Description
[1] 細胞傷害性T細胞(CTL)の増殖及び抗体の産生を増強させる、Lactococcus lactis subsp.lactisである乳酸菌株又はその培養物若しくは処理物を含む、細胞傷害性T細胞(CTL)の増殖及び抗体の産生を増強するための経口摂取用組成物。
[2] Lactococcus lactis subsp.lactisである乳酸菌株がLactococcus lactis JCM5805である、[1]の経口摂取用組成物。
[3] 医薬組成物である、[1]又は[2]の経口摂取用組成物。
[4] 飲食品である、[1]又は[2]の経口摂取用組成物。
[5] ワクチンアジュバントである、[1]〜[4]のいずれかの経口摂取用組成物。
[6] 細胞傷害性T細胞(CTL)の増殖及び抗体の産生を増強させる、Lactococcus lactis subsp.lactisである乳酸菌株又はその培養物若しくは処理物を含む、ワクチンである抗原と併用される、該抗原に特異的な細胞傷害性T細胞(CTL)の増殖及び該抗原に特異的な抗体の産生を増強するための、[1]〜[5]のいずれかの経口摂取用組成物。
[7] ワクチンである抗原を投与する被験体に、事前に経口摂取させ、該抗原に特異的な細胞傷害性T細胞(CTL)の増殖及び該抗原に特異的な抗体の産生を増強する、[6]の経口摂取用組成物。
[8] 抗原が感染症の原因となる細菌、ウイルス若しくはリケッチアに特異的な抗原、又はがん特異的抗原である、[6]又は[7]の経口摂取用組成物。
[9] 7〜20日間、1日1〜3回経口摂取させる[1]〜[8]のいずれかの経口摂取用組成物。
[10] [1]〜[9]のいずれかの経口摂取用組成物、及び抗原を含む、該抗原に特異的な細胞傷害性T細胞(CTL)の増殖及び該抗原に特異的な抗体の産生を増強するためのキット。
[11] 感染症の予防又は治療に用いられる、[10]のキット。
[12] がんの予防又は治療に用いられる、[10]のキット。
6週齢メスのC57BL/6Jマウス(チャールズリバー社)を使用した。1週間馴化させた後、体重を基にコントロール群(n=8)とJCM5805群(n=8)の2群にマウスを群分けした。コントロール群には標準食であるAIN-93Gを摂取させ、JCM5805群にはLactococcus lactis JCM5805株加熱死菌体を1 mg/day/mouse含んだAIN-93Gを摂取させた。温度25 ± 1 ℃、湿度60 ± 15%、明暗周期12時間に保たれた無菌環境の室内で1ケージあたり1匹のマウスを飼育した。
JCM5805株を14日間混餌投与した後、抗原の注入を行った。抗原注入後もJCM5805株の摂取は解剖時まで継続的に行われた。抗原にはInfluenza PA Peptide(MBL社製)を使用した。10 mg/ml H-2DbInfluenza PA Peptide(MBL社製)を1μl、10 mg/ml I-Ab HBc Helper Peptide(MBL社製)を12μl、Complete Freund’s Adjuvant(SIGMA社製)を100μl、PBS を87μl混合した後にエマルジョン化したもの200μlをマウス腹腔内へ投与した。抗原投与12日後にマウスを安楽殺し、脾臓及び血液を回収した。
抗原投与スケジュールを図1に示す。
回収した脾臓から細胞調製液を作製し、1個体あたり1 × 106 cellsの脾臓細胞調製液を用いた。細胞の染色には、CD8-FITC(KT15)(MBL社製)、H-2DbInfluenza PA Tetramer-PE(MBL社製)、7-AAD(BD Pharmingen社製)、CD4-APC(RM4-5)(eBioscience社製)、CD3e-APC-Cy7(145-2C11)(BD Pharmingen社製)を使用した。測定にはFACS Cant II(BD Biosciences社製)を用い、解析にはFCS Express Software (De Novo Software社製)を用いた。7-AAD- CD3+ CD8+の細胞をCD8+ T細胞と定義し、CD8+ T細胞の内Influenza PA Tetramer+ (Tet+)の割合、すなわち抗原特異的細胞傷害性T細胞(CTL)の割合を解析した。
回収した血液1 mlを遠心(4 ℃、30分、2000 rpm)し、上清(血漿)を回収した。血漿中の抗原特異的Whole IgGの検出にはELISA法を用いた。H-2Db Influenza PA Peptide(MBL社製)を終濃度100μg/mlになるように0.05 M Sodium Bicarbonate(pH 9.6)で調製した後、調製した抗原100μlを96 wellプレートに添加、4 ℃、over nightでインキュベートした。PBS-T(0.05% Tween20含有PBS)で3回洗浄を行い、Assay Diluent buffer(50 mM Tris、0.14 M NaCl、1% BSA、0.05% Tween20)200μlを添加後、室温で2時間インキュベートした。PBS-Tで3回洗浄を行い、100倍希釈した血漿100μlを添加し、室温で2時間インキュベートした。PBS-Tで3回洗浄を行い、Assay Diluent で1000倍希釈したHRP標識二次抗体(Bethyl社製)100μlを添加し、室温で2時間インキュベートした。PBS-Tで5回洗浄を行い、TMB substrate(eBioscience社製)100μlを添加し、室温で15分インキュベートした。Stop buffer(1 M H3PO4)100μlを添加し、プレートリーダーを用いて450 nmの吸光度を測定した。
Claims (7)
- Lactococcus lactis subsp.lactis JCM5805である乳酸菌株生菌体又は死菌体を有効成分として含む、ワクチンである抗原と併用される、該抗原に特異的な細胞傷害性T細胞(CTL)の増殖及び該抗原に特異的な抗体産生の増強用組成物。
- ワクチンである抗原を投与する被験体に、事前に経口摂取させ、該抗原に特異的な細胞傷害性T細胞(CTL)の増殖及び該抗原に特異的な抗体の産生を増強する、請求項1記載の組成物。
- 抗原が感染症の原因となる細菌、ウイルス若しくはリケッチアに特異的な抗原、又はがん特異的抗原である、請求項1又は2に記載の組成物。
- 7〜20日間、1日1〜3回経口摂取させる請求項1〜3のいずれか1項に記載の組成物。
- 請求項1〜4のいずれか1項に記載の組成物、及び抗原を含む、該抗原に特異的な細胞傷害性T細胞(CTL)の増殖及び該抗原に特異的な抗体の産生を増強するためのキット。
- 感染症の予防又は治療に用いられる、請求項5記載のキット。
- がんの予防又は治療に用いられる、請求項5記載のキット。
