本発明は、架橋セルロースエーテルを用いた崩壊剤に関するものである。特に、医薬、食品、トイレタリー、飼料等の錠剤、顆粒等の成形剤において、崩壊剤と基質薬剤乃至有効成分を混合して成形することにより、これら成形剤が水に投入された場合、崩壊剤が吸水し、膨潤することにより速やかに成形剤が崩壊し、有用な薬剤乃至有効成分が水中に容易に分散されるようにした成形剤に使用される崩壊剤に関するものである。

【従来の技術】
医薬品添加物辞典(1995追補版、日本医薬品添加物協会編集、薬事日報社発行)によれば、現在医薬品添加剤の中で崩壊剤として収載されているものは71種類ある。この中で汎用的に使用されているものとして、低置換度ヒドロキシプロピルセルロース、カルメロースカルシウム(カルボキシルメチルセルロースカルシウム)、クロスカルメロースナトリウム(架橋カルボキシルメチルセルロースナトリウム)、カルボキシメチルスターチナトリウム、部分アルファー化デンプン、クロスポビドン(架橋ポリビニルピロリドン)等が挙げられる。これらの物質は、通常、中性の水には溶解しないが膨潤する。この特性を利用して医薬品、トイレタリー、農薬等の錠剤、顆粒等の崩壊剤に用いられる。更に詳細な医薬用途使用例を挙げれば、有効薬剤に加え、結合剤、滑沢剤、色素、フィラー等に崩壊剤を混合し、加圧下錠剤を成形する。その錠剤を人が服用すると、消化器官中の水と接触し、崩壊剤が水を吸収して崩壊するため、その他の基材をバラバラに壊し、有効成分を速やかに吸収せしめる。

一方、この崩壊剤が使用されないときは、錠剤の崩壊は極めて緩やかであるか又は崩壊はせずに錠剤の成分が溶解するのを待つしかない。しかし、薬剤には難溶性の物質も多数存在し、実質上崩壊剤の助け無しには有効に薬剤を使用できない。従って、崩壊剤は、医薬用途には極めて一般的に使用されている。

医薬品以外の分野としては、農薬用製剤がある。例えば、水田に農薬を散布する場合、小規模水田では、航空散布や噴霧機械が使用できず、人の手で散布する必要がある。こうした場合、微粉農薬は風による飛散を避けて顆粒状にすることが一般的であるが、一旦散布した顆粒状農薬は細かく分散しづらく、時間と共に流出してしまう。また、苗の消毒においては同じく顆粒が使用されるが、分散しないために、田植え作業を通して実際に苗に付着する有効成分の割合は低い。従って、この用途でも崩壊剤は広く使用されている。

更に、トイレタリー分野において、崩壊剤が使用されていない顆粒状洗剤が水に溶け残ることは屡々観察され、崩壊剤適用の一例となる。また、釣り用の撒き餌に崩壊剤を使用し、細かく分散させることにより、少量の魚誘因餌で大きな体積をカバーすることも考えられる。

このように多くの用途を持つ崩壊剤であるが、先に挙げた既存の崩壊剤はそれぞれ欠点を有している。

まず、カルメロースカルシウム(カルボキシルメチルセルロースカルシウム)、クロスカルメロースナトリウム(架橋カルボキシルメチルセルロースナトリウム)、カルボキシメチルスターチナトリウムは、セルロース系高分子の金属塩、つまりイオン性であるために薬物との安定性が悪い。具体的にいえばアスピリン(アセチルサリチル酸)と混合した場合、アスピリンの分解を助長し、サリチル酸の結晶が析出する場合やパンクレアチン(アミラーゼ活性酵素)の失活を起こす。クロスポビドン(架橋ポリビニルピロリドン)は薬剤との相互作用は少ないが、吸湿性が高いために、やはり湿気が多いところで保存した場合、アスピリンの分解を引き起こす場合がある。部分アルファー化デンプンは元々の膨潤率が低く、崩壊力が他の崩壊剤に較べ劣る。低置換度ヒドロキシプロピルセルロースに関しては、安定性は良好であるが崩壊力が若干低く、薬剤によっては添加量を多く使用しなければならない場合がある。

本発明は上記事情に鑑みてなされたものであり、既存の崩壊剤に比べ、膨潤率が高く、かつ薬物と安定である崩壊剤を提供することを目的とするものである。

【課題を解決するための手段及び発明の実施の形態】
本発明者らは、上記課題を解決するために種々検討した結果、セルロースエーテルを製造する工程において、エーテル基を導入する際に、併せて架橋反応することにより得られる架橋セルロースエーテルが優れた崩壊剤であることを知見し、本発明をなすに至ったものである。

