U87細胞株(GBM cell)にゴシポール(Gossypol)を0.5、1、5、10および50μMの濃度で72時間処理した。(図1)
U87細胞株を96ウェルプレートに接種し、37℃で24時間培養した後、下記表1に記載の実施例および比較例の薬学的組成物を処理し、MTS薬剤を20μl/ウェルの濃度で処理した後、37℃で4時間培養した。以後、490nmで吸光度を測定して、何らかの処理を施していない対照群対比の吸光度の変化を細胞生存率で計算して、その結果を図2にグラフで示した。
U87細胞株を2重量%1xB27、0.02重量%のbFGF20ng/ml、0.02重量%のEGF20ng/ml、および50U/mlペニシリン−50mg/mlストレプトマイシン(100x、Gibco、Invitrogen Korea、Seoul、South Korea)が添加されたDMEM/F−12培地で腫瘍球(tumor sphere)培養した。以後、細胞を96ウェルプレートに10細胞/セルで接種し、ゴシポール1μM、フェンホルミン10μM、ゴシポール1μMおよびフェンホルミン10μMの混合物を処理した後、37℃で3週間培養した。U87細胞の形状とサイズを確認するために、得られた細胞培養物を倒立位相差顕微鏡(inverted phase−contrast microscope)(1x71 Inverted Microscope;Olympus、Tokyo、Japan)で観察し、デジタルカメラ(DP70 Digital Microscope Camera;Olympus)で撮影して、その写真を図3に示した。また、各処理ごとに神経球の半径の変化を図4にグラフで示し、神経球の形成程度を図5にグラフで示した。
U87細胞(2x105細胞/ウェル)を成長培地0.1mlに浮遊させた後、ゴシポール1μM、フェンホルミン10μM、ゴシポール1μMおよびフェンホルミン10μMの混合物を処理し、トランスウェルチャンバ(8mmポアサイズ;Corning Glass)の上部ウェルに細胞をローディングした。ただし、前記トランスウェルチャンバの下部は成長培地0.5mlで満たされており、上部は成長因子の減少したマトリゲル(Matrigel)(Corning Matrigel Matrix)8.4mg/mlで前処理コーティングされた。細胞を前記チャンバ内で37℃で48時間培養した後、フィルタの上面で非浸潤細胞は綿棒で除去し、フィルタの下面に移動した細胞を固定した後、Diff−Quick kit(Fisher)で染色した後、得られた細胞培養物を位相差顕微鏡(phase−contrast microscope)(Olympus)で観察して、その写真を図6に示した。浸潤能は、ウェルあたり10顕微鏡フィールド(microscopic field)で細胞数を測定した。
動物実験に使用されるゴシポールおよびフェンホルミンはそれぞれ、DMSOとPBSに溶解して準備した。ゴシポールおよびフェンホルミン併用投与用組成物は、10重量%DMSOと10重量%クレモフォール(cremophor)−PBSが混合した溶液に溶解して準備した。
Claims (6)
- ゴシポール及びフェンホルミンを含む、膠芽腫の治療用薬学的組成物。
- ゴシポールとフェンホルミンが、1:1〜100の重量比で含まれる、請求項1に記載の薬学的組成物。
- ゴシポールとフェンホルミンが、1:2〜20の重量比で含まれる、請求項1又は2に記載の薬学的組成物。
- ゴシポールが、0.5〜50μMの量で含まれる、請求項1〜3のいずれか一項に記載の薬学的組成物。
- フェンホルミンが、10〜1000μMの量で含まれる、請求項1〜4のいずれか一項に記載の薬学的組成物。
- 前記膠芽腫の成長抑制は、脳癌幹細胞の維持の抑制、脳癌幹細胞の悪性化の抑制、または脳癌幹細胞の浸潤活性の抑制を含む、請求項1〜5のいずれか一項に記載の薬学的組成物。
