Description
a.微生物、酵素、および補因子の組成物を、栄養素の混合物と共に、廃水に添加するステップと、
b.前記組成物を、所定の所要時間、廃水と反応させるステップと、
c.反応を定量して、廃水の浄化の程度を決定するステップとを含む。
個々の微生物株を、対数増殖期まで純粋培養で別々に培養する。次いで、上述の微生物株の全てを含む共培養物を調製するために、各純粋培養物の10%(v/v)を採取する。このように調製された共培養物を、最短で8〜14時間にわたりさらにインキュベートして、最終配合物を調製するための全ての株を最大限に増殖させる。本発明の実施形態では、各株の純粋培養物の10%v/v(共培養物の全体積に対する特定株の培養物の体積)であって、それらの増殖サイクルの対数期にあるものを選択する。
個々の微生物株を、対数増殖期まで、純粋培養で栄養素内で別々に培養する。次いで、上述の微生物株の全てを含む共培養物を調製するために、各純粋培養物の10%(v/v)を採取する。このように調製された共培養物を、最短で8〜14時間にわたりさらにインキュベートして、最終配合物を調製するための全ての株を最大限に増殖させる。40%から60%体積/体積の範囲で採取された微生物株の共培養物に、60%から40%体積/体積の範囲の酵素を添加する。5%〜30%の範囲の補因子を、酵素、微生物、および栄養素の組合せと共に添加する。
南インドの織物加工ユニットの通気タンクの入口から収集された廃液について実験を行った。廃液のpHを、0.1N HClで6.5に調節した。「試験」サンプルの場合、30%過酸化水素0.16mlを硫酸第一鉄30mgと共に、廃水サンプル200mlに添加し、これを磁気スターラ上で20分間保持することにより、個々の試験のそれぞれに使用される適正な混合および拡散を促進させた。微生物、酵素、および栄養素の組合せは、栄養素中の既に記述された微生物の共培養物40%(v/v)と、既に記述された酵素からなる酵素ブレンド60%(v/v)とを添加することによって調製した。この組合せ0.1mlを廃水サンプルに添加し、磁気スターラ上で20分間保持した。この後、サンプルを、オービタル・シェーカ上で、30℃および50rpmでインキュベーションした。微生物、酵素、補因子、および栄養素を添加しない廃水からなる対照サンプルを、上述の条件で、「試験」と同じ長さの時間にわたりインキュベートした。5mlのサンプルを、「試験」ならびに「対照」集合の両方から、実験開始から0.1時間、4時間、8時間、および16時間で収集した。サンプルを、5000rpmで5分間、遠心分離した。遠心分離後に得られた上澄みの色、化学的酸素要求量、および全溶解固形分を、標準的な手法により評価した。
インドの石油化学精製所からのフェノールに富む廃液を研究に使用した。廃液のpHを、0.1N HClで6.5に調節した。「試験」サンプルの場合、30%過酸化水素0.16mlを硫酸第一鉄30mgと共に、廃水サンプル200mlに添加し、これを磁気スターラ上で20分間保持することにより、個々の試験のそれぞれに使用される適正な混合および拡散を促進させた。微生物、酵素、および栄養素の組合せは、栄養素中の微生物の共培養物40%(v/v)と、既に記述された酵素からなる酵素ブレンド60%(v/v)とを添加することによって調製した。この組合せ0.1mlを廃水サンプルに添加し、磁気スターラ上で20分間保持した。この後サンプルを、オービタル・シェーカ上で、30℃および50rpmでインキュベーションした。微生物、酵素、補因子、および栄養素を添加しない廃水からなる対照サンプルを、上述の条件で、「試験」と同じ長さの時間にわたりインキュベートした。5mlのサンプルを、「試験」ならびに「対照」集合の両方から、実験開始から0時間、6時間、12時間、および24時間で収集した。サンプルを、5000rpmで5分間、遠心分離した。遠心分離後に得られた上澄みのフェノール含量を、メタノール:水(60:40)を移動相としたC18カラムを用いてRP−HPLCにより予測し、UV検出器により254nmで検出した。
