この結果、Duro-Tak87-2677 とMASCOS10Cとは自己架橋性があるが、Duro-Tak87-235AとDuro-Tak387-2287とは自己架橋性がないことが判明した。

リバスチグミン、粘着剤、固化剤を含むリバスチグミン経皮吸収製剤を作成しボランティアに貼付して粘着性を評価した。
リバスチグミン経皮吸収製剤は、効果を上げるため大量のリバスチグミン遊離塩基を含ませる必要があり、そのため製剤が柔らかくなり皮膚貼付後の糊残りが課題であるので、この評価を実施した。

製剤構成成分1覧及び評価結果を第2表に示す。構成成分の単位は重量%である。また、10重量%以上の構成成分について、小数点以下は4捨5入した。

C: 製剤は固く粘着性が弱いので皮膚に貼付する時皮膚から剥がれおちることがある。製剤としては不適である。
これらの結果より、自己架橋型粘着剤を選択し、適切な量の固化剤を使用することにより好適な経皮吸収製剤が得られると結論できる。

From these results, it can be concluded that a suitable transdermally absorbable preparation can be obtained by selecting a self-crosslinking adhesive and using an appropriate amount of a solidifying agent.

3.透過特性
全層ヒト皮膚を用い、リバスチグミンの経皮透過を次のようなインビトロ透過試験で検査した。
試作リバスチグミンパッチ(実施例1組成 , 面積=1.3 cm2)を貼付したヒト皮膚を改良フランツ型セルにセットし、経皮吸収試験自動サンプリング装置(TransViewC12、コスメディ製薬製)で経時的にサンプリングを行った。PEG400:H2O(1:3 w/w) 混合液5.75 mlをレセプター媒体として使用した。試験温度は、水浴によりインビボヒト皮膚の温度に対応する32℃に調節した。対象としてリバスチグミンの市販経皮吸収製剤である市販品(Exelon Patch)を用いた。レセプター媒体中のリバスチグミン濃度をHPLCで測定した。透過試験の結果は図1にグラフ的に示す。遊離塩基で存在するリバスチグミンのヒト皮膚を通る浸透量は、リバスチグミンの含有率を30.0%に設定した時、市販品と同等あるいはそれ以上の透過性を示した。
図1において、三角印(▲)は実施例1の経皮吸収製剤の結果である。また四角印(■)は市販品(Exelon Patch)を同一条件で測定した結果である。

比較例9においてはリバスチグミン濃度が高すぎて粘着物性が破たんし泥状となっており薬物透過量試験は実施しなかった。
リバスチグミンの濃度と経皮透過性は相関しており濃度20%以下では薬物透過量が小さく、実用性に劣る。また濃度が50%以上では粘着剤系が極めて柔らかくなり経皮吸収製剤としての実用性が問題となる。この表の結果より、リバスチグミンの粘着剤層中の量(重量%)は10〜50%が好ましく、20〜45%がより好ましいと結論できる。
