Abstract
【解決手段】エタノール含有試料を、酸素、水及びアルコールオキシダーゼの存在下に酵素反応させることにより過酸化水素を発生させ、該過酸化水素と、4−アミノアンチピリン、フェノールを、ペルオキシダーゼの存在下に酵素反応させることにより赤色キノン色素を発色させるエタノール測定方法及び上述したエタノール測定方法をより簡易、迅速に実施するためのエタノール測定キットにより解決する。

Description
(A液)アルコールオキシダーゼ、ペルオキシダーゼ及び4−アミノアンチピリンを含有する溶液
(B液)リン酸緩衝液及びフェノールを含有する溶液
以下の組成からなるA液及びB液を調製した。なお、調製後は5℃で保存した。
酵素力価1000U/mlのアルコールオキシダーゼ(Pichea pastoris由来、SIGUMA社製)900μl、酵素力価50,000U/gのペルオキシダーゼ(わさび由来、オリエンタル酵母社製)1mg、4−アミノアンチピリン1mgを1L容メスフラスコに入れ、蒸留水で1Lになるように定容することにより、A液を調製した。
リン酸二水素ナトリウム(二水和物)(Sodium Dihydrogen Phosphate, NaH2PO4・2H2O, M.W 156.01(国産化学株式会社製))10.95gを1L容メスフラスコに定容し70mmolリン酸二水素ナトリウム溶液を調製した。また、リン酸水素二ナトリウム(12水和物)(di- sodium Hydorogen Phosphate, Na2HPO4・12H2O, M.W 358.14(国産化学株式会社製))25.0gを1L容メスフラスコに定容し70mmolリン酸水素二ナトリウム溶液を調製した。そして、リン酸二水素ナトリウム溶液:リン酸水素二ナトリウム溶液を4:6の割合で混合しpHを7.1に調整し、70mmolリン酸緩衝液(pH7.1)を得た。
使用前にA液500μLとB液500μLをエッペンドルフチューブに入れて混合した。その際、空気中に揮発しているエタノールの混入に留意した。これに、エタノール含有試料としてエタノール溶液を調製したものをサンプルとして約20μl採取し、A液とB液の混合液が入ったエッペンドルフチューブに添加した。そして、数秒後、発色したサンプルの吸光度(515nm)を吸光度計(日本分光社製)で測定し、相関関数を算出した。結果を図2に示す。
Claims (7)
- エタノール含有試料を、酸素、水及びアルコールオキシダーゼの存在下に酵素反応させることにより過酸化水素を発生させ、
該過酸化水素と、4−アミノアンチピリン及びフェノールを、ペルオキシダーゼの存在下に酵素反応させることにより赤色キノン色素を発色させる、
エタノール測定方法。 - 前記エタノール含有試料を酵素反応させる前に、界面活性剤を添加する、請求項1に記載のエタノール測定方法。
- 前記界面活性剤が、ポリオキシエチレンソルビタン脂肪酸エステル類である、請求項2に記載のエタノール測定方法。
- 下記A液及びB液を備えた、エタノール測定用キット。
(A液)アルコールオキシダーゼ、ペルオキシダーゼ及び4−アミノアンチピリンを含有する溶液
(B液)リン酸緩衝液及びフェノールを含有する溶液 - 前記A液及び/又は前記B液が、界面活性剤を含有する、請求項4に記載のエタノール測定用キット。
- 前記界面活性剤が、ポリオキシエチレンソルビタン脂肪酸エステル類である、請求項5に記載のエタノール測定用キット。
- さらに、エタノール濃度に応じて色分けされた比色表を備えた、請求項4〜6のいずれか1項に記載のエタノール測定用キット。
