Description
[材料および方法]
上述したAlvarezら、2008に記載されているように、HIの仔ブタモデルを使用した。麻酔を施した1〜3日齢の仔ブタの左右頸動脈を閉塞し、吸入される酸素を30分間にわたり21%から10%まで低減することで、一時的にHI障害の誘発を行う。
・ビヒクル
・CBD(1mg/kg)
・CBD(1mg/kg)と、CB2拮抗薬であるAM630(1mg/kg)
・CBD(1mg/kg)と、5HT1A拮抗薬であるWAY100635(0.1mg/kg)
・CBD(1mg/kg)と、アデノシン受容体の非特異的拮抗薬であるカフェイン(10mg/kg)
[子ブタモデル]
図1のAおよびBは、低酸素虚血(HI)の終了後6時間に採取した脳組織を示したものである。図1のAは、擬似手術を行った仔ブタ群の脳切片をニッスル染色したものを、HIを受けた後にビヒクルもしくはCBD1mg/kg i.v.単独、またはCBD(1mg/kg)とCB2拮抗薬であるAM630(AM)とを併用、CBD(1mg/kg)と5HT1A拮抗薬であるWAY100635(WAY)とを併用、およびCBD(1mg/kg)とアデノシン拮抗薬であるカフェイン(CAF)とを併用処置した仔ブタ群のそれぞれについて採取した脳切片をニッスル染色したものと比較している。
図2は、ラットの脳の磁気共鳴画像(MRI)を示すものであり、この画像によると、HI後から7日目のHI+VEHおよびHI+CBDの損傷の容積は同様であることが明らかになり、脳損傷の重症度は両群ともに同様に高度であることが示唆された。
仔ブタモデルにおいて、CBDを用いることで、HIによって引き起こされる脳の壊死領域の量を減少させることが可能であることが示された。
[材料および方法]
鎮静状態で人工呼吸器につながれた仔ブタ(日齢1〜2)に、HI脳損傷を経験させた(低酸素−FiO2 10%+両側頸動脈を30分間圧迫)。
神経保護は、CBDと低体温とを組み合わせて処置した動物群で、最も良好であることがわかった(P<0.05)。
HI後のCBD投与により、神経細胞および星状膠細胞が保護され、小膠細胞の活性化が調整される。

Claims (3)
- 低体温療法を組合せて、新生児低酸素性虚血性脳症(NHIE)の治療に使用するための、植物由来カンナビノイドのカンナビジオール(CBD)を含む、医薬組成物。
- 前記CBDが植物抽出物の形態である請求項1に記載の医薬組成物。
- 前記CBDが純粋な形態または単離された形態である請求項1に記載の医薬組成物。
