Description
本発明はコール酸及び/又はそのエステルを含
む培地から微生物が蓄積する3α,7α−ジヒドロ
キシ−12−ケト−5β−コラン酸及び/又はその
エステルを得る方法に関し、さらに詳しくはコー
ル酸及び/又はそのエステルを基質として3α,
7α−ジヒドロキシ−12−ケト−5β−コラン酸及
び/又はそのエステルを生産するアルスロバクタ
ー属、ブレビバクテリウム属、コリネバクテリウ
ム属又はノカルデイア属に属する細菌を、コール
酸及び/又はそのエステルを含む栄養培地で培養
して3α,7α−ジヒドロキシ−12−ケト−5β−コ
ラン酸及び/又はそのエステルを生成せしめ、こ
れを採取することを特徴とする3α,7α−ジヒド
ロキシ−12−ケト−5β−コラン酸及び/又はそ
のエステルの製造方法に関する。
従来、コール酸を基質として3α,7α−ジヒド
ロキシ−12−ケト−5β−コラン酸を微生物の代
謝産物として得る方法は種々知られている。例え
ば、早川らによるストレプトマイセス・ゲラチカ
ス1164菌株を用いる方法〔ジヤーナル・オブ・バ
イオケミストリー(日本)、第44巻第2号第109〜
113頁(1957年)及びプロシーデイングス・オ
ブ・ジヤパン・アカデミー、第32巻第519〜522頁
(1956年)〕;長谷川らによるアスペルギルス・シ
ンナモメウスHUT2026菌株を用いる方法〔ヒロ
シマ・ジヤーナル・オブ・メデイカル・サイエン
ス、第8巻第3号第277〜283頁(1959年)〕;及び
菊池らによるスタフイロコツカス・エピデルミデ
イスH−1菌株を用いる方法〔ジヤーナル・オ
ブ・バイオケミストリー、第72巻第1号第165〜
172頁(1972年)〕などである。
本発明者らは微生物の代謝産物として3α,7α
−ジヒドロキシ−12−ケト−5β−コラン酸及
び/又はそのエステルを有利に得るためその基質
としてコール酸及び/又はそのエステルを資化す
る微生物のスクリーニングを長期間行なつてきた
結果、アルスロバクター属、ブレビバクテリウム
属、コリネバクテリウム属又はノカルデイア属に
属する特定の細菌がコール酸及び/又はそのエス
テルを基質として3α,7α−ジヒドロキシ−12−
ケト−5β−コラン酸及び/又はそのエステルを
生産すること、しかもコール酸及び/又はそのエ
ステルを可溶化することなく懸濁状もしくは結晶
状のままで上記のコラン酸及び/又はそのエステ
ルに変換しうることを見出し、本発明を完成する
に至つた。
本発明者らが得たコール酸及び/又はそのエス
テルを基質として3α,7α−ジヒドロキシ−12−
ケト−5β−コラン酸及び/又はそのエステルを
生産する能力を有するアルスロバクター属、ブレ
ビバクテリウム属、コリネバクテリウム属又はノ
カルデイア属に属する細菌としては、アルスロバ
クターCA−35(Arthrobacter CA−35)菌株
(微工研菌寄第5145号)、アルスロバクターCA−
35−A589−29−32(Arthrobacter CA−35−
A589−29−32)菌株(微工研菌寄第5522号)、ブ
レビバクテリウムCA−6(Brevibacterium CA
−6)菌株(微工研菌寄第5144号)、コリネバク
テリウムCA−53(Corynebacterium CA−53)菌
株(微工研菌寄第5532号)及びノカルデイアCA
−173(Nocardia CA−173)菌株(微工研菌寄第
5531号)があり、それぞれの菌学的性質を列挙す
ると次表のとおりである。

上記の表に示した菌学的性質に基づき、アルス
ロバクターCA−35菌株、アルスロバクターCA−
35−A589−29−32菌株、ブレビバクテリウムCA
−6菌株、コリネバクテリウムCA−53菌株及び
ノカルデイアCA−173菌株の同定をバージエイ
ズ・マニユアル・オブ・デイターミネイテイブ・
バクテリオロジー第7版及び第8版に基づき行な
つた。アルスロバクターCA−35菌株はその形態、
グラム染色などの顕微鏡的所見及び生理学的性質
などからアルスロバクター属に属する菌であると
