【発明の詳細な説明】
本発明は、新規抗生物質サツカロシン
(Saccharocin)およびその製造法に関する。
本発明の新規アミノ糖抗生物質サツカロシン
(以下サツカロシン)の物理化学的性質は次のと
おりである。

1340、1140、1090、990cm-1付近の各波長に吸収
帯を有する。
核磁気共鳴スペクトル(水素核):第2図に示
す通り(D2O中、100MHz、内部基準Dss)

上記の物理化学的性状より、サツカロシンは新
規なアミノ糖化合物と認められ、またその平面構
造としては下記の構造が推定される新規化合物で
ある
また、サツカロシンの寒天希釈法による抗菌ス
ペクトル(最小発育阻止濃度を示す)は次表のと
うりである。

さらに本発明の抗菌性、特にグラム陰性菌に対
する優れた抗菌性を有するサツカロシンは、通常
医薬的に許容される鉱酸や、有機酸などの無毒性
酸付加塩の形で使用でき、例えば、塩酸、ヨウ化
水素酸、硫酸、リン酸、炭酸、酢酸、フマル酸、
リンゴ酸、クエン酸、マンデル酸、コハク酸、ア
スコルビン酸、アスパラギン酸、グルタミン酸な
どの酸類との塩の形で使用できる。また本発明の
サツカロシンまたはその無毒性酸付加塩は、例え
ば20〜100mg用バイアルまたは20〜100mg含有アン
プルとして注射用製剤として適用され、さらに1
g当り20〜100mg含有の坐剤用製剤として製剤化
してもよい。さらに本発明のサツカロシンまたは
その無毒性酸付加塩は、家蓄、家禽または魚貝類
等動物用の抗菌性治療用製剤もしくは予防用製剤
や飼料用組成物としても使用できる。これらの用
途において、サツカロシンは水によく溶解する
が、好ましくは酸付加塩の形で動物の飲料水に加
えて治療または予防、生育促進のための有効な濃
度の溶液として用いるかまたは、飼料中に添加し
てもよい。さらに注射用製剤となして、水、生理
食塩水などの注射用希釈剤とともに用いて投与し
ても有効である。
さらにこのサツカロシンは、その製剤設計上、
生体内にてサツカロシンとして効力を奏するため
のプロドラツグとなして用いてもよいものであ
る。
本発明のサツカロシン生産菌は、山口県武部郡
阿東町の畑土壌より分離した放線菌Ac3440株で
あつて、サツカロポリスポラ
(Saccharopolyspora)属に属するものと同定し、
サツカロポリスポラ・エスピーAc3440
(Saccharopolyspora sp.Ac3440)と称すること
とした〔工業技術院微生物工業技術研究所受託番
号:微工研菌寄第6238号(FERMP−6238)〕。
以下に本菌の菌学的諸性状について述べる。
形態的性状
スターチ・無機塩寒天培地(IPS培地4)
〔Inter.J.System.Bacteriol.16:313−340
(1966)〕上、37℃、10〜14日間培養し、観察し
た所見は次の通りである。
基生菌糸は曲線状または直線状で、分枝を伴
つて伸長し、菌糸の部分により、あるいは培養
後期に分断を生じ、直径0.4〜0.6μであり、飽
子は着生しない。
基生菌糸より生じた気菌糸は曲線状または直
線状で単純分岐をなし、直径0.5〜0.7μであり、
その先端はループ状またはゆるく2〜3回巻い
た螺旋を呈するものと、曲線状または直線状を
呈するものがある。気菌糸は分節してビーズ様
に多数連鎖(通常10個以上)した飽子を形成
し、しばしば胞子と胞子の間は空の菌糸部分に
より仕切られている。
胞子は卵形または短円筒形で、大きさは0.5
〜0.7×0.7〜1.3μであり、その表面は直線状ま
たは曲線状の長い毛様物質が房状に多数生えた
殻で覆われている。
基生菌糸や気菌糸に胞子のう、菌核または鞭
毛胞子を形成しない。
染色性
グラム染色は陽性で、抗酸性染色は陰性であ
る。
菌体組成
(1) B.Becker等の方法〔Appl.Microbiol12:
421−423(1964)〕により分析したジアミノピ
メリン酸(DAP)はメゾ−型(meso−
type)が検出され、Lechevalierの方法〔J.
Lab.Clin.Med.71:934−944(1968)〕で分析
した糖は、アラビノースとガラクトースが検
出された。
(2) H.Mordarska等の方法〔J.Gen.Microbiol.
71:77−86(1972)〕による脂質の分析におい
て、脂質LCN−Aは検出されず、また、D.
E.Minnikin等〔J.Gen.Microbiol.88:200−
204(1975)〕による分析において、ノカルド
ミコール酸(nocardomycolic acid)または
ミコール酸(mycolic acid)は検出されな
かつた。
培養的性状
各種培地上で、37℃、14日間培養し観察した
所見を第1表に示す。色の表示はColor
harmony manual第4版1958年(Container
Corporation of America)による色の分類に
従つた。

