【発明の属する技術分野】
本発明は、食品または医薬として有用な肝機能賦活剤、特に臓器の加水分解物と植物由来の肝機能賦活成分とを組み合わせた肝機能賦活剤に関する。

【従来の技術】
肝臓は人体の臓器の中で最大の臓器であり、胃、腸、膵臓、脾臓などの他の消化器官と血管を通じて関係している生体機能の中心的な臓器である。その働きは、栄養物の代謝、胆汁の分泌、有害物や毒物の解毒、免疫力の活性など、多岐にわたっている。肝臓は普段はたくましい丈夫な臓器であるが、いったん病気(ウィルス性肝炎、アルコール性肝炎、薬剤性肝炎など)になると、回復がなかなか困難であり、今日でもなお肝機能障害に対し優れた予防治療薬を求める声は大きい。

肝機能を高める薬剤としては、アミノ酸製剤、グルタチオンや、田七ニンジン、ウコンなどの生薬などが使用されているが、これらは肝機能賦活作用が緩和で優れたものは少ない。また、肝臓病治療剤として、プロトポルフィリン2ナトリウム製剤、チオプロニン製剤、グリチルリチン製剤、ジクロロ酢酸ジイソプロピルアミン製剤などの肝再生増強剤、抗炎症剤や種々の解毒剤が用いられるが、これらは対症療法の薬で肝臓病を根本的に治すものではなく進行を遅らせるのが目的であるだけである。さらに副作用が強くでる恐れもあり、使用する際は充分注意する必要がある。
医薬分野が進んだ今日でも、優れた肝機能改善薬はいまだ現れず、画期的な医薬として唯一、ウィルス性肝炎の予防ができるワクチン製剤が使用されるようになっているだけである。また、肝機能障害の患者は、肝機能低下がかなり進行してから医師にかかる場合が多く、そのような状況にあって、OTC医薬品や食品といった比較的手軽に、いわゆるセルフメディケーションとして使用できるものが強く望まれている。

【発明が解決しようとする課題】
本発明の目的は、疾病その他種々の原因に由来する肝機能障害や肝機能低下を治療、軽減あるいは予防するための肝機能賦活剤を提供することにある。
本発明の他の目的は、副作用が少なく、それゆえにOTC医薬品や食品などとして比較的手軽に利用できる肝機能賦活剤を提供することにある。

【課題を解決するための手段】
かかる実情から、本発明者らは鋭意研究を行った結果、臓器の加水分解物と植物由来の肝機能賦活成分とを組み合わせた肝機能賦活剤が、優れた肝機能亢進作用と肝障害抑制作用を有し、さらに意外にもこれらそれぞれ単独の使用時よりも有意に優れた肝機能賦活作用を示すことを見出した。
従来、肝臓の加水分解物が肝機能賦活作用を有することは知られており(Gilbert, A., Carnot, P., Compt Rend. Soc. Biol., 48, 934 (1894))、また植物由来の肝機能賦活成分は上述のように種々のものが知られているが、臓器の加水分解物と植物由来の肝機能賦活成分とを組み合わせることは知られていなかった。それゆえに、これら2種の成分の組み合わることにより、それぞれを単独で使用するときよりも有意に優れた肝機能賦活作用を示すことも知られていなかった。特に、肝臓加水分解物とマリアアザミ種子抽出物とを組み合わせると、肝機能賦活作用が大きかった。
本発明者らは、さらに検討を重ねて本発明を完成した。

すなわち、本発明は、
(1)臓器の加水分解物と植物由来の肝機能賦活成分とを含有することを特徴とする肝機能賦活剤、
(2)臓器が肝臓であることを特徴とする前記(1)に記載の肝機能賦活剤、
(3)植物由来の肝機能賦活成分がマリアアザミ種子抽出物であることを特徴とする前記(1)に記載の肝機能賦活剤、
(4)肝臓加水分解物およびマリアアザミ種子抽出物を含有することを特徴とする前記(1)に記載の肝機能賦活剤、
(5)経口用形態である前記(1)〜(4)記載の肝機能賦活剤、
(6)食品である前記(5)記載の肝機能賦活剤、および、
(7)成人1日あたりの摂取または投与量が、肝臓加水分解物0.1〜10g、マリアアザミ種子抽出物(原植物換算で)2〜15gである前記(4)記載の肝機能賦活剤、
に関する。