即ち、本発明は、
(1)セルロースの水酸基の水素原子がアルキル基及び/又はヒドロキシアルキル基で置換されていると共に、ポリグリシジルエーテルにより架橋された架橋セルロースエーテルからなることを特徴とする崩壊剤、
(2)架橋セルロースエーテルが、ポリグリシジルエーテルによる架橋反応とアルキル基及び/又はヒドロキシアルキル基の置換導入反応が同じ反応工程で行われることにより得られたものであることを特徴とする(1)に記載の崩壊剤、
(3)架橋反応により置換されるグルコースユニット上の水酸基割合が、0.004〜0.06であることを特徴とする(1)又は(2)に記載の崩壊剤
を提供する。

以下、本発明につき更に詳しく説明する。
本発明の崩壊剤は、架橋セルロースエーテルからなるものであり、この場合、この架橋セルロースエーテルは、セルロースの水酸基の水素原子がアルキル基及び/又はヒドロキシアルキル基で置換されていると共に、ポリグリシジルエーテルにより架橋されているもので、これはセルロースエーテルを製造する際、セルロースをアルカリ金属水溶液と混合し、エーテル置換基を導入するためのアルキル化剤又はヒドロキシアルキル化剤と同時に架橋剤を添加して反応させることにより、実質上一段階で崩壊剤を製造することができる。

目的とする架橋セルロースエーテルを得ようとする場合、エーテル基を導入したセルロースエーテルを反応、精製、乾燥、また必要ならば粉砕といった別工程で製造し、それを用いて架橋反応を行うことは可能であるが、工程が複雑になる一方、反応、精製、乾燥、粉砕等の工程が重複するため、コスト的に全く見合わない。

例えば、架橋剤を使用してメチルセルロースやヒドロキシプロピルセルロースを架橋する試みは、米国特許第3,168,421号公報に見られるが、セルロースエーテルを水溶液にし、架橋剤と混ぜてから架橋する手法により不溶性フィルムを得るというものであり、本発明と異なる。

本発明者らは、いかに簡単な製造工程を採用することにより、架橋セルロースエーテルを安価に製造できるかを検討した結果、このエーテル基の導入と架橋反応とを実質上同一工程で行うことにより達成できることを見出した。この場合、好ましくはベースとなるセルロースを反応触媒となる水酸化アルカリ金属水溶液と混合した後、同一反応器内でこれらエーテル基導入と架橋の両反応を行うものである。更に詳しく述べれば、エーテル化反応及び架橋反応は、触媒を加えられたセルロースにポリグリシジルエーテルを加えて架橋反応をさせ、その後同じ反応器を用い、エーテル化剤を加えてエーテル化反応させてもよいし、エーテル化反応を終了させてから架橋剤を加えて架橋反応させてもよい。但し、最も短時間で両反応を終了させるには、エーテル化剤と架橋剤をほぼ同時期に加えて反応させるのが最も望ましい。

ここで、セルロースとしては、セルロース自体が天然に広く産出されるものなので、特に限定されないが、あえて例示するとすれば、市場で容易に入手できる綿、コットンリンター、木材を精製したバルクやシート、つまりパルプが挙げられる。

水酸化アルカリ金属水溶液は、例えば水酸化カリウム、水酸化リチウム等の水溶液が挙げられるが、コスト的には水酸化ナトリウムが有利である。

エーテル化剤の添加量は特に限定されないが、あまり少量すぎるとセルロースの結晶構造が十分壊れず、吸収性が低下するおそれがあるため、メチルセルロースを例にとれば、グルコースユニットに対するメトキシル基置換度(セルロースのグルコースユニット当りメトキシル基で置換された水酸基の平均個数(Degree of Substitution)、以下「DS」と略す)として1.0〜2.2程度が好ましい。また、ヒドロキシアルキルセルロースを例にとれば、ヒドロキシアルキル基置換度(セルロースのグルコースユニット当りに付加したヒドロキシアルキル基の平均モル数(Molar Substitution)、以下「MS」と略す)0.1〜1.0程度が好ましい。