同定された。アルスロバクターCA−35菌株は黄
色〜クリーム色の色素を生産するが、アルスロバ
クター属に属する菌で色素生産菌としてはアルス
ロバクター・オキシダンス、アルスロバクター・
オーレツセンス及びアルスロバクター・ウレアフ
アシエンスが存在する。しかし、アルスロバクタ
ー・オキシダンス及びアルスロバクター・オーレ
ツセンスは通常グラム染色が陰性であること及び
デンプンの加水分解性を有していることより、ア
ルスロバクターCA−35菌株とは異なる。また、
アルスロバクター・ウレアフアシエンスは通常グ
ラム染色が陰性であること及び硝酸塩の還元性を
有していないことより、アルスロバクターCA−
35菌株とは異なる。さらにアルスロバクターCA
−35菌株はアルスロバクター・シンプレツクスに
近縁の菌であると同定されるが、アルスロバクタ
ー・シンプレツクスIAM1660菌株はコール酸塩
の10g/濃度の培地で生育しないので、アルス
ロバクターCA−35菌株はコール酸塩に対する態
度において明らかにアルスロバクター・シンプレ
ツクスIAM1660菌株と異なる。従つて、アルス
ロバクターCA−35菌株はアルスロバクター属に
属するいずれの標準種の菌とも異なつていること
より、アルスロバクター属に属する新菌種を構成
する細菌であると判断した。アルスロバクター
CA−35−A589−29−32菌株はアルスロバクター
CA−35菌株をN−メチル−N′−ニトロ−N−ニ
トロソグアニジンと接触させることにより得られ
た突然変異菌株である。一般に突然変異菌株はそ
の親株と同じ種に属するものと考えられているこ
とより、アルスロバクターCA−35−A589−29−
32菌株はその親株と同じ新菌種に属するものと判
定した。ブレビバクテリウムCA−6菌株はその
グラム染色などの顕微鏡的所見及び生理学的性質
などからブレビバクテリウム属に属する細菌と同
定された。しかしながら、ブレビバクテリウム属
に属する細菌が周鞭毛を有しているのに対し、ブ
レビバクテリウムCA−6菌株は極単鞭毛を有す
るなど、ブレビバクテリウムCA−6菌株はブレ
ビバクテリウム属に属する細菌とは若干異なる性
質を有する。コリネバクテリウムCA−53菌株は
その形態、グラム染色などの顕微鏡的所見及び生
理学的性質などからコリネバクテリウム・エクイ
に近縁の細菌であると同定された。ノカルデイア
CA−173菌株は顕微鏡的所見及び生理学的性質な
どからコリネバクテリウム属又はノカルデイア属
に属する細菌と思われるが、O−Fテストが酸化
的(オキシデイテイブ)であることよりコリネバ
クテリウム属に属する細菌とは明らかに異なる。
従つて、ノカルデイアCA−173菌株はノカルデイ
ア属に属する細菌であると判断した。
本発明の方法による3α,7α−ジヒドロキシ−
12−ケト−5β−コラン酸及び/又はそのエステ
ルの生産は、前記のコール酸及び/又はそのエス
テルを基質として3α,7α−ジヒドロキシ−12−
ケト−5β−コラン酸及び/又はそのエステルを
生産する細菌を、コール酸及び/又はそのエステ
ルを含む培地で培養することにより行なわれる。
コール酸エステルは好適にはコール酸のメチルエ
ステル、エチルエステル、プロピルエステル、ブ
チルエステルなどの低級アルキルエステルであ
る。コール酸及び/又はそのエステルの濃度は通
常約1〜500g/の範囲でよいが、生産される
3α,7α−ジヒドロキシ−12−ケト−5β−コラン
酸及び/又はそのエステルの収量、培養条件及び
操作性などの経済的観点から約10〜500g/の
範囲が好ましく、さらに10〜300g/の範囲が
より好ましい。培養方法は原則的には一般微生物
の培養で採用される方法と同じであるが、通常は
液体培地による振盪培養法又は通気撹拌培養法が
用いられる。培地としては上記のコール酸及び/
又はそのエステルを基質として3α,7α−ジヒド
ロキシ−12−ケト−5β−コラン酸及び/又はそ
のエステルを生産する細菌が資化利用できる栄養
源を含有するものであればよい。炭素源としては