Claims (10)
- シュードモナス・イルージノウザ(Pseudomonas aeruginosa)、シュードモナス・フルオレッセンス(Pseudomonas fluorescence)、シュードモナス・プチダ(Pseudomonas putida)、シュードモナス・デスモリティカム(Pseudomonas desmolyticum)、コリオルス・ベルシコル(Coriolus versicolour)、ラクトバチルス・エスピー(Lactobacillus sp.)、バチルス・サブチリス(Bacillus subtilis)、バチリス・セレウス(Bacillus cereus)、スタフィロコッカス・エスピー(Staphylococcus sp.)、およびファネロキーテ・クリソポリウム(Phanerochaete chrysosporiu)からなる微生物と;
ペルオキシダーゼ(マンガン依存性およびマンガン非依存性)、リグニンペルオキシダーゼ、ラッカーゼ、カタラーゼ、シトクロムcオキシダーゼ、グルコースオキシダーゼ、フェノールオキシダーゼ、n−およびo−デメチラーゼ、プロテアーゼ、リパーゼ、α−アミラーゼ、およびバクテリオシンからなる酵素と;
栄養素と;
酸化剤ならびに第一鉄および第二鉄イオンの塩からなる補因子と
の組み合わせを含む、廃水を処理するための組成物。 - 前記酵素が、10%〜20%v/vの前記ペルオキシダーゼ(マンガン依存性およびマンガン非依存性)、7%〜10%v/vの前記リグニンペルオキシダーゼ、7%〜10%v/vの前記ラッカーゼ、1%〜5%v/vの前記カタラーゼ、0.5%〜3%v/vの前記シトクロムcオキシダーゼ、5%〜10%v/vの前記グルコースオキシダーゼ、3%〜5%v/vの前記フェノールオキシダーゼ、1%〜2%v/vの前記n−およびo−デメチラーゼ、5%〜7%v/vの前記プロテアーゼ、5%〜7%v/vの前記リパーゼ、5%v/vの前記α−アミラーゼ、および1%v/vの前記バクテリオシンから選択される、請求項1に記載の組成物。
- 前記酸化剤が過酸化水素である、請求項1に記載の組成物。
- 前記塩が、硫酸第一鉄・7水和物および塩化第二鉄である、請求項1に記載の組成物。
- 前記栄養素が、酵母エキス、動物組織のペプシン消化物、リン酸二アンモニウムおよび塩化ナトリウム、トリプトン、大豆エキス、ゼラチンの酵素消化物、硫酸マグネシウム、塩化カルシウム、塩化マグネシウム、および硫酸カリウムを含む、請求項1に記載の組成物。
- 前記栄養素が、0.2%〜0.5%の前記酵母エキス、0.3〜0.6%の前記動物組織のペプシン消化物、0.05%〜0.2%の前記リン酸二アンモニウムおよび0.3%〜0.8%の前記塩化ナトリウム、0.3%〜0.6%の前記トリプトン、0.2%〜0.5%の前記大豆エキス、0.2%〜0.5%の前記ゼラチンの酵素消化物、0.3%〜0.8%の前記硫酸マグネシウム、0.3%〜0.8%の前記塩化カルシウム、0.3%〜0.8%の前記塩化マグネシウム、および0.3%〜0.8%の前記硫酸カリウムを含む、請求項5に記載の組成物。

- 前記塩が、硫酸第一鉄・7水和物および塩化第二鉄である、請求項7に記載の方法。
- 前記栄養素が、酵母エキス、動物組織のペプシン消化物、リン酸二アンモニウムおよび塩化ナトリウム、トリプトン、大豆エキス、ゼラチンの酵素消化物、硫酸マグネシウム、塩化カルシウム、塩化マグネシウム、および硫酸カリウムを含む、請求項7に記載の方法。
