Abstract
Description
The present invention relates to a whole blood platelet aggregating ability measuring method for measuring platelet aggregating ability using whole blood without using platelet-rich plasma separated from blood, its device, and aspiration used in a whole blood platelet aggregating ability measuring device. This relates to the nozzle.
Conventionally, the light transmission method is mainly used for the measurement of platelet aggregation ability. FIG. 8 shows the configuration of the platelet aggregating ability measuring apparatus, and 1 shows a photoelectric aggregation degree measuring means. A
In the measurement of platelet aggregation ability by the light transmission method, it is necessary to prepare PRP and PPP from whole blood. First, sodium citrate is added to the collected whole blood to obtain anticoagulated blood, and the blood is centrifuged. The erythrocyte concentrate and PRP are separated, and the erythrocyte concentrate is further centrifuged to separate erythrocytes and PPP. However, PRP contains some white blood cells, red blood cells, etc., and has a problem of affecting the transmitted light, and it is necessary to separate the PRP and PPP from the whole blood sample collected during the platelet aggregation test. There was such a trouble. Moreover, this separation operation has been considered to have some adverse effects on the platelet aggregation ability test. Further, the conventional example has a drawback in that the aggregation rate curve is created after creating the relationship between the aggregation rate of PRP at each concentration of the aggregation-inducing agent and time, and the creation procedure is complicated.
The present invention has been made in view of the problems as described above, and eliminates the complicated preprocessing in the platelet agglutination ability test, and automatically processes the platelet agglutination ability test result to determine the agglutination ability.
Next, with reference to FIGS. 1-7, the platelet aggregation ability measurement by the whole blood of this embodiment is demonstrated. First, whole blood is placed in four
By operating the measurement start button of the
Subsequently, the
When the suction pressure of the sample in the
In this embodiment, in order to find the lowest concentration of ADP that is estimated that platelets caused by platelets cause secondary aggregation, whole blood to which ADP having a different concentration is added is prepared, and the whole blood is subjected to agglutination reaction. The agglutination curve is prepared by measuring the suction pressure according to the clogging state of the
Next, the platelet agglutination ability test form will be described in detail with reference to FIG. This inspection form is provided with printing areas R1 to R5, and R1 is printed with measurement date, measurement time, measurement mode (ANALY 1), agglutination reaction time (minutes and seconds), and measurement number (specimen number). Then, the drug name, unit (ADP, μM), and reagent concentration (0.5, 1.0, 2.0, 4.0 (μM)) of the agglutination reagent (platelet aggregation inducer) are printed. A measurement result is shown in the print region R2, a platelet aggregation threshold coefficient (PATI coefficient) is shown, and a grade of aggregation ability is shown. Grade 1 (normal) is printed on this inspection form. PATI indicates the minimum concentration of a platelet aggregation inducer that induces secondary aggregation, and is the value of the reagent concentration when the suction pressure is 50%, and this value is used as the platelet aggregation threshold coefficient.
III ,II,I ,-I,-II が表示され、I を三角印で指し示している。
A graph is shown in the printing region R3, the horizontal axis indicates 0 to 4 channels (reagent concentration), the vertical axis indicates the suction pressure (0 to 100%), and the range of the coagulation ability grade is indicated by diagonal lines. The agglomeration curve α is illustrated, and the measurement results are indicated by triangles in the grade type (GT) column. In the print region R4, III, II, I, -I, and -II are displayed as GT, and I is indicated by a triangle mark. In the printing region R5, the suction pressure when whole blood is not sucked is shown in 0ch, and the concentration of ADP (0.5 μM, 1.0 μM, 2.0 μM, 4.0 μM) is shown in 1-4 channels (ch). In the measurement result of this inspection form, -3.1%, 0.7%, 3.0%, 73.5%, and 100.0% are displayed. In the percentage of the printing area R5, 4ch is 100.0%, and the result of this suction pressure indicates that the
The aggregation curve α is a straight line between the channels, and these straight lines are represented by linear functions. Therefore, a PATI value, that is, a value obtained by substituting a known 50% suction pressure value into the linear function of the straight line. Is a PATI value coefficient, and the PATI value coefficient can be calculated by a simple arithmetic expression. Which range the PATI value coefficient belongs to can be easily obtained from a comparison value indicating a grade range. Incidentally, the comparison values are, for example, grade III is −1 to 0 μM, grade II is 0 to 1 μM, grade I is 1 to 2 μM,
Further, as shown in FIG. 7, the