コール酸及び/又はそのエステルの他に、アラビ
ノースなどのペントース類;グルコース、マンノ
ース、フラクトース、ガラクトースなどのヘキソ
ース類;マルトースなどの二糖類;デンプン分解
物、糖アルコール類、グリセリンなどの多価アル
コール類;又はポリペプトン、ペプトン、肉エキ
ス、麦芽エキス、コーンステイープリカー、酵母
エキス、各種アミノ酸などが用いられる。また窒
素源としては、例えば硫酸アンモニウム、塩化ア
ンモニウム、燐酸アンモニウム、硝酸アンモニウ
ム、硝酸ナトリウム、硝酸カリウムなどが用いら
れる。また、この他に燐酸水素2カリウム、燐酸
2水素カリウム、硫酸マグネシウムなどの無機塩
が添加される。培養条件に特徴はないが、通常25
〜35℃で6時間〜8日間振盪培養又は通気撹拌培
養を行なう。
このようにして培養液中に蓄積された3α,7α
−ジヒドロキシ−12−ケト−5β−コラン酸及
び/又はそのエステルを分離・採取するには、ま
ず培養液中の菌体、培養液中にほとんど不溶のま
ま存在している3α,7α−ジヒドロキシ−12−ケ
ト−5β−コラン酸及び/又はそのエステルをは
じめとするコール酸及び/又はそのエステルの酸
化生成物並びに未変換のコール酸及び/又はその
エステルを濾過又は遠心分離などにより分離取得
する。これに例えばクロロホルム−エタノール混
合液を加えてコール酸及び/又はそのエステルの
酸化生成物並びに未変換のコール酸及び/又はそ
のエステルを溶解したのち、濾過又は遠心分離な
どにより菌体を分離除去し、得られた濾液又は上
清を蒸発乾固する。得られたコール酸及び/又は
そのエステルの酸化生成物並びに未変換のコール
酸及び/又はそのエステルの混合物に、先の培養
濾液又は上清に例えば酢酸エチルを加えて残存す
るコール酸及び/又はそのエステルの酸化生成物
並びに未変換のコール酸及び/又はそのエステル
を抽出し、しかるのち酢酸エチルを溜去して得ら
れたものを合わせることにより、ほぼ完全に3α,
7α−ジヒドロキシ−12−ケト−5β−コラン酸及
び/又はそのエステルをはじめとするコール酸及
び/又はそのエステルの酸化生成物を採取し、未
変換のコール酸及び/又はそのエステルを回収す
ることができる。このようにして得られたコール
酸及び/又はそのエステルの酸化生成物並びに未
変換のコール酸及び/又はそのエステルの混合物
を例えばシリカゲルカラムに吸着させ、クロロホ
ルム−エタノール混合液により順次溶出させる。
このようにして3α,7α−ジヒドロキシ−12−ケ
ト−5β−コラン酸及びそのエステルを取得する
ことができる。
得られた3α,7α−ジヒドロキシ−12−ケト−
5β−コラン酸のエステルは常法により加水分解
することにより3α,7α−ジヒドロキシ−12−ケ
ト−5β−コラン酸とすることができる。
本発明の方法により得られる3α,7α−ジヒド
ロキシ−12−ケト−5β−コラン酸はフアン・ミ
ンロン還元反応に供することにより胆石溶解剤と
して有用なケノデオキシコール酸(CDCA)に容
易に変換できる。
以下実施例によつて本発明をさらに詳細に説明
する。
参考例 1
アルスロバクターCA−35−A589−29−32菌株
(微工研菌寄第5522号)の取得
培地1(組成:水酸化ナトリウム0.5%、コール
酸5.0%、ペプトン0.5%、酵母エキス0.5%、塩化
ナトリウム0.5%及び寒天1.5%)のスラントに生
育させたアルスロバクターCA−35菌株の1白金
耳を、予め試験管(内径21mm、長さ200mm)内に
準備した培地2(組成:水酸化ナトリウム1%、
コール酸10%、硝酸アンモニウム0.2%、燐酸2
水素カリウム0.2%、燐酸1水素カリウム0.5%、
硫酸マグネシウム・7水和物0.02%及び酵母エキ
ス0.01%)の10mlに植菌し、30℃で24時間振盪培
養した。この培養液の0.3mlを予め試験管(内径
21mm、長さ200mm)に準備した培地3(組成:水酸
化ナトリウム0.05%、コール酸0.5%、グルコー
ス0.5%、硝酸アンモニウム0.2%、燐酸2水素カ
