Claims (3)
- 流体サンプルを処理するためのマルチウェルデバイスであって、
上側受容プレートと、中間濾過プレートと、下側収集プレートとを備え、
(a)前記上側受容プレートが、細胞を含む流体サンプルを受容するための複数のウェルを備え、前記上側受容プレート内の前記複数のウェルそれぞれが、内面を有する側壁と、下方向に突出するカラー及び底部流体流れポートを含む底端とを含み、前記上側受容プレートが、2つの対の対向する側壁を含み、前記上側受容プレート内の前記2つの対の対向する側壁の少なくとも1つ内の各対向する側壁が、前記中間濾過プレート上の対応する側壁にある対応するウェルの側壁を受容するための下側開口部を有する溝を含み、
(b)前記中間濾過プレートが、複数のウェルを備え、前記中間濾過プレート内の前記複数のウェルそれぞれが、前記上側受容プレート内の対応するウェルの前記底部流体流れポートを通過した前記流体サンプルを濾過するためのフィルタを備え、
各フィルタが、約1マイクロメートル~約20マイクロメートルの範囲内の平均ポアサイズ、上流面、及び下流面を有する上側深層フィルタ層であって、前記深層フィルタ層の前記上流面が前記フィルタの上流面を構成する、上側深層フィルタ層と、約0.4マイクロメートル~約0.8マイクロメートルの範囲内の平均ポアサイズを有するマイクロポーラス膜を備える中間層と、約0.2マイクロメートルの平均ポアサイズを有するマイクロポーラス膜を備える、上流面及び下流面を有する底層であって、約0.2マイクロメートルの平均ポアサイズを有する前記マイクロポーラス膜の前記下流面が前記フィルタの下流面を構成する、底層とを備え、
前記中間濾過プレート内の前記複数のウェルそれぞれが、内面を有する側壁と、前記フィルタの前記下流面の下方に位置する底壁であって、流体流れポートと、前記底壁から上方向に突出するリブ配置とを含み、前記リブ配置が、前記側壁の前記内面から離間された上面を有する、底壁とを含み、前記フィルタが、前記上側受容プレートの前記下方向に突出するカラーの端部と、前記中間濾過プレート内の前記リブ配置の前記上面との間の圧縮によって前記ウェル内に密封され、
前記中間濾過プレート内の前記複数のウェルのそれぞれの前記底壁はまた、前記流体流れポートを取り囲む下方向に突出するカラーを含み、隣接する中間濾過プレートウェルの下方向に突出するカラー間の空間が、前記下側収集プレート内の対応するウェルの上端を受容するための下側開口部を備えた溝を形成し、
前記中間濾過プレート内の前記流体流れポートが、濾過された流体サンプルが前記中間濾過プレート内のウェルから前記下側収集プレート内の対応するウェルまで進むことを可能にするように配置され、
前記中間濾過プレートの側壁にあるウェルの側壁が、前記上側受容プレートの前記対応する対向する側壁内の対応する溝内に受容可能であり、
(c)前記下側収集プレートが、前記中間濾過プレートの前記対応するウェル内の前記流体流れポートを通過した前記濾過された流体サンプルを受容するように配置された複数のウェルを備え、前記複数のウェルそれぞれが、上端及び底端を有し、前記下側収集プレート内の前記複数のウェルのそれぞれの前記上端が、前記中間濾過プレート内の対応するウェルの前記下方向に突出するカラーを取り囲む溝内に受容可能である、マルチウェルデバイス。 - 前記上側受容プレート内の前記下方向に突出するカラーそれぞれが、内面及び外面を有し、前記上側受容プレート内の隣接する下方向に突出するカラーの前記外面間の空間が、前記中間濾過プレート内の対応するウェルのテーパ状上端を受容するための下側開口部を備えた溝を形成する、請求項1に記載のマルチウェルデバイス。
- 細胞含有流体からタンパク質を得るための方法であって、
請求項1又は2に記載のマルチウェルデバイスの上側受容プレートのウェル内に細胞含有流体のサンプルを配置するステップと、
前記サンプルを濾過するステップと、
下側収集プレート内のウェル内にタンパク質含有流体を得るステップと
を含む、方法。
