Description
1−1.タンパク質発現水準測定
動脈硬化症と区別されるもやもや病だけの特異的なバイオマーカーを選別するために、血管撮影術によって区分されたもやもや病(MMD)の患者139人、頭蓋内動脈硬化性狭窄患者(ICAS)61人、正常群(control)68人からそれぞれ血清を採取した後、ELISA(enzyme−linked immunosorbent assays)キットを使用して下記のタンパク質の発現水準を測定した。
ADMA ELISA kit(catalog #K7828;Immundiagnostik)、
humans E−selectin ELISA kit(catalog #BEK−2089−2P;Biosensis)、
human Endostatin ELISA(catalog #DNST0;R&D systems)、
human lipoprotein−associated phospholipase A2(Lp−PLA2)ELISA kit(cat #SEA867Hu;Cloud−Clone Corp.)
ROC(receiver operating characteristic)カーブは、特定検査の性能を評価するためのモデルであって、敏感度と特異度の相関関係を表現したグラフである。グラフのX軸は、False positive rateを示し、Y軸は、True positive rateを示し、ROCカーブが左側の頂点に近く描かれるほど、classification性能に優れていることを意味する。
2−1.RNF213変異とcaveolin−1水準の相関関係評価
前記実施例1のように、血管撮影術によって区分されたもやもや病と動脈硬化性狭窄をカベオリン−1の血中水準で鑑別する以外に、もやもや病の遺伝的素因によってカベオリン−1の血中水準に差異があるかを確認するために、頭蓋内狭窄がある患者及び正常人から血清を採取し、カベオリン−1水準を確認した。すなわち、もやもや病と関連した遺伝子として明らかにされたRNF213(Ring finger 213)(GenBank accession number NM_001256071.1)変異群とカベオリン−1水準との関連性を調査した。

実施例2−1の検査性能を評価するために、STATA ver.13.1プログラムのロジスティック回帰分析(code:lroc、nograph)方法でROC curveを作成した。
カベオリン−1水準変化が、もやもや病の発病において、angiogenesis因子(VEGF、VEGFR2、endostatin)、endothelial dysfunction因子(ADMA、NO、nitrite、nitrate)の作用を媒介するかを評価するために、path analysisを行った。
Claims (11)
- カベオリン−1(caveolin−1)mRNAまたはタンパク質の発現水準を測定する製剤を含む、もやもや病の診断用組成物。
- 前記mRNA発現水準を測定する製剤は、カベオリン−1遺伝子に特異的に結合するアンチセンスオリゴヌクレオチド、プライマーまたはプローブを含むことを特徴とする請求項1に記載の診断用組成物。
- 前記タンパク質発現水準を測定する製剤は、カベオリン−1タンパク質に特異的に結合する抗体を含むことを特徴とする請求項1に記載の診断用組成物。
- 請求項1に記載の組成物を含む、もやもや病の診断用キット。
- 前記キットは、DNAマイクロアレイチップまたはタンパク質チップであることを特徴とする請求項4に記載の診断用キット。
- 個体の試料からカベオリン−1(caveolin−1)mRNAまたはタンパク質の発現水準を測定する段階を含む、もやもや病診断のための情報提供方法。
- 前記個体の試料からカベオリン−1発現水準を測定した結果、正常対照群より減少した場合、もやもや病であると判定する段階をさらに含むことを特徴とする請求項6に記載の情報提供方法。
- 前記カベオリン−1のmRNA発現水準を測定する段階は、逆転写重合酵素反応(RT−PCR)、競争的逆転写重合酵素反応(Competitive RT−PCR)、リアルタイム逆転写重合酵素反応(Realtime RT−PCR)、RNase保護分析法(RPA;RNase protection assay)、またはノーザンブロッティング(Northern blotting)を利用することを特徴とする請求項6に記載の情報提供方法。
- 前記カベオリン−1のタンパク質発現水準を測定する段階は、抗原−抗体反応を利用することを特徴とする請求項6に記載の情報提供方法。
- 前記抗原−抗体反応は、ELISA(enzymelinked immunosorbent assay)、ウェスタンブロット、放射線免疫分析(RIA:Radioimmunoassay)、放射免疫拡散法(radioimmunodiffusion)、オクタロニー(Ouchterlony)免疫拡散法、ロケット(rocket)免疫電気泳動、組織免疫染色、免疫沈降分析法(Immunoprecipitation assay)、補体固定分析法(Complement Fixation Assay)、FACS(Fluorescence activated cell sorter)またはマイクロアレイ(microarray)分析法で行うことを特徴とする請求項9に記載の情報提供方法。
- 前記試料は、血液、血清、血漿、唾液、リンパ液または小便であることを特徴とする請求項6に記載の情報提供方法。