で生育、11%以上では生育しない。〔基礎培

以上のことにより、本菌Ac3440の特徴的性状
としては(1)形態において、分断性のある基生菌糸
より生じた気菌糸に、ゆるく巻いた螺旋状及び直
線状あるいは曲線状の胞子連鎖を形成し、胞子は
長い毛様物質の生えた殻に覆われており、(2)菌体
分析において、メゾ−ジアミノピメリン酸、アラ
ビノース及びガラクトースが検出され、脂質
LCN−A、ノカルドミコール酸及びミコール酸
は検出されず、(3)染色性において、グラム染色は
陽性、抗酸性染色は蔭性であり(4)好気性であると
の点が挙られる。
これらの特徴的性状をもとに、種々の文献より
検索したところ、サツカロポリスポラ属
(Saccharopolyspora Lacey & Goodfellow)
〔J.Gen.Microbiol.88:75−85(1975)〕に極めて
良く一致したことから、本菌Ac3440はサツカロ
ポリスポラ属に属するものと同定し、サツカロポ
リスポラ・エスピー・Ac3440
(Saccharopolyspora sp.Ac3440)と命名したも
のである。
次に本発明を実施し、サツカロシンを得るに当
つては、上記のサツカロポリスポラ・エスピー・
Ac3440を用いてなる培養物からの採取が簡便か
つ良好である。またその使用菌としては、上記の
菌はその一例であつて、本菌だけでなくサツカロ
シンを生産するサツカロポリスポラ属に属する菌
はすべて本発明において使用でき、例えばサツカ
ロポリスポラ属に属するサツカロシン生産菌また
はその変異株が挙られる。
次いで、本発明の新規抗生物質たるサツカロシ
ンを製造するに当つて例示すれば、上記サツカロ
ポリスポラ属に属するサツカロシン生産菌を通常
の微生物の培養に使用する培地成分を含む培地に
て好気的に培養することによつて得られる。培地
としては、固型培地または液体培地が用いられる
が、特に大量生産のためには液体培地、特に水性
培地が適当である。
培地の栄養源としては、微生物の培養に通常用
いられるものが広く使用されうる。炭素源として
は同化可能な炭素化合物であればよく、例えば、
グルコース、シユクロース、デキストリン、スタ
ーチ、モラツセなどが使用される。窒素源として
は利用可能な窒素化合物であればよく、例えば、
コーンスチープリカー、大豆粉、綿実粉、ペプト
ン、肉エキス、酵母エキス、アンモニウム塩、硝
酸塩などが使用される。さらに必要に応じて、リ
ン酸塩、炭酸塩、硫酸塩、塩酸塩、カルシウム、
カリウム、ナトリウム、銅、鉄、亜鉛、マンガ
ン、コバルトなどの塩類が使用される。
培養温度は菌が発育し、サツカロシンを生産す
る範囲内で適宜変更しうるが、特に好ましくは、
25〜35℃である。培養時間は条件によつて多少異
なるが、通常、72〜140時間程度であつて、サツ
カロシンが最高力価に達する時期を見計つて適当
な時期に培養を終了すればよい。
このようにして得られた、サツカロシン生産菌
の液体培養物においてサツカロシンは大部分が、
液体部分に生産されている。
次いでこの培養物からサツカロシンを採取する
のであるが、サツカロシンは水溶性の塩基性アミ
ノ糖化合物であることを利用して分離精製を行う
ことが簡便である。また生産されたサツカロシン
はバチルス、ズブチリスPCI219を被検菌として、
通常の寒天平板法により、活性区分の確認、およ
