Description
Claims (44)
- 生物学的サンプルおよび化学サンプルの精製方法であって、
少なくとも部分的に疎水性である表面を備えるウェルを有する物品を提供する工程;
前記ウェル中に液体サンプルを配置する工程;
前記表面にしっかりと結合する前記サンプル中の第一種類の溶質と、前記表面にしっかりと結合しない前記サンプル中の第二種類の溶質とを有する前記液体サンプルを蒸発させる工程であって、前記表面はウェルの壁面である工程;
前記ウェル中に、前記表面から前記第二種類の溶質を分離する第一緩衝液を配置する工程:
前記第一緩衝液と前記分離された第二種類の溶質とを前記ウェルから除去する工程:並びに、
前記第一緩衝液を除去した後に、前記ウェル中に、前記表面から第一種類の溶質を分離する第二緩衝液を配置する工程、
を含むことを特徴とする方法。 - 前記物品が、マルチ−ウェルプレートであることを特徴とする請求項1に記載の方法。
- 前記物品が、カラムであることを特徴とする請求項1に記載の方法。
- 前記物品が、ピペットであることを特徴とする請求項1に記載の方法。
- 前記物品は、離間された上面および底面を有するターゲット支持プレートと、前記上面および底面の間に伸び、かつ、前記上面および底面を通って伸びる少なくとも1個のカラムと、前記ターゲット支持プレートに取り外せるように固定されるターゲットデバイスとを含み、前記ターゲットデバイスが、少なくとも1個の集積部を有し、前記カラムが前記集積部と正しく合い、前記カラムおよび前記集積部が前記ウェルをひとまとめにして規定することを特徴とする請求項1に記載の方法。
- 前記ターゲットデバイスが、前記ターゲット支持プレートに取り外せるように固定されることを特徴とする請求項5に記載の方法。
- 前記ターゲット支持プレートの少なくとも底面が、前記ターゲットデバイスに取り外せるように接着されるエラストマー性材料から形成されることを特徴とする請求項5に記載の方法。
- 前記エラストマー性材料がシリコンポリマーを含むことを特徴とする請求項7に記載の方法。
- 前記エラストマー性材料が、ポリ(ジメチル)シロキサンを含むことを特徴とする請求項7に記載の方法。
- 前記ターゲット支持プレートが、前記エラストマー性材料から完全に形成されることを特徴とする請求項7に記載の方法。
- 前記ターゲットデバイスが、質量分析法のプレートであることを特徴とする請求項5に記載の方法。
- 前記物品が、エラストマー性材料から少なくとも部分的に形成されることを特徴とする請求項1に記載の方法。
- 前記エラストマー性材料が、シリコンポリマーを含むことを特徴とする請求項12に記載の方法。
- 前記エラストマー性材料が、ポリ(ジメチル)シロキサンを含むことを特徴とする請求項12に記載の方法。
- 前記物品が、前記エラストマー性材料から完全に形成されることを特徴とする請求項12に記載の方法。
- 前記液体サンプルが、トリプシン消化物を含むことを特徴とする請求項1に記載の方法。
- 前記第一種類の溶質がペプチドを含むことを特徴とする請求項1に記載の方法。
- 前記第一種類の溶質が、蛋白質を含むことを特徴とする請求項1に記載の方法。
- 前記第二種類の溶質が塩を含むことを特徴とする請求項1に記載の方法。
- 前記第二種類の溶質が、小さい分子の不純物を含むことを特徴とする請求項1に記載の方法。
- 前記第一緩衝液が水を含むことを特徴とする請求項1に記載の方法。
- 前記第二緩衝液が有機溶媒を含むことを特徴とする請求項1に記載の方法。
- 前記第二緩衝液が、エネルギー吸収性マトリックスを含むことを特徴とする請求項1に記載の方法。
- 前記表面が、アルキルシランおよび疎水性ポリマーからなる群から選択される1種または複数種を含むコーティングであることを特徴とする請求項1に記載の方法。
- 生物学的サンプルおよび化学サンプルの精製方法であって、
生体特異的リガンドで少なくとも部分的に変性される表面を備えるウェルを有する物品を提供する工程;
前記ウェル中に液体サンプルを配置する工程;
前記表面にしっかりと結合する前記サンプル中の第一種類の溶質と、前記表面にしっかりと結合しない前記サンプル中の第二種類の溶質とを有する前記液体サンプルを蒸発させる工程であって、前記表面はウェルの壁面である工程;
前記ウェル中に、前記表面から前記第二種類の溶質を分離する第一緩衝液を配置する工程:
前記第一緩衝液と前記分離された第二種類の溶質とを前記ウェルから除去する工程:並びに、
前記第一緩衝液を除去した後に、前記ウェル中に、前記表面から第一種類の溶質を分離する第二緩衝液を配置する工程、
を含むことを特徴とする方法。 - 前記物品が、マルチ−ウェルプレートであることを特徴とする請求項25に記載の方法。
- 前記物品が、カラムであることを特徴とする請求項25に記載の方法。
- 前記物品が、ピペットであることを特徴とする請求項25に記載の方法。

- 前記ターゲットデバイスが、前記ターゲット支持プレートに取り外せるように固定されることを特徴とする請求項29に記載の方法。
- 前記ターゲット支持プレートの少なくとも底面が、前記ターゲットデバイスに取り外せるように接着されるエラストマー性材料から形成されることを特徴とする請求項29に記載の方法。
- 前記エラストマー性材料がシリコンポリマーを含むことを特徴とする請求項31に記載の方法。
- 前記エラストマー性材料が、ポリ(ジメチル)シロキサンを含むことを特徴とする請求項31に記載の方法。
- 前記ターゲット支持プレートが、前記エラストマー性材料から完全に形成されることを特徴とする請求項31に記載の方法。
- 前記ターゲットデバイスが、質量分析法のプレートであることを特徴とする請求項29に記載の方法。
- 前記液体サンプルが、トリプシン消化物を含むことを特徴とする請求項25に記載の方法。

- 前記生体特異的リガンドが、リン酸化ペプチド/蛋白質またはポリ(ヒスチジン)融合ペプチド/蛋白質用の固定化金属イオンアフィニティークロマトグラフィー(IMAC)マトリックス、ビオチニル化ペプチド/蛋白質用のビオチンアフィニティーマトリックス、およびグルタチオンS−トランスフェラーゼ(GST)融合ペプチド/蛋白質用のチオール−ジスルフィド交換クロマトグラフィーマトリックスからなる群から選択される1種または複数種であってもよいことを特徴とする請求項25に記載の方法。