リウム0.2%、燐酸1水素カリウム0.5%、硫酸マ
グネシウム・7水和物0.02%及び酵母エキス0.01
%)の10mlに加え、30℃で15〜16時間振盪培養し
た。ついで、この対数増殖期にある菌体を0.45μ
のメンブランフイルターで無菌的に濾過集菌し、
0.1M燐酸塩緩衝液(PH7.0)20mlで洗滌後、同じ
緩衝液25mlに懸濁させた。この菌懸濁液4mlを試
験管(内径21mm、長さ200mm)に入れ、これに終
濃度が50μg/mlになるようにN−メチル−N′−
ニトロ−N−ニトロソグアニジンを添加し、30℃
で45分間振盪することによつて突然変異処理を行
なつた。なお、この処理条件下でのアルスロバク
ターCA−35菌株の死滅率は約80%であつた。突
然変異処理を施した菌体を0.45μのメンブランフ
イルターで無菌的に濾過集菌し、0.1M燐酸塩緩
衝液(PH7.0)20mlで洗滌後、同じ緩衝液25mlに
懸濁させた。得られた菌懸濁液を滅菌した生理食
塩水で希釈し、それを培地4(組成:水酸化ナト
リウム0.1%、コール酸1.0%、硝酸アンモニウム
0.2%、燐酸2水素カリウム0.2%、燐酸1水素カ
リウム0.5%、硫酸マグネシウム・7水和物0.02
%、酵母エキス0.01%及び寒天1.5%)の寒天平
板上に300〜500個のコロニーを出現させるように
塗布したのち、30℃で4日間培養した。出現した
コロニー中の極小コロニーを単離し、培地5(組
成:ペプトン0.5%、酵母エキス0.5%、塩化ナト
リウム0.5%及び寒天1.5%)の寒天平板上で30℃
で24時間培養した。ここに出現したコロニーを培
地4の寒天平板上にレプリカし、30℃で20時間培
養した。この培地4で生育しないコロニーに対応
する上記の培地5のコロニーを培地6(組成:水
酸化ナトリウム0.1%、コール酸1.0%、ペプトン
0.5%、酵母エキス0.5%、塩化ナトリウム0.5%及
び寒天1.5%)のスラントにそれぞれ移し、30℃
で24時間培養した。このスラントから1白金耳の
菌体を予め試験管(内径21mm、長さ200mm)内に
準備した培地7(組成:水酸化ナトリウム0.5%、
コール酸5.0%、グルコース0.5%、硝酸アンモニ
ウム0.2%、燐酸2水素カリウム0.2%、燐酸1水
素カリウム0.5%、硫酸マグネシウム・7水和物
0.02%及び酵母エキス0.01%)の10mlに植菌し、
30℃で3日間振盪培養した。得られたそれぞれの
培養液中の生産物を薄層クロマトグラフイーで検
定し、目的とする3α,7α−ジヒドロキシ−12−
ケト−5β−コラン酸及び/又はその塩を選択的
に蓄積する1菌株を見出し、これをアルスロバク
ターCA−35−M−965−3菌株と命名した。以
後、この突然変異菌株の取得方法を総括して単に
NTG処理と称する。
アルスロバクターCA−35−M−965−3菌株を
親株とし、これにNTG処理を施すことにより
3α,7α−ジヒドロキシ−12−ケト−5β−コラン
酸及び/又はその塩を選択的に蓄積する3菌株を
見出し、これらの中の1菌株をアルスロバクター
CA−35−A589菌株と命名した。アルスロバクタ
ーCA−35−A589菌株のコロニーから2個の優良
菌株を単離し、これらの中の1菌株をアルスロバ
クターCA−35−A589−29−32菌株と命名した。
実施例 1
アルスロバクターCA−35−A589−29−32菌株
(微工研菌寄第5522号)を次に示す方法で培養し
た。コール酸5.0g、グルコース0.5g、硝酸アン
モニウム0.2g、燐酸2水素カリウム0.2g、燐酸
水素2カリウム0.5g、硫酸マグネシウム・7水
和物0.02g及び酵母エキス0.05gに水道水を加え
て容量を100mlに調整し、これを培地とした。こ
の培地を500ml容坂口フラスコに入れ、120℃で15
分間、蒸気殺菌を行なつた。予め上記の培地と同
じ培地で試験管振盪機にて2日間増殖させた種菌
10mlを上記の500ml容坂口フラスコ中の培地に接
種し、5日間振盪培養を行なつた。培地中におい
てコール酸は未溶解の状態であり、培養が進むに
