Description
Claims (18)
- 繊維芽細胞から間葉系幹細胞(MSC)を区別する方法であって、
繊維芽細胞からMSCを区別するように、MSC上で発現されるマーカーEphA2を使用して繊維芽細胞からMSCを単離することを含み、
MSCおよび繊維芽細胞は胎盤関連組織に由来する細胞の初代培養からである、方法。 - 胎盤関連組織が、羊膜、絨毛膜板(chorionic disk)、絨毛膜(chorionic membrane)および臍帯からなる群から選択される、請求項1に記載の方法。
- 単離する工程が、抗体ベースの、またはヌクレオチドベースの単離方法によって行われる、請求項1に記載の方法。
- 単離した胎盤関連組織からMSCおよび繊維芽細胞を回収すること;
初代培養を調製するために培養培地においてMSCおよび繊維芽細胞を培養すること
の予備段階をさらに含む、請求項1に記載の方法。 - 抗体ベースの単離方法が抗体ベースの磁気細胞選別または抗体ベースのフローサイトメトリーである、請求項3に記載の方法。
- ヌクレオチドベースの単離方法がヌクレオチドベースのフローサイトメトリーである、請求項3に記載の方法。
- 間葉系幹細胞(MSC)集団の純度を増加させる方法であって、
MSC上で発現されるマーカーEphA2を使用してMSCを単離および回収すること、ここでMSCは胎盤関連組織に由来する細胞の初代培養からであり;および
MSCを培養すること
を含む、方法。 - 胎盤関連組織が、羊膜、絨毛膜板、絨毛膜および臍帯からなる群から選択される、請求項7に記載の方法。
- 単離する工程が、抗体ベースの、またはヌクレオチドベースの単離方法によって行われる、請求項7に記載の方法。
- 胎盤関連組織に由来する細胞がMSCのための培養培地において培養される、請求項7に記載の方法。
- MSC集団の純度が、単離工程後にP0で少なくとも95%である、請求項7に記載の方法。
- 抗体ベースの単離方法が抗体ベースの磁気細胞選別または抗体ベースのフローサイトメトリーである、請求項9に記載の方法。
- ヌクレオチドベースの単離方法がヌクレオチドベースのフローサイトメトリーである、請求項9に記載の方法。
- 炎症環境においてより応答性である間葉系幹細胞(MSC)集団を単離する方法であって、
MSC上で発現されるマーカーEphA2を使用してMSCを単離および回収することを含み、MSCは胎盤関連組織に由来する細胞の初代培養からであり、MSCの集団は炎症環境においてより応答性である、方法。 - 単離する工程が、抗体ベースの、またはヌクレオチドベースの単離方法によって行われる、請求項14に記載の方法。
- MSC集団が、TNF−αシグナル伝達またはTNF−α−依存性シグナル伝達に対してより応答性である、請求項14に記載の方法。
- 抗体ベースの単離方法が抗体ベースの磁気細胞選別または抗体ベースのフローサイトメトリーである、請求項15に記載の方法。
- ヌクレオチドベースの単離方法がヌクレオチドベースのフローサイトメトリーである、請求項15に記載の方法。