【発明の実施の形態】
肝機能賦活作用としては、肝機能亢進作用または肝障害抑制作用が挙げられる。より具体的には、肝血流の増加作用、胆汁分泌促進作用、肝臓組織呼吸促進作用、脂肪の肝臓への蓄積の阻止もしくは抑制作用、ウィルス性・薬剤性・アルコール性肝障害抑制作用等が挙げられる。

本発明における臓器の加水分解物としては、動物から得られる任意の臓器を加水分解することによって得られるものが挙げられる。また、異なる2種以上の臓器の加水分解物の混合物であってもよいし、1種類の臓器から異なる加水分解方法で得られる加水分解物の混合物(以下、これらをまとめて「加水分解物の混合物」と総称する。)であってもよい。異なる2種以上の臓器を用いる場合は、異なる動物から得られる臓器であってもよいし、同一の動物から得られる異った臓器であってもよい。
動物臓器としては、動物の種類、臓器の種類を問わずいずれのものでも使用することができ、例えばウシ、ブタまたはヒツジなどの心臓、肝臓、胎盤、脳などが用いられる。
加水分解の方法は特に限定されず、例えば、動物臓器を酸・塩基で加水分解する方法、動物臓器をペプシン、トリプシン、キモトリプシン、パパイン、プロメライン、フィシン等のプロテアーゼで加水分解する方法、または動物臓器を動物の膵臓や組織と混合してホモジナイズすることにより加水分解する方法などを用いることができる。
得られた加水分解物は、所望により濾過、滅菌等の操作を行ってもよいし、公知方法により乾燥、粉末化してもよい。これらの処理を2以上組み合わせてもよい。
本発明において臓器の加水分解物としては、畜産動物(ウシ、ブタ、ヒツジなど)の肝臓の加水分解物又は肝臓加水分解物の混合物が特に好ましい。

上記肝臓加水分解物は、アミノ酸、ペプチドなどに富んだ素材で、HAP(動物蛋白加水分解物)として調味料に使用されるほか、栄養強化、肝機能改善薬などに使用されている。

肝機能賦活作用を示す植物としては、自体公知のものを用いてよいが、具体的には、マリアアザミ(Silybum marianum、別名オオヒレアザミ、Milk Thistle)、ウコン(Curcuma longa L.)、藍、タンポポ、西洋タンポポ、ゴボウ、ニンニク、キク、西洋ノコギリソウ、クチナシ、ゴマ、田七ニンジン(Panax pseudo ginseng)、アスパラガス、タマネギ(Allium Cepa)、チコリ、薬用サルビア、朝鮮アザミ(アーティチョーク)、クコ、マメ科・アヤメ科・バラ科の植物(例えば大豆やクズ、マメ科に属するアスパラサス・リネアリス)、ミヤマウズラ(Goodyera schlechtendaliana REICHB. fil. )、エルバ・デ・パサリーニョ(Struthantus Flexicaulis Mart.)、セテサングリア(Cuphea balsamona Cham.)、アカメガシワ(Mallotus Japonicus Mueller Agroviensis, 大戟科Euphorbiaceae )、紅茶(茶の葉を公知手段により加工したもの)などが挙げられる。また、これら植物の変種であってもよい。

