Description
A)少なくとも1つの表面露出ポリペプチド(例えば、病原性または腫瘍細胞由来)またはその抗原部分を含むN−またはC−末端抗原領域であって、前記抗原領域は、
B)オリゴマー化領域(OMD)に融合され、前記オリゴマー化領域は、
C)リンカー領域を介して、
D)単一コピーのLysM領域からなり、グラム陽性菌から得られる不活化バクテリア様粒子(BLP)である前記ポリペプチドのペプチドグリカン担体粒子への前記非共有結合性連結を仲介することが可能なペプチドグリカン結合領域(PBD)に融合され、かつ前記ポリペプチド全体として単一コピーのLysM領域のみを含む組み替えポリペプチド、に関する。
i.粒子状担体としての、グラム陽性菌から得られた不活化バクテリア様粒子(BLP)と、
ii.前記BLPに非共有結合的に連結された、組み替えにより製造されたポリペプチドのオリゴマーであって、前記組み替えポリペプチドは、
A)N−またはC−末端抗原領域であって、病原性または腫瘍起源の少なくとも1つの表面露出ポリペプチドもしくはその抗原部分を含み、前記抗原領域は、
B)オリゴマー化領域(OMD)に融合され、前記オリゴマー化領域は、
C)リンカー領域を介して、
D)前記ポリペプチドの前記BLPへの前記非共有結合的連結を仲介する単一コピーのLysM領域からなり、前記ポリペプチド全体として単一コピーのLysM領域のみを含有するペプチドグリカン結合領域(PBD)に融合された、前記ポリペプチドのオリゴマーと、
iii.調剤的に許容可能な希釈剤または賦形剤とを含む疫組成物、を提供する。
−コリンを含む担体に結合するための、例えば、肺炎球菌酵素LytA(例えば、Pfam PF01473)といったオートリシンからのコリン結合領域。
−グルコース含有ポリマーからなる粒子に結合するための、例えば、グルカンスクラーゼ(例えばPfam PF08363)といった腔連鎖球菌からのグリコシルトランスフェラーゼグルカン結合領域。
−ホスファチジル・イノシトール−リピッド結合領域、例えばプレクストリン(Pleckstrin)相同(PH)ドメイン(Pfam PF00169)。
A)病原体由来の少なくとも1つの表面露出ポリペプチドまたはその抗原部分を含むN−またはC−末端抗原領域であって、インフルエンザ・ヘムアグルチニン(HA)細胞外ドメインまたはその一部分、インフルエンザ・ノイラミニダーゼ(NA)細胞外ドメインまたはその一部分、コロナウイルス・スパイク(S)タンパク質細胞外ドメインまたはその一部分、RSV糖タンパク質FまたはG細胞外ドメインまたはその一部分、およびHIV gp140細胞外ドメインまたはその一部分からなる群より選択されるN−またはC−末端抗原領域を含み、前記抗原領域は、
B)オリゴマー化領域(OMD)に融合され、前記オリゴマー化領域は、
C)リンカー領域を介して、
D)単一コピーのLysM領域からなり、グラム陽性菌から得られる不活化バクテリア様粒子(BLP)である前記ポリペプチドのペプチドグリカン担体粒子への前記非共有結合性連結を仲介することが可能なペプチドグリカン結合領域(PBD)に融合され、かつ前記ポリペプチド全体として単一コピーのLysM領域のみを含む。
B.シチメンチョウRBCを用いる血球凝集反応試験。融合に使用されるHAはH1Mである。レーン1:陰性対照、RBCs+緩衝液。レーン2:RBCs+BLP。レーン3:RBCs+4μgのHA−P1Ltriを結合されたBLP。レーン4:RBCs+2μgのHA−P1Ltriを結合されたBLP。レーン5:RBCs+1μgのHA−P1Ltriを結合されたBLP。レーン6:RBCs+0.5μgのHA−P1Ltriを結合されたBLP。レーン7:RBCs+4μgのHA−P1Ltri。レーン8:RBCs+4μgのHA−P2Lmonを結合されたBLP。レーン9:RBCs+2μgのHA−P2Lmonを結合されたBLP。レーン10:RBCs+1μgのHA−P2Lmonを結合されたBLP。レーン11:RBCs+0.5μgのHA−P2Lmonを結合されたBLP。レーン12:RBCs+4μgのHA−P2Lmon。この実験が明らかに示しているように、オリゴマーHAのみがBLPs(BLP−HAtri)に結合されていると、生物学的な機能を有するが、これに対して、単量体HAがBLPs(BLP−HAtri)に結合されている場合は、生物学的な機能を有しない。
