Description
本出願は、2014年3月6日に出願された米国仮特許出願第61/948,792号明細書の利益を主張するものであり、この仮特許出願の開示は、その内容全体が参照により本明細書に組み込まれるものとする。
本発明は、国立衛生研究所(National Institute of Health)からの助成金第1PO1AG034906号の下で、政府の支援によりなされた。政府は本発明に対して一定の権利を有する。
「AL」は、自由に(ad lib)を意味する。
「DSS」は、差動ストレス感作を意味する。
「DXR」は、ドキソルビシンを意味する。
「FMD」は、絶食模倣食を意味する。
「KB」は、ケトン体を意味する。
「mTOR」は、哺乳動物のラパマイシン標的を意味する。
「PKA」は、プロテインキナーゼAを意味する。
「Rapa」は、ラパマイシンを意味する。
「STS」は、短期間絶食を意味する。
「TVD」は、トポテカン+ビンクリスチン+ドキソルビシンを意味する。
本発明の方法は、主として、in vitroおよびin vivoのマウスモデルで試験した。STSおよびFMDはまた、種々の臨床試験で試験されており、2つの食餌介入の安全性および実現可能性が示されている。FMD制限食は、DSSとDSRの両方を誘発する際にSTSと同様に効果的であることが示されている。
STSおよび化学療法剤と併用したmTORキナーゼ阻害剤ラパマイシンを、癌増殖を遅延させ、生存率を改善するように、動物、ヒト被験体、または癌細胞に投与した。試験した化学療法剤は、アントラサイクリン系ドキソルビシン単剤、または2つの他の化学療法剤、トポテカンおよびビンクリスチンとの併用(TVD)としたが、これらは、癌を有する成人および小児を処置するために通常使用されるものである。
Claims (7)
- 癌を処置するための制限食パッケージであって、当該制限食パッケージは、被験者に第1の期間に提供されるカロリー制限食を含み、前記カロリー制限食は1日当たり最大で1000kcalであり、前記カロリー制限食が前記被験者に提供される前、その期間、またはその後にキナーゼ阻害剤が前記被験体に投与され、前記カロリー制限食が短期間絶食(STS)であり、前記キナーゼ阻害剤がラパマイシンであることを特徴とする制限食パッケージ。
- 請求項1に記載の制限食パッケージにおいて、前記第1の期間が3〜10日であることを特徴とする制限食パッケージ。
- 請求項1に記載の制限食パッケージにおいて、前記キナーゼ阻害剤が、前記第1の期間中に投与されることを特徴とする制限食パッケージ。
- 請求項1に記載の制限食パッケージにおいて、前記キナーゼ阻害剤が、前記第1の期間の後であるがその1週間以内に投与されることを特徴とする制限食パッケージ。
- 請求項1に記載の制限食パッケージにおいて、前記第1の期間後に前記被験体に化学療法剤が投与されることを特徴とする制限食パッケージ。
- 請求項5に記載の制限食パッケージにおいて、前記化学療法剤による処置が開始した後の第2の期間、カロリー制限食が前記被験体に与えられることを特徴とする制限食パッケージ。
- 請求項1に記載の制限食パッケージにおいて、前記被験体が黒色腫または神経芽細胞腫または乳癌を有することを特徴とする制限食パッケージ。
