Description

i.化学基X2を有する連結分子であって、化学基X2が、表面の化学基Y1と反応して化学基Y2を含む反応生成物を形成するのに適したものであり、化学基Y2が、分析物認識分子と反応するのに適したものである、連結分子と、
ii.分析物認識分子と
を同時に含む、溶液に関する。
a.化学基X2を有する連結分子を含む第1溶液であって、化学基X2が、表面の化学基Y1と反応して化学基Y2を含む反応生成物を形成するのに適したものであり、化学基Y2が、分析物認識分子と反応するのに適したものである、第1溶液と、
b.分析物認識分子を含む第2溶液と
を同時に含み、
第1溶液と第2溶液とが、ひとたび混ざり合えば、
・少なくとも1μMに等しく最大でも1mMに等しい濃度の連結分子と
・少なくとも1×10−12mol/lに等しい、好ましくは1×10−8mol/lに等しい濃度の分析物認識分子と
を含む溶液を形成するようになっている、パーツのキット。
a1.裸出表面を提供するステップと、
a2.少なくとも1つの第1分子種を含む第1組成物に裸出表面を接触させるステップと
を含み得、少なくとも1つの第1分子種は、化学基Y1と、少なくとも1つの第1分子種を裸出表面に付着させるのに適した化学基X1とを含む。
・X1aは、−SiZx(CH3)y(式中、Zは、Cl、OCH3およびOCH2CH5から構成される群から選択され、y=0、1または2であり、x=3−yである)、−COOH、−PO(OH)2、−NH2、
から構成される群から選択され、式中、*はSi原子との結合点を指し示し、
・X1bは−SHまたは−SeHであり、
・X1cは、−CH=CH2および−C≡CHから構成される群から選択され、
ここで、X1がX1aである場合、裸出表面はOH基(X1aに対して反応性)を含み、
ここで、X1がX1bである場合、裸出表面は金属表面であり、
ここで、X1がX1cである場合、基Y1は−N3ではなく、裸出表面はSi−H基を含む。
・Y1が−N3である場合、X2は、歪みを有するかまたは有さないアルキンであり、アルキンが歪みを有さない場合、ステップbは、歪みを有さないアルキンと−N3との反応を可能にするのに適した触媒の存在下で行われ、
・Y1が、歪みを有するかまたは有さないアルキンである場合、X2は−N3であり、アルキンが歪みを有さない場合、ステップbは、歪みを有さないアルキンと−N3との反応を可能にするのに適した触媒の存在下で行われ、
・Y1が−COOHである場合、X2はカルボジイミドかまたはジアゾ基=N2かのどちらかである。
i.化学基X2を有する連結分子であって、化学基X2が、表面の化学基Y1と反応して化学基Y2を含む反応生成物を形成するのに適したものであり、化学基Y2が、分析物認識分子と反応するのに適したものである、連結分子と、
ii.分析物認識分子と
を同時に含む、溶液に関する。
a.化学基X2を有する連結分子を含む第1溶液であって、化学基X2が、表面の化学基Y1と反応して化学基Y2を含む反応生成物を形成するのに適したものであり、化学基Y2が、分析物認識分子と反応するのに適したものである、第1溶液と、
b.分析物認識分子を含む第2溶液と
を同時に含み、
第1溶液と第2溶液とが、ひとたび混ざり合えば、
・少なくとも1μMに等しく最大でも1mMに等しい濃度の連結分子(7)と
・少なくとも1×10−12mol/lに等しい、好ましくは少なくとも1×10−8mol/lに等しい濃度の分析物認識分子(1)と
を含む溶液(6)を形成するようになっている、パーツのキットに関する。
i.化学基Y1で官能基化された表面であって、化学基Y1と連結分子とを反応させて分析物認識分子との反応に適する反応生成物を形成することによって分析物認識分子を固定するのに適した、表面、および
ii.官能基化表面に連結された変換器であって、分析物と固定された分析物認識分子との相互作用に際して可読信号出力を生成するのに適した、変換器
を含む。
i.先の態様において定義したデバイス、および
ii.表面に連結された変換器であって、分析物と固定された分析物認識分子との相互作用に際して可読信号出力を生成するのに適した、変換器
を含む。
Claims (5)
- 官能基化された表面に分析物認識分子を固定する方法であって、
a.−N 3 基で官能基化された表面を提供するステップ、および
b.前記官能基化された表面を、
i.アルキン基とN−ヒドロキシスクシンイミドエステルとを有し、式
を有する連結分子と、
ii.−NH 2 基を有する分析物認識分子との両方を含む溶液と接触させるステップを含み、
前記分析物認識分子を前記表面に固定するように、前記連結分子のN−ヒドロキシスクシンイミド基が、前記分析物認識分子の−NH 2 基と反応し、前記連結分子のアルキン基が、前記表面の−N 3 基と反応し、
前記溶液が、前記連結分子を1μMから1mMの濃度範囲で含有し、
前記分析物認識分子と前記連結分子を含む前記溶液が、ステップbの前に前記分析物認識分子と前記連結分子とを最大でも30分混合することによって調製される、方法。 - ステップbが、前記官能基化された表面への前記溶液の液滴の堆積を含む、請求項1に記載の方法。
- 前記官能基化された表面は、分析物と固定された前記分析物認識分子との相互作用に際して可読信号出力を生成するための変換器と連結される、請求項1に記載の方法。
- ステップbに続いて、前記官能基化された表面をブロッキング剤と接触させることをさらに含む、請求項1に記載の方法。
- 前記ブロッキング剤が、DBCO−PEG 5kDa、または5%ウシ血清アルブミンである、請求項4に記載の方法。