Claims (27)
- 微粒子型免疫組成物であって、
i.ペプチドグリカン担体粒子としての、グラム陽性菌から得られた不活化バクテリア様粒子(BLP)と、
ii.前記BLPに非共有結合的に連結された、組み替えにより製造されたポリペプチドのオリゴマーであって、前記組み替えポリペプチドは、
A)N−またはC−末端抗原領域であって、病原性または腫瘍起源の少なくとも1つの表面露出ポリペプチドもしくはその抗原部分を含み、前記抗原領域は、
B)オリゴマー化領域(OMD)に融合され、前記オリゴマー化領域は、
C)リンカー領域を介して、
D)前記ポリペプチドの前記BLPへの前記非共有結合的連結を仲介する単一コピーのLysM領域からなり、前記ポリペプチド全体として単一コピーのLysM領域のみを含有するペプチドグリカン結合領域(PBD)に融合された、前記ポリペプチドのオリゴマーと、
iii.調剤的に許容可能な希釈剤または賦形剤と、を含み、
前記LysM領域は、L.lactis AcmAにおける3つのLysM領域のうちの1つと、少なくとも70%以上の配列同一性を有する免疫組成物。 - 請求項1に記載の免疫組成物において、
前記組み替えポリペプチドの前記抗原領域における前記表面露出ポリペプチドは、エンベロープを有するウイルスのタンパク質の細胞外ドメインを含む免疫組成物。 - 請求項2に記載の免疫組成物において、
前記ウイルスは、インフルエンザ・ウイルス、動物コロナウイルス、ヒト呼吸器コロナウイルス、ヒト免疫不全ウイルス(HIV)、パラミクソウイルスである免疫組成物。 - 請求項3に記載の免疫組成物において、
前記ウイルスは、呼吸器合胞体ウイルス(RSV)またはメタ肺炎ウイルスである免疫組成物。 - 請求項2に記載の免疫組成物において、
前記表面露出ポリペプチドまたはその抗原部分は、インフルエンザ・ヘムアグルチニン(HA)細胞外ドメインまたはその一部分、インフルエンザ・ノイラミニダーゼ(NA)細胞外ドメインまたはその一部分、コロナウイルス・スパイク(S)タンパク質の細胞外ドメインまたはその一部分、RSV糖タンパク質FもしくはGの細胞外ドメインまたはその一部分、およびHIVgp140細胞外ドメインまたはその一部分からなる群より選択される免疫組成物。 - 請求項1〜5のいずれか一項に記載の免疫組成物において、
前記組み替えポリペプチドの前記リンカー領域は、10〜60個のアミノ酸からなる免疫粗組成物。 - 請求項6に記載の免疫組成物において、
前記組み替えポリペプチドの前記リンカー領域は、20〜50個のアミノ酸からなる免疫組成物。 - 請求項7に記載の免疫組成物において、
前記組み替えポリペプチドの前記リンカー領域は、25〜40個のアミノ酸からなる免疫組成物。 - 請求項6に記載の免疫組成物において、
前記リンカー領域は、GASSAGNTNSGGSTTTITNNNSGTNSSST、GSASSTNSGGSNNSASTTPTTSVTPAKPTSQと、QSAAASNPSTGSGSTATNNSNSTSSNSNASとからなる群より選択されるアミノ酸配列を含む免疫組成物。 - 請求項1〜9のいずれか一項に記載の免疫組成物において、
前記組み替えポリペプチドの前記オリゴマー化領域(OMD)は、二量体化、三量体化または四量体化領域である免疫組成物。 - 請求項10に記載の免疫組成物において、
前記オリゴマー化領域は、GCN4−に基づく二−、三−、または四量体化、T4フィブリチン(フォルドン)の前記C−末端領域配列またはその機能性部分あるいはその類似体(C−末端27〜30残基)およびニワトリ軟骨基質(CART)タンパク質の前記可溶性三量体化領域から選択される免疫組成物。 - 請求項1〜11のいずれか一項に記載の免疫組成物において、