ポリグリシジルエーテルとしては、例えばエチレングリコールジグリシジルエーテル、ポリエチレングリコールジグリシジルエーテル、プロピレングリコールジグリシジルエーテル、ポリプロピレングリコールジグリシジルエーテルが挙げられる。かかるポリグリシジルエーテルは、これら両末端のエポキシ官能基がセルロース中の水酸基又はヒドロキシアルキル基の水酸基と結合した場合、一方のエポキシ基のみ結合した場合のいずれの場合においても非イオン性であり、全体的に非イオン性であることは、崩壊剤として使用した場合に、薬剤との相互作用を起こしにくい。

このように、ポリグリシジルエーテルは2つのエポキシ基を持ち、セルロースの水酸基に導入されるが、必ずしも両エポキシ基が水酸基と反応するわけではなく、一方のみセルロースに導入される場合もあり得る。完全に2つのエポキシ基がセルロースと反応したと仮定したMS置換度は、0.004〜0.06、特に0.01〜0.04が好ましい。この範囲より少ないと架橋が十分でなく、セルロースエーテルは部分的に溶解し、粘性を発揮するため、水の浸透が妨げられ、有効に崩壊剤として機能しない場合がある。またこの範囲を超えると、架橋があまりに密であるため膨潤しにくく、またコスト的にも見合わない場合がある。
また、添加方法として、添加するポリグリシジルエーテル量が少ない場合は、エーテル化剤、水、反応を妨害しない有機溶媒に希釈して添加することもできる。

なお、エーテル化反応及び架橋反応の反応条件としては、それぞれ30〜120℃、特に40〜100℃で反応を行わせることが好ましい。反応時間は特に制限されないが、通常2〜8時間である。

本発明においては、このようにして得られる架橋セルロースエーテルを崩壊剤として使用するが、その使用方法は、従来の崩壊剤の場合と同様である。具体的には医薬、農薬、食品、トイレタリー、飼料等において、錠剤、顆粒等の成形剤を得る場合に、従来の崩壊剤に代えて又は従来の崩壊剤と共に使用される。

【実施例】
以下、実施例及び比較例を示し、本発明を具体的に説明するが、本発明は下記の実施例に制限されるものではない。なお、下記例で部は重量部を示す。

[実施例、比較例]
内容積5Lの内部撹拌型オートクレーブに250gのコットンリンターパルプを仕込み、−98kPaまで減圧した。次いで50重量%水酸化ナトリウム300gを添加し、10分間撹拌した。架橋剤としてエチレングリコールジグリシジルエーテル(分子量174.2g/mol)を合計30gとなるようアセトンで希釈して加え、更に塩化メチルを200g添加し、60〜70℃で2時間、さらに90℃まで昇温し、30分間温度を維持し、反応を完結させた。内部残ガスをパージした後、粗反応物を熱水で精製し、乾燥後、0.5mmの開口部を持つスクリーンを備えた粉砕機にて処理・整粒し、試料とした。

上記の手法により、架橋剤の添加量を変えて調製した6種類の試料の物性を下記表1に示す。なお、エチレングリコールジグリシジルエーテルのMS置換度は、完全に架橋した、つまり2つのエポキシ基が両方ともセルロースに結合したと仮定した場合の値である。

膨潤特性は、試料100mg、アルミナ400mgの混合粉末からなる錠剤を直径15mmφ、製錠圧力55MPaにてシリンダー内に調製し、導水管付きの上杵を錠剤に接するように配置し、導水して7分後の錠剤厚みから下記式により膨潤率を算出した。また、試験後の錠剤を取り出し、目視にて膨潤状態、水の浸透状態(水が錠剤下部まで浸透しているか否か)を観察した。結果を表1に示す。

250 g of cotton linter pulp was charged into an internal stirring type autoclave having an internal volume of 5 L, and the pressure was reduced to -98 kPa. Next, 300 g of 50% by weight sodium hydroxide was added and stirred for 10 minutes. As a crosslinking agent, ethylene glycol diglycidyl ether (molecular weight 174.2 g / mol) diluted with acetone to a total of 30 g was added, and further 200 g of methyl chloride was added, and the temperature was further raised to 90 ° C. at 60 to 70 ° C. for 2 hours. The swelling characteristics were as follows. A tablet consisting of a mixed powder of 100 mg of a sample and 400 mg of alumina was prepared in a cylinder with a diameter of 15 mm and a tableting pressure of 55 MPa, and an upper punch with a water pipe was placed so as to be in contact with the tablet. The swelling ratio was calculated from the tablet thickness after one minute by the following formula.
