Abstract
【解決手段】ヒト脂肪組織から採取した脂肪前駆細胞に対し、継代培養を繰り返すこと。

Description
また、本発明は、そのヒト脂肪前駆細胞株の利用方法を提供する。
以下に本明細書において特に使用される用語の定義を列挙する。
本実施例では、まず、本実験に対して同意を示したヒトから細胞を脂肪組織から調整した。脂肪吸引物を生理食塩水で十分に洗浄した。上層に脂肪吸引物、下層に生理食塩水が十分に分離したのを確認し、下層を捨て、肉眼で見て生理食塩水はほぼ透明になるまでこれを繰り返した。この実験例では、7回行った。脂肪組織を40mlと同量40mlの0.075%コラゲナーゼ/PBS(Gibco)を加え、37℃でよく攪拌しながら1時間インキュベートした。この試料に、同量の10%血清加DMEMを加え、1200×gで10分間遠心分離した。
脂肪前駆細胞を5×103cells/cm2の濃度で細胞培養用プレートあるいはディッシュにまく。4日から6日たつとほぼ一面にはえひろがる。そこで、細胞を0.25%トリプシン(1mM EDTA含有)溶液を用いて回収する。回収した細胞数を数えた後、再び5×103cells/cm2の濃度でまく。この操作を繰り返す。
不死化したと考えられる細胞群を1穴に細胞が0.5個入ると考えられる濃度に希釈し、96穴プレートにまく。2週間ほど後に増殖が観察された穴から細胞を回収する。それぞれのクローンについて、その脂肪細胞への分化能を確かめ、脂肪細胞へと分化するクローンを選び出した。
コンフルエントまで増殖させた脂肪前駆細胞を脂肪細胞分化用培地で2日おきに処理した。分化開始日と2日目には、0.5mM IBMX、1μMデキサメタゾン、1μMトリグリタゾン、5μg/mlインスリンを含んだ10%CCS−DMEM培地で処理した。4日目には、トリグリタゾンとインスリンを含んだ10%CCS−DMEM培地で、6日目以降は、500ng/mlインスリンを含んだ10%CCS−DMEMで処理した。分化の割合は、分化誘導後10日目以降に観察した。図2に結果を示す。
2)再生医療分野では、現在汎用されているマウス由来フィーダー細胞(NIH3T3細胞やJ2細胞)に取って代わる可能性がある。すなわち、異種移植でなく同種移植となることができる。
3)本細胞は脂肪幹細胞として多系統に分化しうるものであり、再生医療の臓器或いは細胞ソースになる可能性がある。
Claims (6)
- 脂肪細胞に分化されやすいことを特徴とするヒト脂肪前駆細胞株。
- 細胞が不死化されたものである、請求項1記載のヒト脂肪前駆細胞株。
- 脂肪幹細胞、線維芽細胞、及びフィーダー細胞として利用できる特徴を有する、請求項1、2いずれか1項記載のヒト脂肪前駆細胞株。
- ヒト脂肪組織から採取した脂肪前駆細胞に対し、継代培養を繰り返すことを特徴とするヒト脂肪前駆細胞株製造方法。
- 請求項1〜3のヒト脂肪前駆細胞株を利用した医薬品スクリーニング方法。
- 請求項1〜3のヒト脂肪前駆細胞株を利用した食品、或いは食品成分スクリーニング方法。