前記組み替えポリペプチドは、さらに、N−末端抗原領域の場合は、C−末端キャッピング配列、またはC−末端抗原領域の場合は、N−末端キャッピング配列を含む免疫組成物。 - 請求項1〜12のいずれか一項に記載の免疫組成物において、
前記バクテリアは、非病原性バクテリアである免疫組成物。 - 請求項13に記載の免疫組成物において、
前記バクテリアは、食品品質等級のバクテリアである免疫組成物。 - 請求項14に記載の免疫組成物において、
前記バクテリアは、ラクトコッカス、ラクトバチルス、バチルスおよびミコバクテリウム亜種からなる群より選択される免疫組成物。 - 請求項1〜15のいずれか一項に記載の免疫組成物において、
前記ペプチドグリカン担体粒子は、少なくとも第1の病原体に由来する表面露出ポリペプチドまたはその抗原部分を含むポリペプチドの第1のオリゴマーと第2の病原体に由来する表面露出ポリペプチドまたはその抗原部分を含むポリペプチドの第2のオリゴマーとを非共有結合的に設けられている免疫組成物。 - 請求項1〜16のいずれか一項に記載された免疫組成物の製造方法であって、
a)請求項1〜12のいずれか一項に記載の組み替えポリペプチドを用意する工程であって、前記ポリペプチドをエンコードする発現ベクターを含む宿主細胞を適切な培地中に培養し、前記ポリペプチドの発現を許容し、かつ前記ポリペプチドを単離することを含む工程と、
b)グラム陽性菌から得られる不活化バクテリア様粒子(BLP)を用意する工程と、
c)一種以上の前記ポリペプチドの前記BLPへの非共有結合を許容して、病原体由来の表面露出ポリペプチドまたはその抗原部分のオリゴマーを含み、ペプチドグリカン担体粒子に非共有結合的に結合された免疫複合体を形成する工程と、
d)前記免疫複合体を免疫組成物中に処方する工程と、を備える免疫組成物の製造方法。 - 請求項1〜16のいずれか一項に記載の免疫組成物を備える、病原体により引き起こされる疾患の治療剤または予防剤。
- 請求項18に記載の疾患の治療剤または予防剤において、
前記病原体はウィルスである、疾患の治療剤または予防剤。 - 請求項19に記載の疾患の治療剤または予防剤において、
前記ウィルスは、インフルエンザ・ウイルス、動物コロナウイルス、ヒト呼吸器コロナウイルス、ヒト免疫不全ウイルス(HIV)、パラミクソウイルスである、疾患の治療剤または予防剤。 - 請求項19に記載の疾患の治療剤または予防剤において、前記ウィルスは、呼吸器合胞体ウイルス(RSV)またはメタ肺炎ウイルスである、疾患の治療剤または予防剤。
- 組み替えポリペプチドであって、
A)少なくとも1つの表面露出ポリペプチド(例えば、病原性または腫瘍細胞由来)またはその抗原部分を含むN−またはC−末端抗原領域であって、前記抗原領域は、
B)オリゴマー化領域(OMD)に融合され、前記オリゴマー化領域は、
C)リンカー領域を介して、
D)単一コピーのLysM領域からなり、前記ポリペプチドの、グラム陽性菌から得られる不活化バクテリア様粒子(BLP)であるペプチドグリカン担体粒子への前記非共有結合性連結を仲介することが可能なペプチドグリカン結合領域(PBD)に融合され、かつ前記ポリペプチド全体として単一コピーのLysM領域のみを含み、
前記LysM領域は、L.lactis AcmAにおける3つのLysM領域のうちの1つと、少なくとも70%以上の配列同一性を有する組み替えポリペプチド。 - 請求項22に記載のポリペプチドをエンコードする核酸配列。
- 請求項23に記載の核酸配列を含むベクター。
- 請求項23に記載の核酸配列または請求項24に記載のベクターを含む宿主細胞。
- 請求項25に記載の宿主細胞において、
前記宿主細胞は、真核宿主細胞である宿主細胞。 - 請求項26に記載の宿主細胞において、
前記宿主細胞は哺乳動物宿主細胞である宿主細胞。
