Abstract
【解決手段】Ca/Pのモル比が1.67より大きく、ヒドロキシアパタイト構造のBおよびA部位において置換されているCO3 2-イオンを5重量%までの量で含んで成り、該CO3 2-イオンの少なくとも50%がB部位において置換され、Na+またはNH4 +イオンを含有しない、単相カーボネート置換ヒドロキシアパタイト組成物。

Description
(i)H+イオンおよびCa2+以外のカチオンを含まず、CO3 2-およびPO4 3-イオンを含有する水溶液を調製し;
(ii)工程(i)からの溶液を、カルシウム化合物の水溶液または懸濁液と混合し;
(iii)工程(ii)において形成された沈殿物を、回収し、乾燥する
工程を含み、カルシウム含有溶液または懸濁液および燐含有溶液を混合した際に、Ca/P比を1.67を超える値に保持する方法により、製造することができる。
混合した際に、カルシウム含有溶液または懸濁液、および燐含有溶液におけるCa/P比を1.67より高く保持して、AおよびB部位の両方における置換を促進して、式:
実施例1〜5
38.25gのCa(OH)2(AnalaR、BDH)を、1リットルの脱イオン水に分散させて、水酸化カルシウムの懸濁液を調製した。次の反応の前に、この懸濁液を15分間攪拌して溶液Aを生成した。
本発明によって製造されるカーボネート置換ヒドロキシアパタイトは、750℃〜1050℃の所定温度について、未置換ヒドロキシアパタイトより高い密度に焼結する。図3は、未置換ヒドロキシアパタイトおよび実施例3において製造されるカーボネート置換ヒドロキシアパタイトの焼結密度に対する焼結温度の効果を示す(焼結条件は以下の通りである:カーボネート置換ヒドロキシアパタイトについては、方法において記載されるCO2/H2Oの雰囲気、および、未置換ヒドロキシアパタイトについては、空気雰囲気。両方の物質について、所定温度まで2.5℃/分の加熱速度、2時間の滞留時間、および室温まで10℃/分の冷却速度)。ヒドロキシアパタイト格子へのカーボネートイオンの置換は、焼結により、一般にセラミックに関して低いと考えられる温度において高い密度を有するセラミックを形成する粉末を生じる。
ヒト血漿に存在するイオンの種類および濃度を含有するSimulated Body Fluid(SBF)中でセラミック試験片を試験することによって、本発明によって製造されるカーボネート置換ヒドロキシアパタイトの生物活性を、未置換ヒドロキシアパタイトと比較して評価した。この試験は、新しい骨様アパタイト層が、セラミック試験片の表面に沈積するのに要する時間を示し、その時間が短いほど、その物質はより高い生物活性を有する。本発明によって製造されたカーボネート置換ヒドロキシアパタイト(実施例3)は、7日未満で新しい骨様アパタイトの形成を生じ、一方、未置換ヒドロキシアパタイトは24〜28日を要とした。この試験は、本発明によって製造されるカーボネート置換ヒドロキシアパタイトが、未置換ヒドロキシアパタイトと比較して、向上した生物学的特性を有することを早期に示すものである。
Claims (3)
- 単相カーボネート置換ヒドロキシアパタイト組成物であって、Ca/Pのモル比が1.67より大きく、ヒドロキシアパタイト構造のBおよびA部位において置換されているCO3 2-イオンを5重量%までの量で含んで成り、該CO3 2-イオンの少なくとも50%がB部位において置換され、Na+またはNH4 +イオンを含有しない組成物。
- ヒドロキシアパタイト構造のB部位およびA部位において置換されているCO3 2-イオンを3〜5重量%で含んで成る請求項1に記載の単相カーボネート置換ヒドロキシアパタイト。
- 該CO3 2-イオンの50〜85%がB部位において置換されている請求項1または2に記載の単相カーボネート置換ヒドロキシアパタイト。
