Description
タンパク質4重量%、pH5.89の乳清タンパク質分離物(Prolacta 90)の分散液を85℃/15分で熱処理し、精密濾過により総固形分22重量%まで濃縮し、噴霧乾燥して、乳清タンパク質ミセル粉末(WPM)を調製した。
−表面電荷
粒子の電気泳動移動度に対応する表面電荷(ζ電位)を粒子移動度分布計(Zetasizer Nanoseries、Malvern,UK)を用いて測定した。1M HCl及びNaOHの滴定液を用いる多目的滴定装置(MPT2、Malvern)を使用して、増分0.5及びpH精度目標0.3で、pHを8から2まで変化させた。セルDTS1060Cを使用し、25℃で測定を行った。15mLの0.1重量%溶液を用いた。データ処理は自動で行った。
マスターサイザーSロングベンチ(Malvern,UK)による多角度静的光散乱を使用して、粒径分布を測定した。分散相の屈折率1.36、連続相の屈折率1.33及び後方散乱係数0.1(3JHD表示)を計算に使用した。残りの値は常に1.5未満であった。使用した分散相の屈折率及び数学モデル(粒子は球状であると仮定)の任意選択を考慮すると、本測定値は、粒径の定量的測定ではなく、系の凝集の定性的指標のみを提供する。
I.WPM/ペクチン静電的複合体の形成を可能にするpH条件の特定
WPM及びペクチンの表面電荷(ζ電位)をpHとの関数として図1に示す。pHが2から8に上がるにつれて、ペクチンのζ電位は、中性から−45mVに下がった。この変化は、ペクチン主鎖上のカルボキシル基に関係する可能性があり、低いpHでは、これらの基が中和されて、ζ電位値がゼロに近づいた。WPMの場合、ζ電位は、pH2の20mVからpH3.8の40mVまで変化し、pH8で−45mVまで減少し、pH4.6で電気的中性が測定された。後者は、WPMの主要構成タンパク質であるβ−ラクトグロブリンの等電点に関係する可能性がある。
WPMへのペクチンの添加によって生じる変化を評価するために、粒径分布を測定した。図2及び3は、WPM:ペクチンの重量比が10:1〜1:1に相当する、1重量%のWPMを含み、ペクチンの量を0.1重量%から1重量%まで増加させた系について得られた結果を示す。
発明者らは、健康なミニブタにおける無作為化二重盲検交差試験にて、血漿アミノ酸濃度の食後応答を観察した。2つの食事の間には少なくとも6日のウォッシュアウト期間を設け、この間は通常の食餌をミニブタに与えた。
Claims (15)
- 筋肉減少症、筋萎縮症及び負の窒素バランスからなる群から選択される状態の治療又は予防における使用のための、乳清タンパク質ミセルとペクチンの複合体を含む組成物であって、前記組成物における乳清タンパク質ミセルのペクチンに対する重量比は、30:1〜0.8:1であり、前記乳清タンパク質ミセルは、乳清タンパク質水溶液のpHを3.0〜8.0の間の値に調節し、前記水溶液を80〜98℃の間の温度に供することによって得ることができる、組成物。
- 前記組成物が重篤患者、術後患者、外傷患者、がん患者又は安静中及び安静後の患者に投与される、請求項1に記載の使用のための組成物。
- 前記組成物が食事と組み合わせて投与される、請求項1又は2に記載の使用のための組成物。
- 手術及び/又は入院の少なくとも1日前から手術及び/又は入院後の少なくとも1週間の間、患者に投与される、請求項1〜3のいずれか一項に記載の使用のための組成物。
- 前記組成物が1kg体重あたり0.1g〜2.0gの乾燥重量の乳清タンパク質ミセルを供給するような一日量で投与される、請求項1〜4のいずれか一項に記載の使用のための組成物。
- 前記組成物が熱処理した液体である、請求項1〜5のいずれか一項に記載の使用のための組成物。
- 前記組成物が流動性食事代替物である、請求項1〜6のいずれか一項に記載の使用のための組成物。
- 前記流動性食事代替物が経腸経管栄養法に適した形態である、請求項7に記載の使用のための組成物。
- 前記組成物中の乳清タンパク質ミセルの総含有量が少なくとも5重量%である、請求項1〜8のいずれか一項に記載の使用のための組成物。
- 体重を維持し、筋量を増加させ、又は筋力を増大させるための、乳清タンパク質ミセルとペクチンの複合体を含む組成物の非治療的使用であって、前記組成物における乳清タンパク質ミセルのペクチンに対する重量比は、30:1〜0.8:1であり、前記乳清タンパク質ミセルは、乳清タンパク質水溶液のpHを3.0〜8.0の間の値に調節し、前記水溶液を80〜98℃の間の温度に供することによって得ることができる、組成物の非治療的使用。
- 乳清タンパク質ミセルとペクチンの複合体を含む食品組成物であって、前記組成物における乳清タンパク質ミセルのペクチンに対する重量比は、30:1〜0.8:1であり、前記乳清タンパク質ミセルは、乳清タンパク質水溶液のpHを3.0〜8.0の間の値に調節し、前記水溶液を80〜98℃の間の温度に供することによって得ることができる、食品組成物。
- 前記組成物中の乳清タンパク質ミセルの総含有量が少なくとも5重量%である、請求項11に記載の食品組成物。
- 総乾燥重量のうち5〜20重量%のタンパク質、1〜15重量%の脂質、25〜50重量%の炭水化物及び0.1〜10重量%の繊維を含む、請求項11又は12に記載の食品組成物。
- 前記食品組成物がヨーグルト、食事代替物又は経口栄養補助食品である、請求項11〜13のいずれか一項に記載の食品組成物。
- 前記食品組成物がペットフードである、請求項11〜13のいずれか一項に記載の食品組成物。
