Description
すなわち、エチルアルコール含有の発酵生産物である酒粕には、通常20度未満のエチルアルコールが含有されているが、それ以上に高濃度のエチルアルコールによる抽出物を酒類以外の分野で利用することについての知見はなかった。
これらの場合において、濃度20%以上のエチルアルコールその他の極性溶媒を抽出溶媒として抽出された酒粕の極性溶媒抽出物を含有する健康食品または化粧料とすることが好ましい。
(1)特定の化合物に対する感受性の有無についての機能であり、例えばこれら3種のDNA合成酵素は、共にN-メチルマレイミドおよびブチルフェニル−dGTPに対する感受性を持つが、ジデオキシTTP(ddTTP)に対する感受性を持たない。
酒粕極性溶媒抽出物は、I型およびII型のDNAトポイソメラーゼを阻害した。DNAトポイソメラーゼは、DNAの二本鎖を切断・再結合させることによりねじれを解消させ、DNAのトポロジーを変化させる酵素である。
この発明における上記したような各阻害剤の有効な投与量は、疾病の種類と病状の程度、患者の年齢、性別、体重、症状の程度または投与方法によって異なり、一律に限定する必要性はないが、例えば、通常の成人の1人当たり30〜3000mg程度、好ましくは30〜1000mg程度、より好ましくは100〜300mg程度を1日1〜4回程度にわけて経口的または非経口的に投与できる。
化粧料の発明に用いる血管新生抑制性抽出物は、紫外線によって誘導される新生血管が本来の血管の有益性よりも皮膚組織に与えるダメージが大きく、このような血管の新生がしわ形成の一因となる可能性もあるため、前記した皮膚組織のダメージとしわ形成の原因となる血管新生を阻害することによって、健康な皮膚状態を維持しようとするものである。
DNAポリメラーゼとして、哺乳類由来のDNAポリメラーゼα(複製型)およびβ(修復型)について試験を行なった。DNAポリメラーゼαは、牛胸腺から常法により抽出精製した標品を、DNAポリメラーゼβは、ラット由来の該当遺伝子を用いた。
DNAトポイソメラーゼ群に対する活性を以下の方法で測定した。I型およびII型DNAトポイソメラーゼは、組換えヒト由来の市販標品(TopoGEN Inc.,USA)を用いた。文献(Mizushima他,Biochem.J.誌,2002年、第350巻,757-763頁)に記載された方法に従ってI型DNAトポイソメラーゼ活性は、カンプトテシンを、II型DNAトポイソメラーゼは、エトポシド(etoposide,VP-16)を陽性対照として、スーパーコイル(超ラセン、フォームI)を持った環状プラスミドDNAの弛緩(relaxation,フォームII)反応で酵素活性を測定した。反応後、反応液を平板アガロースゲル電気泳動を行い、ゲルを臭化エチジウム染色後、反応生成物のDNA量の変化から、50%阻害濃度(IC50)を求めた。
ヒト子宮癌細胞(HeLa cell)抽出液を用いて、ストレッチPCR法により、東洋紡績社製のTeloChaserキットを使用してテロメラーゼを測定し、テロメラーゼによる6塩基の繰り返し配列(テロメア配列)の生成を検出して、その結果を図5のポリアクリルアミドゲル電気泳動の写真に示した。
酒粕Bの99.5%エチルアルコール抽出液からヘキサンで抽出された残渣中の酢酸エチル抽出液のうち、ヘキサンとジエチルエーテルの所定濃度混液で抽出されたもの[ジエチエーテル配合量%(残部ノルマルヘキサン)をNo1〜6の順に示す。15%、20%、30-50%、50%、100%、0%(アセトン100%)]についてのヒト血管新生抑制性およびヒト胃癌細胞増殖抑制性を調べた。
図6の結果からも明らかなように、エチルアルコール、酢酸エチルおよびジエチルエーテルで抽出されたものには、150μg/mlでヒト胃癌細胞に対する増殖抑制活性が認められた。特に、ジエチルエーテル30−50%とヘキサン70−50%の混液で抽出されたNo.3については、75〜150μg/mlで強い増殖抑制活性が認められた。
酒粕Aおよび酒粕Bの99.5%エチルアルコール抽出液についてのラット動脈片を用いた血管新生抑制性を調べた。

化粧クリームを実際に使用した場合の効果を確認するため、40名のパネラーを10名4群に分け、処方例1〜4のクリームの適量を顔面左半分に、比較例のクリームを顔面右半分に適用した。1ヶ月後、パネラーが肌の状態を顔の両面で比較し、下記の5段階で評価し、結果を表2に示した。なお、この化粧料は、使用期間中に劣化が生じることはなく、皮膚の異常を訴えた者はいなかった。