つれて結晶形が明らかに異なつた3α,7α−ジヒ
ドロキシ−12−ケト−5β−コラン酸の結晶に変
化した。培養後、得られた培養液を遠心分離機に
かけ、菌体並びにコール酸の酸化生成物及び未変
換のコール酸を沈澱させ、上清と分離した。菌体
並びにコール酸の酸化生成物及び未変換のコール
酸からなる沈澱物にクロロホルム/エタノールの
1/1容量比の混合液100mlを加えて、コール酸
の酸化生成物及び未変換のコール酸を該混合液に
溶解させた。濾過により菌体を除去し、濾液を減
圧下で濃縮乾固することによりコール酸の酸化生
成物及び未変換のコール酸の混合物を得、これに
先の培養上清に酢酸エチル100mlを加えて抽出し、
この抽出液を濃縮乾固して得られたコール酸の酸
化生成物及び未変換のコール酸の混合物を合わせ
ることにより、5.2gの混合物を得た。この混合
物の極く一部を取り、これにメタノールを加えて
2%溶液とし、この溶液10μをミクロボンダパ
ツクC−18カラムを備えた高速液体クロマトグラ
フイー(米国ウオーターズ社製、HLC−GPC−
244型)に注入した。移動相としてPH4.0に調整し
た水/メタノールの30/70容量比の混合液を流速
1ml/分で流し、検出を屈折率方式で行なつたと
ころ第1図のクロマトグラムが得られた。このク
ロマトグラムにおけるピークA及びピークBは標
準品のピークと照合することにより各々3α,7α
−ジヒドロキシ−12−ケト−5β−コラン酸及び
コール酸のものと一致した。また、ピークAの部
分に相当する化合物を分取し、この化合物を下記
の方法で3α,7α−ジヒドロキシ−12−ケト−5β
−コラン酸であると同定した。
薄層クロマトグラフイーによる同定
上記の化合物の2%メタノール溶液の10μを
ベンゼン:酢酸エチル:酢酸(30:10:1)の組
成の展開溶剤を用いて10cm展開し、乾燥後、硫
酸:エタノール(1:1)の組成の発色試薬液を
噴霧し、115℃で15分間加熱すると標準品の3α,
7α−ジヒドロキシ−12−ケト−5β−コラン酸と
同一位置(Rf=0.48)に同色のスポツトが見られ
た。

アルスロバクターCA−35−A589−29−32菌株
(前述に同じ)を次に示す方法で培養した。コー
ル酸メチル5.0g、グルコース0.5g、硝酸アンモ
ニウム0.2g、燐酸2水素カリウム0.2g、燐酸水
素2カリウム0.5g、硫酸マグネシウム・7水和
物0.02g及び酵母エキス0.05gに水道水を加えて
容量を100mlに調整し、これを培地とした。この
培地を500ml容坂口フラスコに入れ、120℃で15分
間、蒸気殺菌を行なつた。予め上記の培地と同じ
培地で試験管振盪機にて2日間増殖させた種菌10
mlを上記の試験管中の培地に接種し、7日間振盪
培養を行なつた。培養後、得られた培養液を遠心
分離機にかけ、菌体並びにコール酸メチルの酸化
生成物及び未変換のコール酸メチルを沈澱させ、
上清と分離した。菌体並びにコール酸メチルの酸
化生成物及び未変換のコール酸メチルからなる沈
澱物(含水物)にクロロホルム/エタノールの
1/1容量比の混合液50mlを加えて、コール酸メ
チルの酸化生成物及び未変換のコール酸メチルを
該混合液に溶解させた。分液により菌体及び水を
除去し、得られた濾液を減圧下で濃縮乾固するこ
とにより、コール酸メチルの酸化生成物及び未変
換のコール酸メチルの混合物を4.4g得た。この
混合物の極く一部を取り、これにメタノールを加
えて2%溶液とし、この溶液10μをミクロボン
ダパツクC−18カラムを備えた高速液体クロマト
グラフイー(米国ウオーターズ社製、HLC−
GPC−244型)に注入した。移動相としてPH4.0に
調整した水/メタノールの20/80容量比の混合液
を流速1ml/分で流し、検出を屈折率方式で行な
つたところ第2図のクロマトグラムが得られた。
このクロマトグラムにおけるピークC及びピーク
Dは標準品のピークと照合することにより各々
3α,7α−ジヒドロキシ−12−ケト−5β−コラン
酸メチル及びコール酸メチルのものと一致した。
