法に従い生体外(in vitro)で浮遊培養した。得られた細胞を血清無添加のEagle最少基本培地で(pH7.4)で洗浄し、同培地に約1×107個/mlになるように懸濁した。この懸濁液にセンダイウイルスをml当り約1,000赤血球凝集価添加し、38℃で1日保ってツモア・ネクロシス・ファクター−αを誘導生成させた。これを4℃、約1,000gで遠心分離し、得られた上清をpH7.2、0.01Mリン酸塩緩衝液を含有する生理食塩水で15時間透析し、更に精密濾過して得た濾液を常法に従って、抗インターフェロン抗体のカラムに流し、その非吸着画分を、抗ツモア・ネクロシス・ファクター−α モノクローナル抗体のゲルカラムを用いたアフィニティクロマトグラフィーにより精製し、濃縮して、濃度約0.01w/v%比活性約2×106JRU/mg蛋白質の濃縮液を得た。活性収量は、誘導生成時の懸濁液1L当り約5×104JRUであった。
参考例2の方法で得た結晶性無水トレハロース450重量部にヨウ素3重量部を溶解しメタノール50重量部加えて混合し、更に10%プルラン水溶液200重量部およびマルトース含水結晶50重量部を加えて混合し、室温下で一夜放置してトレハロース含水結晶に変換させ、適度の延び、付着性を示す外傷治療用膏薬を得た。

- トレハロースを含有せしめることを特徴とする香料又は香気成分の揮散防止方法。
- トレハロースを含有せしめ、次いで、真空包装、ガス充填包装、又は、冷蔵することを特徴とする食品、医薬品、化粧品、又は、工業化学品の日持ちを向上、又は、その商品寿命を延長させる方法。
- 請求項1乃至6のいずれかに記載の方法により得ることのできるトレハロース含有組成物。