本発明において植物由来の肝機能賦活成分としては、肝機能賦活作用を示す植物もしくはその処理物を用いることができる。
処理とは、例えば、乾燥裁断、抽出、滲出物採取、粉砕など、人為的手段を施すことが挙げられる。
本発明においては、二種以上の植物またはその処理物を混合してもよく、一種類の植物から得られた抽出物または滲出物などの異なった二以上の処理物を組み合わせて混合してもよい。該処理物は、液状であってもよく、粉末状、粒状などの固形状やペースト状などの半固形状であってもよい。
また、液状の処理物をさらに噴霧乾燥や凍結乾燥など公知方法を用いて粉末状、粒状などの固形状にしてもよい。逆に、固形状や半固形状の処理物をさらに適当な溶媒に溶解または懸濁して液状にしてもよい。

肝機能賦活作用を示す植物の抽出物は慣用の方法により得ることができる。例えば、該植物を、所望により裁断、乾燥、粉砕などの処理を施した後、適当な抽出溶媒を用いて、常圧又は加圧下、室温又は加熱下で抽出し、所望によりその後濾過または濃縮することにより、前記抽出物を得ることができる。植物は一種又は二種以上混合して抽出に供することができる。上記抽出物は、例えば分別抽出、二溶媒間の分配、適当な吸着剤による分別吸着/溶離ないしクロマトグラフィーなどによって精製してもよい。

抽出に供する植物体の部位は、植物の種類によっても異なるが、植物体全体であってもよく、植物体の一部分、例えば、根、茎、葉、果実、種子、果皮、樹皮、幹、技、花などであってもよい。
たとえば、(a)ウコン、田七ニンジンまたはアスパラガスは根茎を、(b)タンポポ、西洋タンポポ、チコリは根を、(c)タマネギ、ニンニクは鱗茎を、(d)西洋ノコギリソウ、薬用サルビア、朝鮮アザミは地上部を、(e)マリアアザミ、クチナシ、クコは果実を、(f)ゴマは種子を、(g)ゴボウは根または種子を、(h)ミヤマウズラ、セテサングリアは全草を、(i)エルバ・デ・パサリーニョ(Struthantus Flexicaulis Mart.)は葉部を、(j)アカメガシワは樹皮を、抽出に供するのが好ましい。

本発明における肝機能賦活作用を示す植物の滲出物も慣用の方法により得ることができる。例えば、植物体の一部、例えば、樹幹などに傷を付け、樹脂などの滲出物を採取したり、植物体又はその一部を、所望により熱水処理などを施した後、圧搾して採取することにより前記滲出物を得ることができる。また、上記滲出物は、例えば分別抽出、二溶媒間の分配、適当な吸着剤による分別吸着/溶離ないしクロマトグラフィーなどによって精製してもよい。
なお、本発明においては、このように該植物から分離した滲出物のみならず、該植物体又はその一部から滲出する滲出物をも利用できる。例えば、破砕、乾燥などの処理を施した植物から、滲出物が滲出する場合がある。このような滲出物も、本発明における「滲出物」に含まれ、本発明に係る植物由来の肝機能賦活成分として使用できる。

本発明に係る植物由来の肝機能賦活成分としては、マリアアザミの種子の抽出物、ウコンエキス、田七ニンジンエキスが好ましい。
以下に、これらについて詳細に記載するが、かかる記載は本発明の好ましい態様であって、これに限定されることはない。
マリアアザミはキク科の植物で、原植物にシリビニン(Silibinin, C25H22O10, Mw = 482.4)を原植物換算で1%以上含有し、さらにシリビニン換算でフラボノリグナン類(通称シリマリン)を1.5%以上(原植物換算)含有するものを用いるのが好ましい。本発明においては、該植物の果実の抽出物を用いるのが好ましい。特に含水エタノール抽出により得られ、ドイツ薬局方DAB 1997の規格以上のものが好ましい。

ウコンは、ショウガ科の植物である。本発明においては、ウコンエキスを用いるのが好ましい。該ウコンエキスとしては、ウコンの乾燥根茎をヘキサン・エタノール及び含水エタノールで抽出し、スプレードライにより得られた乾燥粉末を用いるのが好ましい。
田七ニンジンはウコギ科の植物である。本発明においては、田七ニンジンエキスを用いられるのが好ましい。該エキスとしては、田七人参の根茎の30%エタノール抽出物を約60℃以下で減圧濃縮し、粘凋性エキスとしたものを用いるのが好ましい。