び定量を行なえばよい。
サツカロシンの分離精製手段の一例を示すと次
の通りである。すなわち、サツカロシン生産菌を
前述の如くして培養し、得られた培養物から固形
分を除去して培養液を得るのであるが、サツカ
ロシンがアミノ糖化合物であるためにその培養物
のPHを一旦酸性に調製し、これを中和して過
し、その培養液を得ることが好ましく、次い
で、この液を陽イオン交換樹脂例えばアンバー
ライトIRC−50(NH+ 4型)のカラムに吸着せし
め、これより活性物質を1Nアンモニア水にて溶
出せしめ、さらにその溶出液を濃縮した後、その
PHを、調製し陽イオン交換樹脂、例えばCMセフ
アデツクスC−25(NH+ 4型)のカラムに吸着さ
せて、0.03Nのアンモニウム水にて溶出し、溶出
した活性画分を減圧濃縮し、凍結乾燥することに
よつてサツカロシンの精製白色粉末を遊離塩基の
型にて得られる。またこのようにして得られるサ
ツカロシンは薄層クロマトグラフイーにて単一の
スポツトを示すものであることが簡単になしう
る。
次に本発明の実施例を挙げて具体的に説明する
が本発明はこれによりなんら限定されるものでは
ない。
実施例 1
デキストリン1%、グルコース1%、カゼイン
水解物0.5%、酵母エキス0.5%、炭酸カルシユー
ム0.1%を含有する培地(PH7.0)100mlを500ml容
三角フラスコに分取し、120℃、20分間加熱殺菌
した。この培地にサツカロポリスポラ・エスピ
ー・Ac3440株の斜面培養より一白金耳を接種し、
30℃、72時間振とう培養し、これを種培養物とし
た。
別に、グルコース0.2%、グリセリン4%、ペ
プトン5%、スターチ0.2%、脱脂大豆粉0.5%、
乾燥酵母0.5%、食塩0.5%、炭酸カルシユーム0.2
%(PH7.0)の組成からなる培地100mlを500ml容
フラスコに分注し加熱滅菌後、この培地100本に
前記種培培養物を3%づつ移植した。培養は30℃
で96時間振とう培養を行つた後、これらを併合し
て約10の培養物を得た。
実施例 2
実施例1で得られた培養物を、12N硫酸水溶液
にてPHを2.0に調製し、10分間撹拌した後、濃ア
ンモニア水にてPH7.0に調製し、遠心分離によつ
て上清液9を得た。得られた上清液を、アンバ
ーライトIRC−50(NH+ 4型)(ローム・アンド・
ハース社製)1を充填したカラムにチヤージ
し、水洗した後、1Nアンモニア水2にて溶出
せしめ、その全溶出液を得て、これを30mlまで減
圧濃縮した。
次いでこの濃縮液を6N硫酸によつてPH7.0に調
製し、これを、CMセフアデツクスC−25(フア
ルマシア・フアインケミカル社製)(NH+ 4型)
300mlを充填したカラム(径3cm)にチヤージし
て吸着せしめた。その後、0.03Nのアンモニア水
にて溶出し、溶出液を20mlづつ分画した。各分画
について、クロロホルム−メタノール−濃アンモ
ニア水(2:3:2)を展開溶媒とした薄層クロ
マトグラフイーを行ない、ニンヒドリン発色によ
り目的物を確認した。その結果、第124画分より
第143画分がサツカロシンのみを含有したもので
あつた。次いでこの画分を回収、合せて減圧濃縮
し、次いで凍結乾燥し、白色粉末を得、さらにこ
れを五酸化リンの存在下で40℃、48時間減圧乾燥
して、サツカロシンの精製白色粉末(遊離塩基)
