Description
b 取り出した組織を酵素等で分解して細胞を分離し、必要とする細胞を選別する。培養対象である細胞は種別を問わない。
c 選別した細胞数が不足している場合、又は、増す必要がある場合には、単層培養等で一旦細胞数を増やす。
d 細胞構成体を作成する。
i 不定形構成体の場合(以下の何れかを必要に応じて、成長因子、薬剤等を添加すればよい。)
・培養液に細胞を浮遊させる。
・ハイドロゲルに細胞を浮遊させる。
・ゲル状担体に細胞を混ぜる。
ii 定型構成体の場合(以下の何れかを必要に応じて、成長因子、薬剤等を添加すればよい。)
・培養液に細胞を浮遊させ、これをコラーゲンスポンジ、キトサンスポンジ等の細胞担体に入れ、細胞を担体に付着させる。
・ゾルの状態で担体に細胞を混入させ、これをコラーゲンスポンジ、キトサンスポンジ等の細胞担体に入れ、細胞を担体に付着させるとともに、ゲル化させる。
e 細胞構成体6は、半透膜製のチューブ又は袋に細胞等を入れて、そのチューブ又は袋を密封して構成する。半透膜は、細胞や分子量の大きなものは透過せず、培養液中成分の栄養分、薬剤、酸素等の気体等分子量の小さいものは透過する。
f 細胞構成体6は、培養ベット36に取り付ける。
g 培養液を循環させる場合は、培養液の循環回路を用意しておく。循環回路には、ガス交換部を設けて、培養装置内のガスと培養回路の培養液中のガスとのガス交換ができるようにする。
h 細胞構成体6を培養ベッド36とともに、培養器部4に入れ、培養液48を満たす。
i 培養器部4にカバー部14を取り付け、培養器部4(及び培養回路)を密封する。以上は、クリーンルーム及びクリーンベンチ等のクリーンな環境で行う。以後は、回路が密封させているので、それ以外の場所でも雑菌の汚染はない。
j 培養器部4には、駆動部8を取り付ける。
k 温度、ガス濃度を最適状態に保つ。
l 繰り返し、曲げ動作を細胞構成体6に付加する。曲げ動作の周期、大きさ、動作スケジュール等は前もって設定して運転する。
m 必要に応じて、培養液の循環、加圧を行う。加圧は、一定、断続、周期的な繰り返し等の加圧パターンから最適なパターンを選択する。
n 所定の時間が経過したら、培養器部4から駆動部8を外す。

前記筐体部に摺動可能に支持され、前記レバーに前記スライダと反対方向に復帰力を作用させる第2のスライダと、
を備えることを特徴とする培養装置。
前記レバーを挿入する挿入口と、
前記挿入口から挿入される前記レバーの端部を前記第1のスライダと前記第2のスライダとの間にガイドするガイド部と、
を前記筐体部に備えることを特徴とする培養装置。

Claims (4)
- 細胞及び/又は組織を含む培養物の培養装置であって、
前記培養物を載置するベッドと、
培養空間部を有する本体部とカバー部とを備え、前記培養空間部に前記ベッドとともに該ベッドに載置された前記培養物を収容し、前記本体部と前記カバー部とで前記ベッドを保持する培養器部と、
前記培養器部の内外に貫通させ、前記培養空間部内で回動可能にしたレバーと、
前記レバーを操作して該レバーの先端部を円弧回動させ、該円弧運動によって前記先端部を前記ベッドに接触させて力を付与し、前記ベッドを湾曲変形させて前記培養物に変位を付与する駆動手段と、
を備えることを特徴とする培養装置。 - 請求項1記載の培養装置において、
前記レバーは、前記培養器部の壁部又はその近傍に設定された支点を中心に円弧運動することを特徴とする培養装置。 - 細胞及び/又は組織を含む培養物の培養装置であって、
培養空間部を有する本体部とカバー部とを備え、前記培養空間部に前記培養物の一端を取り付けて収容する培養器部と、
前記培養器部の内外に貫通させ、先端部が前記培養物の他端に取り付けられ、前記培養器部の壁部又はその近傍に設定された支点を中心に前記培養空間部内で円弧運動することが可能なレバーと、
前記レバーを操作して該レバーの先端部を円弧回動させて前記培養物に伸縮力を付与し、前記培養物を伸縮させる駆動手段と、
を備えることを特徴とする培養装置。 - 細胞及び/又は組織を含む培養物の培養方法であって、
培養空間部を有する本体部とカバー部とを備える培養器部の前記培養空間部に前記ベッドとともに該ベッドに載置された前記培養物を収容し、前記本体部と前記カバー部とで前記ベッドを保持し、前記培養器部の内外に貫通させ前記培養空間部内で回動可能にしたレバーに、前記培養器部の壁部又はその近傍に設定された支点を中心に円弧運動を付与し、前記レバーの先端部を円弧回動させ、前記先端部を前記ベッドに接触させて力を付与し、前記培養物に曲げ変位を付与することを特徴とする培養方法。