また、本発明に係る植物由来の肝機能賦活成分として、グルテリンとプロラミンなどの穀類由来の植物タンパクを用いてもよい。また、植物生薬として知られる、例えば、柴胡(さいこ)、桃仁(とうにん)、牡丹皮(ぼたんぴ)、紅花(こうか)、三稜(さんりょう)、莪求(がじゅつ)を用いてもよい。

本発明の肝機能賦活剤の投与量は、患者の性別、年齢、症状などによって異なるので、適宜選択することができる。しかし、1日あたり、以下に挙げる範囲の動物臓器の加水分解物と植物由来の肝機能賦活成分とを含有する肝機能賦活剤を摂取または投与(以下、単に摂取と略す)するのが好ましい。また摂取回数は1日1回であっても、または複数回であってもよい。
臓器の加水分解物、特に肝臓加水分解物の摂取量(乾燥物換算)は、約0.1〜10g程度、好ましくは約0.3〜7g程度、さらに好ましくは約0.4〜5g程度である。
植物由来の肝機能賦活成分については、公知の摂取量を参酌して適宜定めることができる。例えば、マリアアザミ抽出物は、原生薬乾物換算で約2〜15g程度、好ましくは約4〜8g程度摂取するのがよい。田七人参エキスは、原生薬乾物換算で約1〜10g程度、好ましくは約2〜4g程度摂取するのがよい。ウコンエキスは、原生薬乾物換算で約50〜2000mg程度、好ましくは約150〜350mg程度摂取するのがよい。

本発明においては、他の肝機能賦活剤を含有させてもよい。
他の肝機能賦活剤としては、肝機能賦活作用を有する自体公知の化合物を用いてよい。具体的には、例えば、グルタチオン;ビタミンC(アスコルビン酸)、ビタミンE(トコフェノール)、ビタミンB1、B2、B6などのビタミン;α−リポ酸;レシチン;フラボノール類、フラボン類、フラバン類、フラバノール類、カテキン類、イソフラボン類などの植物フラボノイド;リグナン酸;クルクミノイド類などが挙げられる。
また、例えば、バリン、ロイシン、イソロイシン、プロリン、アルギニン、アルギン酸、ヒスチジン、トリプトファンなどのアミノ酸、またはキチンオリゴ糖、キトサンオリゴ糖もしくはその塩を他の肝機能賦活剤として含有させてもよい。

その他にも、他の肝機能賦活剤としてエタノールアミン(モノエタノールアミン、2−アミノエタノールともいう)、ホスホエタノールアミン及びホスホグリセロエタノールアミン又はそれらの塩を用いてもよい。
また、アラキドン酸またはリノール酸を他の肝機能賦活剤として含有させてもよく、 エイコサペンタエン酸、ドコサヘキサエン酸、γ−リノレン酸、これらの酸を含有する油脂をさらに含有させるのも好ましい。

本発明にかかる肝機能賦活剤は、他の機能を有する医薬または機能性食品等を含有していてもよい。
また、本発明の肝機能賦活剤には、肝機能賦活効果を損なわない範囲において許容しうる種々の添加物を配合することができ、具体的にはセルロースおよびその誘導体、デンプンやデキストリンおよびその誘導体などの天然および合成高分子の賦形剤、ステアリン酸およびその塩類などの乳化剤、滑沢剤や糖類、pH調整剤、香料、着色料などを配合することができる。

本発明の肝機能賦活剤の投与経路は特に限定されず、経口または非経口投与のいずれかから予防もしくは治療のために最も効果的な投与経路を適宜選択することができるが、経口投与のほうが好ましい。
本発明に係る肝機能賦活剤の剤形は、摂取方法および摂取経路に応じて散剤、顆粒剤、丸剤、錠剤、硬カプセル剤、軟カプセル剤、シロップ剤など経口用剤、坐剤、注射剤などの種々の形態とすることができるが、経口用剤として使用するのが好ましい。また、本発明の肝機能賦活剤は、医薬のみならず、食品として使用してもよい。食品として用いられる場合には、顆粒、錠菓、ガム、キャンディ、ゼリー、飲料等の形で提供されうるが、形態を問わない。

