Description
(A)−Y1−Y2−Y3−G−Y4−Y5−G−Y6−Y7−G−Y8−Y9−G−(配列番号1):
(但し、Gは、グリシンであり、Y1〜Y9は、任意のアミノ酸である)。
LASColの材料になるコラーゲンまたはアテロコラーゲンは、特に限定されず、周知のコラーゲンおよびアテロコラーゲンであればよい。
(i)高濃度の塩の存在下にて、コラーゲンまたはアテロコラーゲンと、酵素とを接触させる方法。
(ii)高濃度の塩と接触させた後の酵素と、コラーゲンまたはアテロコラーゲンとを接触させる方法。
(iii)低濃度の塩の存在下にて、コラーゲンまたはアテロコラーゲンと、酵素とを接触させる方法。
塩化ナトリウムの濃度が0mMと1500mMである50mMクエン酸緩衝液(pH3.0)を準備した。なお、当該水溶液の溶媒として、水を用いた。
Claims (6)
- LASColを含有する椎間板変性の治療剤であって、
前記LASColは、コラーゲンまたはアテロコラーゲンのトリプルヘリカルドメインの下記(A)に示されるアミノ末端のアミノ酸配列においてα1鎖のY 1 とY 2 との間の化学結合が切断されている若しくは、下記(B)に示されるアミノ末端のアミノ酸配列においてα2鎖のGとX 3 との間の化学結合が切断されているコラーゲンまたはアテロコラーゲンの分解物を含む椎間板変性の治療剤。
(A)−Y 1 −Y 2 −Y 3 −G−Y 4 −Y 5 −G−Y 6 −Y 7 −G−Y 8 −Y 9 −G−(配列番号1);
(但し、Gはグリシンであり、Y 1 〜Y 9 は、任意のアミノ酸である)
(B)−G−X 1 −X 2 −G−X 3 −X 4 −G−X 5 −X 6 −G−(配列番号2);
(但し、Gはグリシンであり、X 1 〜X 6 は、任意のアミノ酸である) - ハイドロゲル、ゼラチンゲル、キトサンゲル、ヒアルロン酸・コラーゲンハイドロゲル、ヒアルロン酸ポリマー、ヒアルロン酸・PEGポリマー、コラーゲン・ヒアルロン酸・PEGハイドロゲル、高純度アルギン酸ゲル、人体親和性のある溶媒の少なくとも一つをさらに含む請求項1に記載された椎間板変性の治療剤。
- コラーゲン及びアテロコラーゲンの少なくともいずれかのアミノ酸配列のN末端を切断する工程と、酵素を用いて切断しLASColを得る工程と、
前記LASColにハイドロゲル、ゼラチンゲル、キトサンゲル、ヒアルロン酸・コラーゲンハイドロゲル、ヒアルロン酸ポリマー、ヒアルロン酸・PEGポリマー、コラーゲン・ヒアルロン酸・PEGハイドロゲル、高純度アルギン酸ゲル、人体親和性のある溶媒の少なくとも一つを添加する工程を有し、
前記LASColは、コラーゲンまたはアテロコラーゲンのトリプルヘリカルドメインの下記(A)に示されるアミノ末端のアミノ酸配列においてα1鎖のY 1 とY 2 との間の化学結合が切断されている若しくは、下記(B)に示されるアミノ末端のアミノ酸配列においてα2鎖のGとX 3 との間の化学結合が切断されているコラーゲンまたはアテロコラーゲンの分解物を含む椎間板変性の治療剤の製造方法。
(A)−Y 1 −Y 2 −Y 3 −G−Y 4 −Y 5 −G−Y 6 −Y 7 −G−Y 8 −Y 9 −G−(配列番号1);
(但し、Gはグリシンであり、Y 1 〜Y 9 は、任意のアミノ酸である)
(B)−G−X 1 −X 2 −G−X 3 −X 4 −G−X 5 −X 6 −G−(配列番号2);
(但し、Gはグリシンであり、X 1 〜X 6 は、任意のアミノ酸である) - LASColを含有する椎間板細胞培養材であって、
前記LASColは、コラーゲンまたはアテロコラーゲンのトリプルヘリカルドメインの下記(A)に示されるアミノ末端のアミノ酸配列においてα1鎖のY 1 とY 2 との間の化学結合が切断されている若しくは、下記(B)に示されるアミノ末端のアミノ酸配列においてα2鎖のGとX 3 との間の化学結合が切断されているコラーゲンまたはアテロコラーゲンの分解物を含む椎間板細胞培養材。
(A)−Y 1 −Y 2 −Y 3 −G−Y 4 −Y 5 −G−Y 6 −Y 7 −G−Y 8 −Y 9 −G−(配列番号1);
(但し、Gはグリシンであり、Y 1 〜Y 9 は、任意のアミノ酸である)
(B)−G−X 1 −X 2 −G−X 3 −X 4 −G−X 5 −X 6 −G−(配列番号2);
(但し、Gはグリシンであり、X 1 〜X 6 は、任意のアミノ酸である) - ハイドロゲル、ゼラチンゲル、キトサンゲル、ヒアルロン酸・コラーゲンハイドロゲル、ヒアルロン酸ポリマー、ヒアルロン酸・PEGポリマー、コラーゲン・ヒアルロン酸・PEGハイドロゲル、高純度アルギン酸ゲル、人体親和性のある溶媒の少なくとも一つをさらに含む請求項4に記載された椎間板細胞培養材。
- コラーゲン及びアテロコラーゲンの少なくともいずれかのアミノ酸配列のN末端を切断する工程と、酵素を用いて切断しLASColを得る工程と、
前記LASColにハイドロゲル、ゼラチンゲル、キトサンゲル、ヒアルロン酸・コラーゲンハイドロゲル、ヒアルロン酸ポリマー、ヒアルロン酸・PEGポリマー、コラーゲン・ヒアルロン酸・PEGハイドロゲル、高純度アルギン酸ゲル、人体親和性のある溶媒
の少なくとも一つを添加する工程を有し、
前記LASColは、コラーゲンまたはアテロコラーゲンのトリプルヘリカルドメインの下記(A)に示されるアミノ末端のアミノ酸配列においてα1鎖のY 1 とY 2 との間の化学結合が切断されている若しくは、下記(B)に示されるアミノ末端のアミノ酸配列においてα2鎖のGとX 3 との間の化学結合が切断されているコラーゲンまたはアテロコラーゲンの分解物を含む椎間板細胞培養材の製造方法。
