Claims (1)
- 【特許請求の範囲】 1)真核微生物由来の対応するイントロン含有構造遺伝子から誘導したイントロ ン欠失構造遺伝子であって、前記イントロン欠失遺伝子が原核または真核微生物 内で産生物を発現できることを条件として、どちらの遺伝子も同一の産生物をコ ードしていることを特徴とする遺伝子。 2)前記遺伝子がフェニルアラニンアンモニアリアーゼ(PAL)またはPAL 活性を示すポリペプチドであることを特徴とする請求項1に記載の遺伝子。 3)前記遺伝子が真核微生物のPAL産生株のイントロン含有遺伝子であること を特徴とする請求項2に記載の遺伝子4)微生物がR.toruloidesで あることを特徴とする請求項3に記載の遺伝子。 5)前記遺伝子がPALまたはPAL活性を示すポリペプチドをコードする下記 のポリヌクレオチド配列:【配列があります】 と同一の構造、または該配列と関連する構造、または該配列から誘導した構造、 または該配列と相補的な構造を有することを特徴とする遺伝子。 6)前記遺伝子がPALまたはPAL活性を示すポリペプチドをコードすること を条件として、前記遺伝子がいくつかの塩基を欠くか、またはいくつかの塩基を 余分に含むか、または列挙したコドンのいくつかを他のコドンと置換することを 特徴とする請求項5に記載の遺伝子。 7)請求項1〜4の何れかに記載の遺伝子を含むことを特徴とする組換えDNA 分子。 8)請求項5または6に記載の遺伝子を含むことを特徴とする組換えDNA分子 。 9)前記組換えDNA分子がプラスミドであることを特徴とする請求項8に記載 の分子。 10)前記プラスミドがペクターであり、遺伝子に作働的に結合した発現制御配 列、および/またはE.coli k12.Bacillus subtili s、Saccharomyces cerevisae、Preudomona sPutida、Erwinia chrysanthemiまたは哺乳類の細 胞系由来のPALまたはPAL活性を示すポリペプチドに対する適当な転写/翻 訳信号を含むことを特徴とする請求項9に記載のプラスミド。 11)前記分子が遺伝子の翻訳開始点の上流側のリポソーム結合部位を含み、並 びにS.cerevisaeホスホグリセラートキナーゼ遺伝子および交配因子 遺伝子から誘導した転写信号をリポソーム結合部位の5′に配置することを特徴 とする請求項10に記載の分子。 12)請求項5に記載の遺伝子を、pUC9のIacZプロモータを有する相内 の遺伝子と共にプラスミドpUC9に挿入して成ることを特徴とする組換えDN A分子。 13)請求項8に記載の組換えDNA分子を用いて形質変換することを特徴とす る宿主微生物。 14)イントロンを欠失した遺伝子の製造方法であって、(I)R.torul oides株からPALのmRNAを単離する段階と、 (II)前記mRNAから、それぞれがPALまたはPAL活性を示すポリペプ チドをコードする遺伝子の一部を含む2つのイントロン欠失cDNA配列を合成 し、2つの遺伝子部分が遺伝子の3′末端と5′末端とを含むようにする段階と 、(III)2つの前記cDNA配列を結合して、PALまたはPAL活性を示 すポリペプチドをコードするイントロン欠失構造遺伝子を形成する段階とを含ん で成ることを特徴とする方法。 15)PALまたはPAL活性を示すポリペプチドをコードするイントロン欠失 遺伝子の一部であり、前記遺伝子の3′末端または5′末端を含むことを特徴と するポリヌクレオチド配列。 16)下記のポリヌクレオチド配列: 【配列があります】 と同じ、または該配列と関連する、または該配列から誘導したポリヌクレオチド 配列を含むことを特徴とする請求項15に記載の配列。 17)下記のポリヌクレオチド配列: 【配列があります】 と同じ、または該配列に関連する、又は該配列から誘導したポリヌクレオチド配 列を含むことを特徴とする請求項15にに記載の配列。 18)いくつかの塩基を欠くか、または他の塩基を含むかまたは列挙したコドン のいくつかを他のコドンと置換することを特徴とする請求項16又は17に記載 のポリヌクレオチド配列。 19)請求項15に記載のポリヌクレオチド配列を含むことを特徴とする組換え DNA分子。 20)請求項16または17に記載のポリヌクレオチド配列を含むことを特徴と する組換えDNA分子。 21)ポリヌクレオチド配列がpBR322またはpUC9と結合していること を特徴とする請求項20に記載の組換えDNA分子。