【実施例】
以下、実施例を挙げて本発明を詳細に説明するが、本発明はこれらに限定されるものではない。

肝機能の状態を簡便に知る方法として、血漿中または血清中のGOT、GPT、LDHなどの酵素活性を測定する方法がある。GOTはアスパラギン酸・α−ケトグルタル酸とグルタミン酸・オキザロ酢酸との間のアミノ基転移酵素で、心筋、肝臓、脳に多く存在しており、心筋梗塞や劇症肝炎などで血清GOTが顕著に上昇することが知られている。GPTはアラニン・α−ケトグルタル酸とグルタミン酸・ピルビン酸との間のアミノ基転移酵素で、肝臓や腎臓に存在し、急性肝炎や慢性肝炎で上昇することが知られている。また、LDHはピルビン酸と乳酸との間の酸化還元反応に関与する酵素で、心筋、肝臓、腎臓などに存在しており、心筋梗塞、肝硬変、腎疾患などによってこれらの臓器に障害が起こったり壊死したりすると血液中にLDHが逸脱するため、血清中のLDHが上昇することが知られている。従って、これらの酵素活性を測定することは肝臓疾患の診断上有力な指標になる。

一方、四塩化炭素は実験動物に投与すると比較的容易に肝細胞の壊死を引き起こし、血清中にGOT、GPT、LDHなどの酵素が逸脱することが知られている。また、脂肪肝を形成しやすく、肝機能や肝損傷の生化学的研究手段として広く用いられている。

〔試験例1〕
マリアアザミ種子抽出物(乾燥粉末、抽出物:原植物=1:35、シリマリンとして72.5重量%含有)〔エミール フラックスマン(Emil Flachsmann AG)社製 商品番号12,066〕および豚肝臓加水分解物(乾燥粉末)を用い、マウスの四塩化炭素誘発肝障害に対する抑制作用について実験を行った。22週齢の雌性、DBA/2マウスを水と飼育用飼料(CE−2、商品名、日本クレア株式会社製)で4日間予備飼育して馴化させた後、対照群、マリアアザミ種子抽出物投与群、肝臓加水分解物投与群、および本発明品投与群の4群(1群あたり6〜7匹)に分けた。対照群には飼育用粉末飼料CE−2を与え、試験群には同飼料にマリアアザミ抽出物を2重量%、肝臓加水分解物を2重量%、およびこれら2素材を2重量%ずつ添加した飼料を与えて6日間飼育した。

実験食摂取後6日目に0.36%四塩化炭素(V/V、オリーブ油)を7.5ml/kgの投与量で腹腔内に投与した。投与24時間後に採血し、遠心分離後、血漿中のグルタミン酸オキザロ酢酸アミノ基転移酵素(GOT)活性、グルタミン酸ピルビン酸アミノ基転移酵素(GPT)活性、および乳酸脱水素酵素(LDH)活性を、和光純薬製の「GOT−UVテストワコー」、「GPT−UVテストワコー」、「Lタイプワコー LDH」を用いて日立自動分析装置7070で測定した。試験結果は、対照群の血漿酵素活性を100(%)としたときの各試験群の血漿酵素活性の割合を求め、肝障害強度として示した。結果を〔表1〕に示す。

結果から明らかなように、本発明品投与群は対照群に比べて血漿GOT、GPTおよびLDH活性が顕著に抑制され、その作用は素材単独投与群よりも明らかに強い効果であった。この結果から、本発明品は肝障害抑制作用が認められ、肝臓疾患の予防あるいは改善に有効であることが示された。

【発明の効果】
本発明の肝機能賦活剤は、優れた肝機能賦活効果を有し、疾病や種々の要因による肝機能低下、肝障害に優れた効果を示す。
